Abstract

This study aims to determine the profile of the ability of senior high school biology teachers in designing and testing local material-based bacteria and fungi cultivating practices during the Covid-19 pandemic. The method used in this study is a descriptive method with a qualitative approach. The participants in this study were 33 senior high school biology teachers at West Java province, Bandung. The data collection technique used the lesson plan assessment rubric, student worksheet, and practicum trial reports, besides that used both teacher profile and activities and obstacles questionnaires during the training. The research data analysis technique used data triangulation, data presentation, and conclusions. The data validity test includes data testing, data reliability testing, external validity/generalization. The results show that the profile of Senior High School biology teachers regarding teaching experience, as many as 63% of teachers had more than 10 years of teaching experience. The achievement of eligibility obtained a score of 85% (good category). These criteria include content, presentation, language, graphics, ease of usage, suitability of appearance, and usability of student worksheet features. The results of the questionnaire show that 91% of participants thought that the design and practical trial activities are very useful, 83% of participants thought that the material that had been delivered could be applied in schools. So that biology teachers can innovate further by using local materials to carry out practicum activities.

Abstract

The purpose of this research was to identify the effect of a virtual laboratory application on the student results in the environmental changes topic at SMAN 16 Bandung. A virtual laboratory is an activity of learning with the use of software as a simulator containing all necessary equipment in preparation, tools, working step demonstration, and practicum observation. During the teaching and learning process, the teacher guided the students to understand the concept of learning, especially in the concept of environmental changes topic (pollution and climate changes). The students were directed to analyze and identify the problem of the topic itself, so they would develop the ability to think critically and solve the problems. The method used in this research was pre-experimental with a one-group pretest-posttest design. The sample in the research consisted of 34 students in one class experiment. Instruments used in this research consisted of 30 multiple choices questions with six options and a questionnaire about the attitude cases to the learning process. Supported research results showed that there was an effective effect in the application of virtual laboratory, with an N-gain value of 0.72 with the category of high effect. The average learning mastery of students after implementing a virtual laboratory was 88%. Learning outcomes about students' attitudes when learning using virtual laboratory showed a very good category with an average of 81%, with the highest attitude on indicators of collaboration and critical thinking.

Abstract

Donuts are processed flour products made from a combination of several ingredients such as wheat flour, sugar, salt, butter, eggs, water, and yeast as a fermentation agent, these ingredients are mixed with potatoes as an additional raw material. The yeast will grow by converting the sugar into carbon dioxide gas and this gaseous-flavored compound is what makes the donuts rise. The method used in this study was an experimental laboratory method to test the effect of different yeast concentrations on the organoleptic characteristics of potato donuts. The three concentrations of yeast used were 1.4% (w/w), 1.8% (w/w), and 2.2% (w/w). Yeast affected all parameters, taste, color, aroma, and texture of the best donuts in the first treatment with a yeast concentration of 1.4% and the highest organoleptic value.

Abstract

Post-pandemic policies encourage teachers and students to carry out learning in offline, where they used to go online before. One learning strategy that can be applied in this condition is blended learning, which combines the advantages of offline and online learning. In improving the quality of learning outcomes and developing students' 21st-century skills, especially argumentation skills, blended learning strategies can be combined with inquiry-based learning models, one of which is the revised Argument-Driven Inquiry (rADI). This study aims to obtain an analysis of data related to the application of blended learning with rADI model in improving students' argumentation skills and concept mastery in Monera materials, especially regarding Bacteria. The research method used in this study was pre-experimental with a one-group pretest-posttest design. This research was conducted on students of class X MIPA at a high school in Bandung which consisted of 26 students. The data were obtained from the pre-test and post-test results of argumentation skills and concept mastery, and questionnaires on students' responses to learning. The results showed an increase in students’ argumentation skills with an N-Gain score of 0.81 and students’ concept mastery with an N-Gain score of 0.78 after applying blended learning with rADI model. The increase in argumentation skills and concept mastery was significant with N-Gain scores included in the high category. In addition, students' response data showed a good response to the application of blended learning with rADI model.

Abstract

Enzim selulase merupakan enzim komersial yang berperan penting sebagai biokatalis dalam berbagai proses industri, seperti industri tekstil, kertas, makanan, medis, detergen, dan lainnya. Produksi enzim selulase dapat dilakukan dengan pemanfaatan limbah selulosa, salah satunya jerami padi. Di Indonesia, jerami padi merupakan limbah pertanian terbesar namun pemanfaatannya belum optimal, biasanya digunakan untuk makanan ternak, media tumbuh jamur atau dibakar di lahan pertanian. Substrat selulosa dapat dihidrolisis oleh mikroorganisme selulolitik, salah satunya bakteri yang berasal dari saluran pencernaan rayap. Penelitian ini dilakukan untuk mengetahui pH dan suhu yang optimal untuk produksi selulase bakteri yang diisolasi dari saluran pencernaan rayap Cryptotermes sp. Seleksi bakteri selulolitik dilakukan pada media Carboxymethylcellulose (CMC) dengan indikator adanya zona bening yang dihasilkan di sekitar koloni bakteri selulolitik. Pre-treatment secara mekanik dan kimiawi dilakukan terlebih dahulu pada substrat jerami padi. Media SmF digunakan sebagai media produksi enzim selulase. Penentuan suhu dan pH optimal yaitu dengan variasi suhu 36,5 oC dan 37,5oC, sedangkan variasi pH medium 7 dan 8. Aktivitas enzim selulase diukur dengan penghitungan gula pereduksi menggunakan reagen 3,5-dinitrosalicylic acid. Dari hasil penelitian diketahui bahwa konsorsium bakteri selulolitik R3-1, R4-3, dan R7-3 berturut-turut merupakan genus bakteri Achromobacter sp., dan Pseudomonas sp. Konsorsium bakteri ini dapat menghasilkan enzim selulase tertinggi sebesar 8,93 U/mL pada waktu inkubasi 24 jam dengan pH medium 7 dan suhu inkubasi 36,5oC.

Abstract

Corncob is an agricultural waste rich in cellulose, making it a promising alternative substrate for cellulase enzyme production, which is widely used in various industrial applications such as bioenergy, food, and textile industries. Aspergillus niger is a well-known cellulolytic fungus capable of efficiently producing cellulase enzymes. This study aimed to optimize cellulase production by A. niger using corncob powder (Zea mays) as a substrate through submerged fermentation (SmF), with variations in temperature (29.5°C and 30.5°C) and pH (4.5 and 5.5). The cellulolytic activity of A. niger was tested on CMC agar medium. Cellulase activity was analyzed based on the concentration of reducing sugars using the DNS method. The highest enzymatic activity was recorded at 1.125 IU/mL with a biomass yield of 0.754 mg/mL at 30.5°C and pH 4.5. These findings suggest that optimizing environmental conditions and utilizing corncob waste as a carbon source can enhance cellulase production by A. niger, offering a sustainable and cost-effective approach for enzyme-based industries.
