Abstract

ABSTRACTBackground of Study: The widespread use of non-steroidal anti-inflammatory drugs (NSAIDs) as analgesic, antipyretic and anti-inflammatory agents still causes some worries in the polulation related to side effects, especially gastric ulcers. Suji leaf (Dracaena angustifolia) is a plant that well known as a food coloring also known contain flavonoids, saponins and phenolics that have the potential as antioxidants and anti-inflammatory. The purpose of this study was to determine the effect of suji leaf extract on gastric pH of male Rattus norvegicus rats induced by NSAIDs.Methods : This study was a laboartory experimental research using Randomized Controlled Trial with Post-test Only Control Group Design. Thirty male rats (Rattus norvegicus) divided into 6 groups, i.e normal control group (KN), negative control (K-), positive control (K+), and three treatment groups with Suji leaf extract dose 50mg/kgbw (KU1), 100mg/kgBW (KU2) and  group with Suji leaf extract dose 200mg/kgbw (KU3) for 14 days once daily after induction of NSAID (aspirin dose 200 mg/kgbw for 5 days) at the Animal Laboratory Faculty of Medicine, Universitas Surabaya. Gastric pH was measured after the rats fasted and sacrificed. Data were analyzed statistically by using One Way ANOVA test and LSD (Least Significant Difference).Results : The results showed that there was no significant difference between each group KU1, KU2 and KU3 on gastric pH of rats induced by NSAIDs (p 0.05). The lowest mean gastric pH level was in the K(+) group.Conclusion : The administration of suji leaf extract did not show a significant difference to the gastric pH of male Rattus noervegicus rats induced by NSAIDs.Keywords : Suji leaf; gastric ulcer; NSAID; gastric pH

Abstract

Background. Free radicals in cigarette smoke will hurt health when they enter to the respiratory tract. An excessive increase of free radicals in the body will cause oxidative stress. Free radicals are generated physiologically by the body's metabolism and can neutralize antioxidants in the body. An imbalance number of free radicals will result in cell damage and death. It has characterized by an increase in malondialdehyde levels in the blood and alveolar macrophages in the lung tissue. Giving red mulberry (Morus rubra) as an intake of antioxidants from outside the body can prevent adverse effects of cigarette smoke. Objective. This study analyses flavonoids' impact on red mulberry in reducing free radicals due to exposure to cigarette smoke by lowering levels of malondialdehyde and alveolar macrophages. Method. This research is experimental with a post-test control group design using male Wistar rats (Rattus novergicus) as experimental animals. Treatment of experimental animals through red mulberry per oral and exposure to cigarette smoke had conducted for 30 days. The parameters used were levels of malondialdehyde and alveolar macrophages in the lung tissue. Results. The research showed an increase in free radicals in the group exposed to cigarette smoke. Increasing intake of red mulberry can further reduce malondialdehyde levels and the number of alveolar macrophages (p<0.05). Conclusions. The antioxidants in red mulberry can reduce malondialdehyde levels in the blood and the number of alveolar macrophages in lung tissue due to exposure to cigarette smoke.

Abstract

Abstract — Diabetes mellitus is widely known as one of the non-transmittable diseases which is a global problem because its incidence is increasing every year throughout the world. The condition of hyperglycemia in diabetes mellitus has a very influential effect on endothelial blood vessels due to the process of glucose auto-oxidation in forming free radicals which in turn will produce macro and microvascular dysfunction. Next, this condition will further increase morbidity and mortality rates in people with diabetes mellitus. The using of antioxidants in patients with diabetes mellitus was found to be effective in reducing the emergence of arising complications. This is supported by various the benefits of antioxidants related to pathological process of diabetes mellitus due to conditions of oxidative stress. In the future, the safety and effectiveness of supplements or food contain antioxidants in an effort to overcome the condition of diabetes must still be proven further. Abstrak— Diabetes melitus banyak dikenal sebagai salah satu penyakit tidak menular yang menjadi masalah global karena insidensinya setiap tahun yang terus meningkat di seluruh dunia. Kondisi hiperglikemia pada diabetes melitus mempunyai efek yang sangat berpengaruh pada endotel pembuluh darah akibat adanya proses auto-oksidasi glukosa dalam membentuk radikal bebas yang pada akhirnya akan menghasilkan disfungsi makro dan mikrovaskular. Kondisi inilah yang selanjutnya akan menimbulkan komplikasi yang selanjutnya akan meningkatkan angka morbiditas maupun mortalitas pada penderita diabetes melitus. Penggunaan antioksidan pada penderita diabetes melitus ternyata diketahui efektif dalam mengurangi munculnya komplikasi yang timbul. Hal ini didukung dengan berbagai penelitian yang membuktikan manfaat antioksidan terkait proses patologi dari diabetes mellitus akibat kondisi stres oksidatif. Pada masa yang akan datang, keamanan dan keefektifan suplemen atau makanan yang mengandung antioksidan dalam upaya mengatasi kondisi diabetes tetap harus dibuktikan lebih lanjut.

Abstract

Diabetes mellitus (DM) is one of disease that its incidence increases every year worldwide. The condition of DM can cause various complications caused by oxidative stress. Stichopus hermanii (SH) or golden sea cucumber extract contains antioxidant compounds that have been proven to reduce oxidative stress conditions. The purpose of this study was to investigate the effect of Stichopus hermanii extract on condition of diabetes mellitus by looking at changes in fasting blood glucose, plasma insulin, and malondialdehyde levels in animal models of Wistar rats. This study was a laboratory experimental study using Randomized Control Trial Design with Post-test only control group design. Thirty-five male Wistar rats divided into five groups, i.e normal control group, positive control, negative control and two treatment groups with SH extract dose 8.5 and 17 mg / kgBW for 14 days once daily after induction of Streptozotocin at the Biochemistry Laboratory of the Faculty of Medicine, Airlangga University. Fasting blood glucose level was measured by a glucometer, plasma insulin measured by ELISA and MDA level was measured by a spectrophotometer. Data were analyzed statistically by using One Way ANOVA test and Kruskal Wallis. There were significant results of SH extract can reduce fasting blood glucose (Kruskal Wallis, p=0.030) and MDA (Kruskal Wallis, p=0.042) but not in plasma insulin (ANOVA, p=0.130). The lowest MDA level occurs in the K4 group that given SH extract dose 17 mg/kg BW than another experimental group. As the conclusion, this study showed SH extract can decrease fasting blood glucose and oxidative stress in diabetic-induced rats.
