Abstract

Cincau Hijau leaves (Cyclea barbata L.Miers ) has been used empirically as a medicinal plant because it contains many potential compounds such as flavonoids. Cincau Hijau leaves has been known as antioxidant activity. Determination of pharmacognosy parameters and phytochemical screening of dry leaves and extracts have been carried out as well as determination of total fl avonoids content. Fresh leaves were extracts with water and Dry leaves were macerated with 96% ethanol and were then concentrated by rotavapor to obtain viscous extracts. Free radical scavenging activity of the extracts was evaluated using DPPH method. Afterwards, determination of specifi c and non-specifi c parameters were performed. Results of phytochemical screening of powder and 50% and 96% ethanol extract showed that tall the tested samples contained alkaloid, fl avonoids, saponins, tannins, steroids/ triterpenoids, coumarin. The examination of specifi c parameter showed that the extract has a thick consistency, tawny color, bitter taste, characteristic odor. In addition, water-soluble compound and 96% ethanol extract are 46.64 and 62.13% respectively whereas ethanol-soluble compounds are 39.22 and 74.72%, respectively. While the results of nonspecifi c parameters of 50% and 96 % ethanol extract displayed total ash content of 9.69 and 9.49%, respectively, acid insoluble ash content of 0.30 and 0.16%, respectively, content of water soluble ash of 9.17 and 4.30%, respectively, loss on drying of 9.35 and 8.9%, respectively, water content of 8.45 and 7.25%, respectively. Based on heavy metal contamination, Pb concentration in 50 and 96% ethanol extract are 0.0227 and 0.0333 mg/kg, respectively whereas Cd concentration are 0.1206 and 0.0022 mg/kg, respectively and total number of CFU of 4,22 x 103 and 2,30 x 103 colonies/g, respectively while molds and yeasts number of colony of 0,48 x 102 and 8,88 x 102 colonies/g, respectively. Moreover, the total flavonoid was 0,19 %. Result of DPPH inhibition test showed that IC50 96 % ethanol extract are 83,280 ppm and water extracts are 102,01 ppm

Abstract

Mulberry Leaf (Morus alba L.) contains oxyresveratrol has the potential as a skin lightener. The activity of mulberry leaf callus from tissue culture is unknown as an inhibitor of the tyrosinase enzyme. The purpose of this study was to determine the activity callus extracts of mulberry leaf which obtained from tissue culture with the addition of growth regulator NAA 1.0 ppm and BAP 2.5 ppm and to formulate callus extract in Nanostructured Lipid Carrier and its characteristics. Callus was extracted by maceration-sonication. NLC is made by solvent evaporation method, NLC characterization including particle size, polydispersity index, zeta potential and particle morphology. The best NLC was made into a gel and evaluated organoleptic, viscosity, flow properties, pH, and tyrosinase enzyme inhibitory on extracts and NLC gels. The characterization of NLC includes particle size of 189.8 nm-632.8 nm; polydispersity index, 0.387-0.582. The morphology of NLC were spherical and zeta potential of -7.37 mV. NLC gel was semi-solid, greenish yellow, homogeneous, viscosity 530000 cPs, plastic thixotropic flow properties, pH 5.26, and inhibitory activity of tyrosinase enzymes (IC50) of callus extract and NLC gel has 72.51 µg/ml and 79.69 µg/ml, respectively. It can be concluded that callus extract of Mulberry leaf can be prepared into Nanostructured Lipid Carriers, NLC gels are physically and chemically stable and have the potential of lightening activity.

Abstract

Callus is a cell mass which is formed by cell tissues and formed in explants surface or explants slice which divide them continuously in a controlled condition. With tissue culture method, the aim in using callus is for reproduce new plants to get the secondary metabolite which match with the secondary metabolite from the origin plants. Mahkota dewa (Phaleria macrocarpa [Scheff.] Boerl.), from Thymelaceae family is a very useful plant which can be used for medicine. Its leaves and seed's skin are the most important parts that usually used. In this experiment, Mahkota’s seed is planted in germination media to get a sterile plant, and then from the plant, we take the leaves for callus initiation. We do a phytochemical filtering, TLC, and HPLC tests with this callus that we produced then we compared the result with Mahkota Dewa's leaves. From the phytochemical filtering result we found that both of them have alkaloid secondary metabolite, flavonoid saponin, tannin, steroid/triterpenoid. We also found some chromatograms which are similar in TLC and HPLC.
