Abstract

Abstract

Radioiodine has been known as an adjuvant therapy for thyroid cancer. Beside for thyroid cell, radioiodine is reported to be taken up by breast cell. It may be proposed as an alternative for breast cancer therapy. Objective: The study aim was to analyze an uptake of radioiodine and cell proliferation rate of breast cancer cell after the exposure of radioiodine. Methods: This study used SKBR-3 cell line as representative of Human Epidermal Growth Factor Receptor (HER2+) subtype. The cells were planted in DMEM medium added 10% fetal bovine serum (FBS), panstrep 1% and amphotericin B. The cells were grown until 80% confluent and then stripped. Subsequently the cells were sub-cultured on plates 12 and 6 wells respectively for measured uptake of radioiodine and cell proliferation rate after the exposure. Radioiodine uptake was calculated by sum efflux of 125 Iodine (125I) and the amount of uptake after the addition of 95% ethanol. Cell proliferation rate was measured by calculated by using clonogenic assay after iodine-131 (131I) exposure. Results: The average of iodine uptake was 194±50 cpm/106 cell. The average cell proliferation after radioiodine exposure was 54%±5%; Conclusions: Radioiodine is taken by SKBR-3 cells and it reduced cell proliferation rate. This finding shows an opportunity for radioiodine as an alternative therapy for breast cancer. Another studies are needed to understand of cell death the mechanism.

Abstract

Kata kunci: new normal, pengetahuan, sikap, tindakan, covid, murid SD ABSTRACT Padang enforces offline education by limiting the hours of education while still paying attention to health promotion. Students are asked to wear masks, keep their distance, and wash their hands before entering their respective classes. One of the efforts to improve health promotion is to make students well educated about COVID and its transmission. Community service activities were carried out at SDN 10 Lambung Bukit in the fostered village of Medical Faculty Universitas Andalas (FK Unand) in grades 3A, 4A, 5A SD as a pilot class for other classes. Activities are carried out continuously for 6 weeks. Activities in the form of education using videos and training on the formation of the Covid task force. Knowledge, attitudes, and actions are assessed before giving the material and reassessed at the end of week 4. At week 5 and 6 an assessment of knowledge, attitudes, and actions towards covid prevention is carried out. This activity was concluded to have formed a covid task force at SDN 10 Lambung Bukit so that it was hoped that a pattern of attitudes and actions that had become a culture in dealing with the new normal in this pandemic era was expected. Abstract

Thyroid carcinoma is the most common malignancy found in the endocrine glands and accounts for 1% of all malignancies. The average incidence of thyroid carcinoma cases according to the National Cancer Institute (NCI) from 2012-2017 was 14.3 per 100,000 total population per year, with an average mortality rate of 0.5 per 100,000 population per year. In Indonesia in 2000, thyroid carcinoma ranked ninth out of the ten most common carcinomas (4.3%) and covered 90% of all endocrine carcinomas. Based on medical record data at Dr. M. Djamil Padang General Hospital, the incidence of thyroid carcinoma in 2015 was 373 people and increased in 2016 to 850 people. Based on the problems described above, and there has not been much research on Thyroglobulin levels as a prognostic factor for recurrence of thyroid carcinoma in Indonesia, especially Padang. Encourage the author to conduct research on differentiated thyroglobulin levels after total thyroidectomy at Dr. M. Djamil Padang General Hospital.

Abstract

Thyroid cancer poses a persistent clinical challenge, particularly when conventional therapies fall short. Growing evidence suggests that molecular iodine (I₂) may exert direct antitumor effects beyond its classical role in thyroid hormone synthesis. This study evaluated the antiproliferative and apoptosis-inducing effects of I₂ on primary cell cultures derived from differentiated thyroid cancer tissue and established dose- and time-response relationships. Primary cultures were exposed to I₂ at concentrations of 5, 10, 20, 40, 80, and 100 µM for 24, 48, and 72 hours. Cell viability was assessed using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, with IC₅₀ calculated accordingly. Apoptosis was characterized by acridine orange/propidium iodide (AO/PI) double-staining, analyzed by fluorescence microscopy and ImageJ software. I₂ demonstrated clear antiproliferative and apoptotic activity in a dose- and time-dependent manner. The strongest effect appeared at 100 µM after 72 hours, reducing viability to 1.39% (p < 0.001). At the IC₅₀ of 2.09 µM, iodine significantly suppressed proliferation and induced apoptosis, with an apoptosis rate of 81.77% ± 12.24% (p = 0.004). These findings confirm the meaningful antineoplastic potential of I₂ against differentiated thyroid cancer cells. This study supports SDG 3 (Good Health and Well-Being), contributing to efforts to develop more equitable, effective cancer treatment options and to reduce cancer-related morbidity globally.
