Abstract

This study was experimental, conducted using a Completely Randomized Design (CRD) with 3 x 2 factorial design, and four replicates have been carried out to evaluate the protease and amylase activities of Anguilla bicolor McClelland. A total of 71 individuals divided into three weight groups were used in this study.  The first group with an average weight of 41.25 ± 0.898 g consisted of 51 eels,  the second with an average weight of 319.8 ± 4.666 g composed of 14 eels, and the third with a mean weight of 569.5 ± 9.150 g consisted of 6 eels. The results showed the protease activity differed significantly based on eel size and intestinal segment (P ˂ 0.05). This research recorded the highest protease activity was in eels within the smallest weight group (41.25 ± 0.898 g). This study also revealed the protease activity in the anterior intestine was higher than the posterior in all size of eels. The amylase activity did not differ significantly (P>0.05) by eel size and intestinal segment. This study concluded the protein digestion capacity of smaller eels was higher than larger eels, and the protein digestion capacity was greater in the anterior intestine than the posterior intestine. The carbohydrate absorption capacity in eel was not affected by the variety of fish size which indicates no change in the feed category.

Abstract

The availability of fish seed is very important in fish propagation. Good quality of fish seeds were produced by a good brooder which characterized a large number of egg production at spawning. The aims of this research were to evaluate the oocytes development in the hard-lipped Barb (Osteochilus hasselti C.V.) given 17β-estradiol, different percentage of protein in the diet, and their combination. This research was conducted experimentally applying Factorial Completely Randomized Design. The first factor was percentage of protein in the diet consisted of 4 levels namely 25% (P1), 30% (P2), 35% (P3), and 40% (P4), the second factor was dose of 17β-estradiol benzoate (EB) consisted of 3 levels namely 0 µg/kg of body weight (D0), 126 µg/kg of body weight (D1), and 210 µg/kg of body weight (D2), thus there were 12 combinations of treatment with 3 replicates. The results showed that neither of protein proportion or 17β-estradiol affected the proportion of oocytes of any developmental stage (p>0,05) within the first two weeks post spawning. The treatments, however, significantly increased the proportion of oocytes at V3-V5, V6-V7, and post-vitellogenic stages (p<0,05) started at week 4th post spawning. The GSI increased in correlation to the proportions of oocytes at stage ≥ V6 (r=0,701; p<0,01) and the dose of 17β-estradiol (r = 0.357, p <0.05). There was no significant different on GSI amongst the experimental groups. The HSI tend to decrease as the dose of 17β-estradiol increased (r = -0.210, p> 0.05). In conclusion, the percentage of protein in the diet and different doses of 17β-estradiol improved oocyte development of Hard-Lipped Barb (Osteochilus hasselti C.V.).

Abstract

Ikan Nilem (Osteochilus hasselti C.V) merupakan ikan omnivora dari familia Cyprinidae. Jenis pakan yang dikonsumsi berkaitan dengan alat pencernaan yang dimilikinya. Pencernaan pada ikan tersebut terjadi di dalam usus halus dan berlangsung secara enzimatis. Enzim-enzim yang aktif pada saluran pencernaannya adalah protease dan amilase yang dihasilkan oleh pankreas dan hepatopankreas. Aktivitas enzim pencernaan diketahui dengan mengukur banyaknya mikromol maltosa dan tirosin yang dihasilkan per menit. Penelitian ini bertujuan untuk mengetahui perbedaan aktivitas protease dan amilase pada hepatopankreas dan intestine ikan Nilem (Osteochilus hasselti C.V.). Hasil penelitian ini diharapkan nantinya akan digunakan sebagai dasar formulasi pemberian pakan yang cocok untuk ikan omnivora salah satunya adalah ikan Nilem (Osteochilus hasselti C.V.). Materi yang digunakan adalah ikan Nilem berukuran 9-16 cm dengan bobot 5-50 gram. Metode penelitian yang dilakukan adalah metode survey dengan purposive random sampling dengan rancangan percobaan berupa Rancangan Acak Lengkap (RAL) dengan pola faktorial 3x4 dan empat kali pengulangan. Sampel dikelompokkan menjadi tiga ukuran yang berbeda dengan bobot 5-10 g dan panjang 9-10 cm, 11-25 g dan panjang 11-12 cm dan 35-50 g dengan panjang 13-16 cm. Metode analisis yang digunakan yaitu one way analisis of variance (ANOVA) dan dilanjutkan dengan uji beda nyata terkecil (BNT). Hasil uji BNT menunjukkan bahwa aktivitas protease dan amilase ikan Nilem tidak berbeda nyata (P>0,05) dengan ukuran tubuh ikan. Aktivitas protease dan amilase ikan Nilem berbeda nyata (P<0,05) dengan organ pencernaan. Dengan demikian dapat disimpulkan bahwa tidak ada hubungan antara aktivitas protease dan amilase dengan ukuran tubuh ikan Nilem; serta ada hubungan antara aktivitas protease dan amilase dengan organ pencernaan ikan Nilem.

Abstract

Eel (Anguilla bicolor) is a catradomous fish, in the growth phase of life in freshwater and upon reaching adult will migrate to ocean waters to reproduce. Eel during the migration will use energy from body reserves to osmoregulation. Increased salinity will affect the osmotic pressure of the body of the fish so that the fish do active transport which requires energy. Energy that used for osmoregulation approximately 20–50% of the total energy from carbohydrates, fats and proteins. Carbohydrates are the first source of energy that is used and then will be replaced by fat and protein as energy after carbohydrate depleted. Thus the energy utilization during osmoregulation will affect the changes in the composition of protein and fat content of the body. Fat content will affect the value of lipostatic fish is one of the growth parameters. The purpose of this study was to evaluate changes in the levels of protein and lipostatic eel (Anguilla bicolor) is maintained in a range of salinity. This research method is experimental research design with CRD, treatment in the form of maintenance medium salinity; 4 ppt (control), 15 ppt, and 30 ppt. Each treatment was repeated 5 times. Test fish used were obtained from the Village Pesanggrahan Cilacap with range size between an average weight of 395–920 g and an average length of 59–81 cm. The variables measured were the levels of protein and lipostatic eel. Protein content measurement was conducted using kjeldahl and the fat content by soxhlet. Research results showed that the eels were exposed to salinity 4 ppt, 15 ppt and 30 ppt no effect on protein content and body lipostatic eel. It can be concluded that the eel is able to adapt well in the range between 4 ppt salinity to 30 ppt.

Abstract

Research to determine the activity of amylase and protease digestion gourami on various temperature has been carried out experimentally using a completely randomized design (CRD) with 3 treatments and 4 replications. The treatments were included gourami nurturing on water temperature 24oC (P1), gourami nurturing on water temperature 28oC (P2), and gourami nurturing on water temperature 32oC (P3). The results the amylase activity is showed no significant difference between treatments (P>0,05) where as protease activity showed there are significant differences between treatments (P0≤0,05). In conclusion, the difference in protease activity of gourami occured in water temperature 24oC (P1) and 28oC (P2) and the protease activity was highest at 28oC water temperature, but the amylase activity did not difference.
