Abstract

Graptophyllum pictum is a well-known folk medicine in Indonesia. The red leaves are widely used for their anti-inflammatory properties to relieve hemorrhoids. However, little is known about to what extent these properties can affect platelet aggregation. This study was aimed to determine the anti-inflammation and anti-platelet aggregation activities of the ethanolic extract of Graptophyllum pictum (EEGP). Apart from the ear and hind-paw edema assay, anti-platelet aggregation and bleeding time assay were also performed on male Wistar rats. The test animals were grouped into six groups: three treatment groups termed EEGP100, EEGP600, and EEGP3000 (received 100, 600, and 3000 mg/kg BW of EEGP, respectively), one control-solvent group (1% CMC-Na), and two positive control groups (aspirin and Na-diclofenac). Based on the percentage of edema inhibition, absorbance change, and bleeding time, EEGP600 and EEGP3000 produced a comparable percentage of edema inhibition to that of Na-diclofenac, which was not found in EEGP100. Also, EEGP600 and EEGP3000, but not EEGP100, inhibited platelet aggregation as effective as aspirin (positive control). In conclusion, if administered at 600 and 3000 mg/kg BW, the ethanolic extract of Graptophyllum pictum can function as an anti-inflammatory agent that inhibits platelet aggregation.

Abstract

Hypercholesterolemia is a disease associated with high levels of cholesterol and LDL levels in the blood. Utilization of the commercial drugs can be given; however apart from the expensive price, adverse side effects might occur. It makes people choose alternative medication with herbal medicine through the use of natural materials. This study aimed to determine the effect of the combination of mangosteen peel extract-extract of roselle calyx and mangosteen peel extract-extract the ant nest plant as lowering cholesterol and triglyceride levels in male rats. The method used in this study was a laboratory experimental method using device posttest only control group design (simple experimental design). This study used 25 male rats of Wistar strain, divided into 5 groups; Group I: group without treatment, group II: control group solvent (NaCMC 1%), group III: positive control group (Simvastatin), Group IV: combination group mangosteen peel extract (200 mg / kg) - extract of roselle calyx (250 mg / kg), group V: group combination of mangosteen peel extract 200 mg / kg) - extract anthill (270 mg / kg). Induction of cholesterol in rats using quail egg yolk (10 ml / kg). The results showed that there was no significant difference in cholesterol and triglycerides between the combination of both extracts of mangosteen peel with a positive control (p<0,05).

Abstract

Telah dilakukan penelitian mengenai pengembangan metode KCKT dalam penetapan kadar tablet ibuprofen dalam plasma darah tikus jantan wistar secara in vitro. Metode  divalidasi berdasarkan parameter linearitas, akurasi, dan presisi. Kondisi KCKT yang digunakan adalah fase terbalik dengan kolom shim pack CLC ODS dan fase gerak asetonitril:bufer fosfat (35:65 v/v), laju alir 0,8 mL/min. Hasil uji linearitas menunjukan nilai r = 0,993 pada kisaran konsentrasi 0,5-5 μg/mL, presisi 0,93 (%RSD), dan perolehan kembali 92,9%. Abstract

Photosensitizer modifications are needed to improve the performance of Photodynamic Therapy (PDT) for the treatment of cancer. This work appraised the photophysical and photobiological characteristics of tetrapyrrolic macrocycles. Meso-tetraphenylporphyrin (m-TPP) and meso-tetraphenylchlorin (m-TPC) represent non-reduced and reduced pyrrole rings tetrapyrrolic macrocycles, respectively. The absorption spectra of m-TPP and m-TPC were performed. The relative quantum yield of singlet oxygen (ΦΔ) was measured at two different pH (6.3 and 7.4) relative to chlorine e6. Dark toxicity was monitored using the MTT assay on breast cancer T47D and NIH 3T3 cell lines. In addition, the stability of the compounds under indirect sunlight was also observed. The absorption maxima of the Q1 band of both compounds are at a longer wavelength. The Q1 peak of a reduced pyrrole ring is more intense than the non-reduced pyrrole ring with the lowest energy transition undergoing a redshift. The singlet oxygen generations are higher at pH 7.4 for both compounds with them-TPC showed a higher relative singlet oxygen quantum yield. In addition, the photosensitizers are stable under indirect sunlight radiation. m-TPC has higher cellular uptake than m-TPP in both cell lines. The dark toxicity of m-TPC is higher than m-TPP in T47D cells. However, the opposite occurred in observing dark toxicity in the NIH 3T3 cell line. Our study concludes that the cellular uptake and ΦΔ can be enhanced by reducing the pyrrole ring of the tetrapyrrole macrocycle. Therefore, m-TPC proved to be a better photosensitizer than m-TPP.
