Abstract

Cervical cancer is a type of cancer possessing the 3rd highest incidence among women worldwide. Todays therapies against cancer are still ineffective, thus people are seeking for alternative method. Erythrina fusca Lour. has been used traditionally as anticancer. This experiment is conducted to study the ability of chloroform extract of E.fusca Lour. leaves and its isolates on HeLa cervical cancer cells. The cytotoxicity assay and doubling time assay were done using direct counting method, while the DNA staining assay was done using acrydine orange. The chloroform extract and isolate #30 showed cytotoxic activity with IC50 of 16 and 5 μg/ml, respectively, while isolate #2 and isolate #8 didnt. The effluent #30 could not affect HeLa cells growth at the given concentration, but it might promote DNA fragmentation indicating apoptosis induction. The molecular mechanisme underlying these effects still need to be further explored.Keywords :  Erythrina fusca Lour., isolate and effluent of chloroform extract, antiproliferative, cervical cancer

Abstract

Buah  adas  mengandung  minyak  atsiri  antara  lain  anethole,  fenchone  dan  metil chavicol. Senyawa anethole dilaporkan memiliki aktivitas antibakteri  pada bakteri Gram  Positif  dan  Gram  Negatif.  Penelitian  ini  bertujuan  untuk  membuat  lotion minyak  atsiri  dari  buah  adas  dan  menentukan  aktivitas  antibakterinya  terhadap Staphylococcus  aureus  dan  Pseudomonas  aeruginosa.  Penelitian  ini  dilakukan untuk membuat lotion menggunakan minyak atsiri buah adas yang diperoleh dari destilasi  uap  air  dengan  konsentrasi  1,  5,   dan  10%  (Formula  I,  II  dan  III). Kemudian  lotion  tersebut  diuji  parameter  fisik  dan  ditentukan  aktivitas antibakterinya.  Hasil  penelitian  menunjukkan  bahwa  semakin  tinggi  konsentrasi minyak  atsiri,  sediaan  lotion  yang  dibuat  memiliki  daya  lekat  dan  viskositas semakin  rendah.  Variasi  konsentrasi  minyak  atsiri  tidak  memberikan  pengaruh pada  sifat organoleptis, pH dan  kestabilan lotion.  Diketahui pula bahwa semakin tinggi  konsentrasi  minyak  atsiri  pada  sediaan  lotion  maka  semakin  tinggi  daya sebar dan diameter zona hambatnya terhadap pertumbuhan Staphylococcus aureusdan  Pseudomonas  aeruginosa.Minyak  atsiri  buah  adas  memiliki  aktifitas antibakteri  setelah  dibuat  dalam  sediaan  lotion  meskipun  lemah.  Formula  yang menunjukkan  aktivitas  antibakteri  yang  paling  tinggi  adalah  formula  III  dengan konsentrasi 10% minyak atsiri. Abstract

Graptophyllum pictum is a well-known folk medicine in Indonesia. The red leaves are widely used for their anti-inflammatory properties to relieve hemorrhoids. However, little is known about to what extent these properties can affect platelet aggregation. This study was aimed to determine the anti-inflammation and anti-platelet aggregation activities of the ethanolic extract of Graptophyllum pictum (EEGP). Apart from the ear and hind-paw edema assay, anti-platelet aggregation and bleeding time assay were also performed on male Wistar rats. The test animals were grouped into six groups: three treatment groups termed EEGP100, EEGP600, and EEGP3000 (received 100, 600, and 3000 mg/kg BW of EEGP, respectively), one control-solvent group (1% CMC-Na), and two positive control groups (aspirin and Na-diclofenac). Based on the percentage of edema inhibition, absorbance change, and bleeding time, EEGP600 and EEGP3000 produced a comparable percentage of edema inhibition to that of Na-diclofenac, which was not found in EEGP100. Also, EEGP600 and EEGP3000, but not EEGP100, inhibited platelet aggregation as effective as aspirin (positive control). In conclusion, if administered at 600 and 3000 mg/kg BW, the ethanolic extract of Graptophyllum pictum can function as an anti-inflammatory agent that inhibits platelet aggregation.

Abstract

COVID-19 global pandemic increases the usage of vitamin supplement products. However, easy access to purchase and consume the product increases the risk for its misuse. Lack of information and or misinformation available for vitamin supplement product usage leads to its abuse. Teachers' and parents' role in deciding the choice and use of vitamin supplement products for the pupil and children is crucial. Thus, a program was held to provide information about the correct usage of vitamin supplement products for teachers and parents. The program was performed for the teachers and pupil parents of TK Aisyiyah Ledug via online. The attractive audio-visual program materials were delivered via animation video, artistic leaflet, and presentation. The teachers and parents responded positively toward the program. All attendants actively participated, and 100% of attendants agreed that the program was interesting and increased their knowledge on the appropriate usage of vitamin supplement products.

Abstract

ABSTRAK Rosela (Hibiscus sabdariffa L) merupakan tanaman yang serba guna. Sebagai obat tradisional, kelopak rosela berkhasiat sebagai antioksidan, antiseptik, diuretik, antikolesterol. Penelitian ini bertujuan untuk mengetahui konsentrasi povidon terhadap persyaratan mutu granul yang baik, tanggapan rasa dan deteksi kandungan flavonoid dalam granul instan ekstrak rosela. Kelopak segar rosela dihaluskan dengan bantuan berupa blender kemudian sari yang diperoleh diuapkan untuk menghilangkan kandungan air sampai terbentuk ekstrak kental. Granul instan jus rosela dibuat dengan metode granulasi basah dalam 3 formula yang berdasarkan konsentrasi povidon yang berbeda yaitu F I 1%, F II 3%, FIII 5%. Uji yang dilakukan terhadap granul instant ekstrak rosela adalah uji fisik granul dan uji tanggapan rasa. Hasil uji fisik granul instan menunjukaan bahwa formula I memenuhi persyaratan sifat fisik granul yang baik. Untuk uji waktu alir granul (8,29 0,38 detik), uji sudut diam (39,11 0,790), uji susut pengeringan %MC (2,99 0,046), uji distribusi ukuran partikel (692,1797 µm). Hasil uji tanggapan rasa menunjukan bahwa granul instan ekstrak rosela dapat diterima oleh responden. Kata kunci: kelopak bunga rosela, povidon, granul instan. ABSTRACT Roselle is a very useful plant. Widely used as traditional medicine, its calyx functions as antioxidant, antiseptic, diuretic, and anticholesterol. This research is aimed at knowing the concentrate of povidone toward the condition of a good quality of granule, taste reaction, and the detection of content of flavonoid in the instant granule of the roselle extract. The fresh calyx roselle was pounded with mixer and then the obtained essence was steamed to lose the water content, until thick extract was derived. The instant granule of rosella juice was formulated with wet granulation method in 3 formulas which is based on differrent concentrate of povidone, they are F I (1%), F II (3%), F III (5%). The test were done to the instant granule from the extract Hibiscus sabdariffa for its granule physical test and taste reaction test. The result of the test of physical instant granule showed that F I meet the requirement of good granule. For flow rate test (8,29 0,38 second), point of quit test (39,11 0,790), moisture content test with %MC (2,99 0,046), and for test distribution of the size of particle with microscopis method (692,1797 µm). The result of the test of taste reaction showed that the extract of instant granule can be accept by the respodent. Keyword ׃ fresh calyx roselle, Povidone, granule instant

Abstract

As the pandemic of the coronavirus disease 2019 (COVID-19) continues while there is no drug and vaccine available, every effort to discover one should be considered. This review aimed to discuss the potential use of jamu, the Indonesian traditional herbal medicine, to deal with COVID-19 by following those of more-established traditional Chinese medicine (TCM). The online literature search using the PubMed database, as well as the circulars from the Indonesian Ministry of Health, were carried out to collect data up to June 07, 2020. The use of TCM for the treatment and prevention of COVID-19 has been officiated in the Chinese National Clinical Guideline on COVID-19 Associated Pneumonia, with shen-fu-tang + su-he-xiang pill and xiang-sha-liu-junzi- tang as the most frequently suggested formulae to treat severe and recovery stages of COVID-19, respectively. In Indonesia, the highly promoted product for the prevention of this disease is likely the adaptation of a TCM formula, while the use of some other formulae, which mainly containing Zingiberaceae, is suggested by the Government for the said purpose. The rationalization of the use of medicinal plants and suggested polyherbal formulae, based on their available pharmacological activity and the bioactive compounds, is also discussed in this review. The use of jamu for COVID-19, however, needs to be carefully considered for the limited scientific data available to support it.

Abstract

Osteoporosis represents a significant global public health issue, particularly among the aging population. Its incidence reaches 18.3% of the total population, with the highest prevalence observed in elderly postmenopausal women. A key factor in osteoporosis is the decreased expression level of estrogen receptor alpha (ERα), attributed to its degradation by the ubiquitin ligase protein complex Cullin4B (CUL4B), DNA damage binding 1 (DDB1), and aryl hydrocarbon receptor (AhR), collectively known as CUL4BAhR. Acanthus ilicifolius L contains compounds exhibiting antiosteoporosis activity, primarily by inhibiting osteoclastogenesis via RANKL. However, no reports exist of antiosteoporosis agents that act by inhibiting ERα degradation via CUL4BAhR. This study employed an in silico approach to predict active compounds from A. ilicifolius that could inhibit ERα degradation via CUL4BAhR, potentially developing them into antiosteoporosis agents. We utilized the 3D structures of proteins CUL4B-DDB1 (PDB ID:4A0L), AhR (5NJ8), and ERα (1A52) in various molecular docking tools, including ClusPro2.0, PyRx0.8, PyMol, PLIP, and SWISS-MODEL for QMEAN and structure assessment analysis. The ligands tested were acancifoliuside, acanthaminoside, acteoside, isoacteoside, (-)-lyoniresinol, (-)-lyioniresinol-3a-O-β-glucopyranoside, and estradiol. Acteoside displayed lower binding affinity energy (-9.7 kcal/mol) compared to estradiol (-8.9 kcal/mol) and was the lowest among all compounds. Acteoside was found to weaken the interaction between CUL4B-Rbx1 and CUL4B-DDB1 but not between AhR and ERα. Consequently, acteoside could be a viable candidate as an antiosteoporosis agent by inhibiting ERα degradation via the CUL4B-DDB1-AhR pathway. Further biochemical, in vitro, and in vivo studies are required to strengthen this evidence.

Abstract

COVID-19 is associated with disseminated lung damage in patients, Dexamethasone can reduce lung injury caused by inflammation and there reduce the progression to respiratory failure and prevent death. This systematic review aimed to determine the benefits and safety of Dexamethasone in COVID-19 treatment. The study was performed by a comprehensive literature search which were published in several databases i.e., PubMed, Science Direct, VHL Regional Portal, and ClinicalTrials.gov within the search time of 28 November 2020. Inclusion criteria were articles on the study on COVID-19 patients who received Dexamethasone, observational and experimental studies on the outcomes use evaluation of Dexamethasone. Exclusion criteria are the articles that do not provide control in controlled studies and do not show clear research results on the use of Dexamethasone. An initial search from four databases by entering keywords resulted in 1,046 articles. After screening articles duplication we obtained 835 studies. Finally, 6 articles were obtained after we screened for the article that it can be obtained its full text and 5 articles joined in articles included in the meta-analysis. The analysis showed that Dexamethasone in Covid-19 patients could reduce the incidence of death within 28 days with RR of 0.78 (95% CI 0.57–0.97 P=0.13) compared with Methylprednisolone, Dexamethasone was compared without corticosteroids with RR 0.89 (95% CI 0.82-0.97 P=0.01). Dexamethasone also reduced mechanical ventilator use during treatment with RR 0.95 (95% CI = 0.86-1.05 P = 0.28) compared without corticosteroids. The conclusion from these results: the use of Dexamethasone can reduce the number of deaths in COVID-19 patients, especially severe and critically ill category patients.
