Abstract

Ciguatera Fish Poisoning (CFP) merupakan salah satu tipe keracunan pada manusia yang terjadi akibat pengkonsumsian ikan-ikan karang yang telah terkontaminasi ciguatoksin. Toksin tersebut dihasilkan oleh beberapa jenis dinoflagellata bentik yang umumnya ditemukan menempel pada makroalga. Penelitian mengenai inventarisasi dan kelimpahan dinoflagellata bentik pada makroalga di perairan Pulau Tidung telah dilakukan pada tanggal 21 September 2014, mengingat lokasi tersebut telah mengalami tekanan dari berbagai aktivitas manusia. Penelitian dilakukan dengan mengkoleksi makroalga Sargassum dan Padina dari rataan terumbu Pulau Tidung, untuk kemudian dimasukkan ke dalam wadah plastik berisi air laut. Setelah itu, untuk melepaskan dinoflagellata bentik dari makroalga, dilakukan proses pengocokan dan penyaringan dengan saringan bertingkat (125μm dan 20μm). Sampel yang telah disaring kemudian diamati dengan Sedgewick rafter cells di bawah mikroskop. Dari hasil penelitian diperoleh sebelas jenis dinoflagellata bentik, dimana enam diantaranya berpotensi menyebabkan CFP yaitu Amphidinium sp., Ostreopsis lenticularis, O. siamensis, Prorocentrum concavum, P. lima, dan P. rhathymum. Jenis-jenis dinoflagellata toksik yang ditemukan mengindikasikan perlunya dilakukan monitoring di perairan Pulau Tidung.Kata kunci : Dinoflagellata bentik, Ciguatera Fish Poisoning, makroalga, Pulau Tidung

Abstract

Species composition and abundances of phytoplankton in the shellfish farming area in Jakarta Bay, Kali Baru-Cilincing, were examined in order to identify harmful algae species. Phytoplankton contained in the shellfish was also observed to detect the presence of toxic species, as well as the phycotoxin contained in  visceral of green mussel (Perna viridis) collected from the farming area using brine shrimp lethality test (BSLT). We detected the presence of  Alexandrium spp. (PSP causing species), Dinophysis caudata, D. miles and D. rotundata (DSP causing species), and Pseudo-nitzschia spp. (ASP causing species). The last mentioned species was also detected in thestomach content of green mussel. The toxicity of green mussels tested was recorded LC 50 ≤ 1,000 ppm and indicatedthe presence of an active compound in the green mussel viscera. Meanwhile, preliminary PSP test of shellfish did not showed any toxin.

Abstract

The Ciguatera Fish Poisoning (CFP) causing microorganisms were observed at the reef flat of Penjaliran Barat Islands and Pramuka Islands District, Seribu Island National Park, North Jakarta, Indonesia. Of the samples collected, four potentially toxic benthic dinoflagellates species were found, which are Gambierdiscus toxicus, Prorocentrum concavum, Prorocentrum lima, and Ostreopsis lenticularis. The dinoflagellates were found attached on the brown macroalgae Padina and Sargassum. Most of the benthic dinoflagellates from both locations were found in Pramuka Islands District, the inhabitated islands where human activities which could damage the coral reef areas were relatively high. Beside the macroalgae substrate preference of each dinoflagellate species, several environmental factors were also obtained in this research.

Abstract

Penelitian mengenai kelimpahan mikroalga epifit di perairan Teluk Hurun belum pernah dilakukan, padahal beberapa mikroalga epifit berpotensi menimbulkan Harmful Algal Blooms. Tujuan dari penelitian ini adalah untuk mengidentifikasi dan menganalisis kelimpahan mikroalga epifit pada makroalga Padina di perairan Teluk Hurun, terutama untuk spesies yang berpotensi menyebabkan HAB sehingga memungkinkan dilakukannya upaya mitigasi. Makroalga Padina diambil secara acak dari kawasan rataan terumbu, dimasukkan ke dalam botol sampel, dan dikocok kuat selama 1 menit. Sampel air disaring menggunakan saringan bertingkat, untuk kemudian diawetkan dan diamati di bawah mikroskop. Analisis eDNA juga dilakukan dengan menyaring sampel air tersebut melalui kertas millipore berukuran pori 0,45 µm menggunakan filtering set, kemudian diamplifikasi dengan set primer 18s v9, dan dilakukan pengurutan basa DNA melalui Illumina iSeq. Hasil morfogenetik menunjukkan bahwa ditemukan mikroalga dari kelompok Bacillariophycea dengan kelimpahan dan proporsi keberadaan tertinggi yaitu Nitzschia dan Navicula. Meskipun kelompok Dinophycea tidak ditemukan pada pengamatan morfologi, namun analisis eDNA menunjukkan 2 spesies yang berpotensi menyebabkan HAB, yaitu Gambierdiscus sp. dan Prorocentrum mexicanum. Sel-sel dari kelompok Dinophyceae mungkin mengalami kerusakan selama proses perlakuan atau pengawetan, namun materi genetik yang terlarut dalam sampel air dapat terdeteksi menggunakan analisis eDNA.

Abstract

Damage to coral reef ecosystems and changes in water quality are capable of increasing the abundance of benthic dinoflagellates which could cause Ciguatera Fish Poisoning (CFP). Therefore, this research aimed to observe spatial and temporal abundance of those potential benthic dinoflagellates and analyze their relationship with environmental factors. Sampling was carried out on September 21-24, 2023 in Pramuka Island, Seribu Islands district, with different treatments, namely depth, location and time. Research conducted by deploy several series of artificial substrates into the waters, and the benthic dinoflagellates were collected from the artificial substrate placed in 1, 3, and 5 meter of water depths for 24, 48, and 72 hours at Odi, Mazu, and Villa Delima Piers. Cell densities were determined by enumeration using a Sedgewick Rafter Counting Chamber under a light microscope, and the relationship with environmental parameters was analyzed by using Principal Component Analysis (PCA). The results showed that 4 genera of benthic dinoflagellates which had the potential to cause CFP, were attached to the artificial substrate, namely Amphidinium, Coolia, Ostreopsis, and Prorocentrum. Prorocentrum cells were observed in highest number on artificial substrates at every treatment with different depths, locations, and times. Furthermore, the PCA analysis showed the abundance of Prorocentrum was influenced by the most typical environmental parameters at each depth, location, and time, namely temperature, nitrate, DO, and light intensity. The research provided valuable information on the benthic dinoflagellates both spatially and temporally, through international standard methods in order to prevent and anticipate negative impacts caused by CFP.
