Abstract

Rhizosphere and endophytic fungi are functional types of microbes capable of producing secondary metabolites that can affect plant growth directly or indirectly. This study aims to isolate and test the rhizosphere and endophytic fungi' ability from Moringa (Moringa oleifera L.) against rice seeds' viability and vigor. Fungus exploration was carried out on soil samples in the rhizosphere and stem and leaf tissue of healthy Moringa plants. Isolation of fungi from the rhizosphere was carried out using 10-2 and 10-3 dilution techniques, while the isolation of endophytic fungi was carried out on the leaf and stem tissue of Moringa, then cultured on potato dextrose agar (PDA) medium. The pathogenicity test of fungi and its effect on in-vitro rice seed germination using the blotter test method, namely growing 25 rice seeds on seven-day old fungal isolates. Nineteen fungal isolates were isolated and collected from the parts of the Moringa plant. Pathogenicity observations showed that there were five fungal isolates as potential pathogens, namely isolates RF2, RF5, RF6, RF8, and EDF6. A total of four fungal isolates tested consistently showed a positive effect on seed viability and vigor with a value of ≥90%, namely isolates RF4, EDF1, EDF2, and EDFbt3.

Palembang: Penerbit & Percetakan Universitas Sriwijaya (UNSRI).Plant pathogens are one of the causes of corn yield loss in Indonesia. Evaluation of the resistance of hybrid maize genotypes against major pathogens is deemed necessary as the first step in the management of the pathogens. This study aimed to evaluate the resistance reaction of the prospective hybrid corn varieties against the primary disease of corn. This study was conducted at two different locations. The location was at the Agricultural Technology Research and Assessment Installation (ATRAI) of the Indonesian Cereals Research Institute in Bajeng District, Gowa Regency, South Sulawesi. The study was conducted using spreader row plants as a source of inoculum for the test pathogen that planted around the experimental block. Spreader rows were inoculated with conidia suspension of the fungus Bipolaris maydis for leaf blight and the pathogen Puccinia polysora for leaf rust in the afternoon with a spore density of about 6 x104 conidia/ml. The results showed that there were 16 genotypes reacted moderately resistant and 7 genotypes reacted susceptible against  B. maydis. In addition, there were 11 genotypes reacted moderately resistance and 12 genotypes that reacted susceptible against P. polysora.

Abstract

Efficiency and effectiveness in controlling crossbreeding are pivotal in yielding superior hybrid varieties with desired traits such as genetic purity and high productivity. Cytoplasmic Male Sterile (CMS) is a crucial component in hybrid variety assembly. This study aims to identify the genotypes that can induce male sterile lines based on genetic diversity and the level of pollen sterility in the prospective male sterile genotypes tested, consisting of seventeen genotypes. The research findings revealed that the genetic diversity among the seventeen observed male-sterile candidate genotypes tends to vary in quantitative traits. Among the quantitative traits, JHD14 exhibited indications of being a male sterility inducer compared to the other genotypes observed, demonstrating a value of 0.00 g in terms of pollen weight. This is further substantiated by the microscopic examination of JHD14 pollen grains, which displayed 100% sterility.Â
