Abstract

Kanker paru merupakan kanker urutan ke-3 terbanyak dan penyebab kematian utama pada pria yang kemoterapinya memberikan banyak efek samping sehingga perlu dicari kemoterapi alternatif. Kumarin psoraldin diketahui memiliki efek kemotoksik pada sel kanker  dan terkandung di dalam purwaceng (Pimpinella alpinna) namun efek ekstrak air Pimpinella alpinna sebagai agen antiproliferatif, sitotoksik, penginduksi apoptosis, dan jalur mekanismenya pada sel kanker belum diteliti secara ilmiah. Penelitian eksperimental murni dengan kultur sel kanker paru A549 ini menggunakan rentang dosis ekstrak purwaceng 1-10.000 μg/mLdan rentang dosis kontrol positif cisplatin 1-10 μg/mL yang direplikasi 6 kali serta dengan rentang waktu inkubasi 24-72 jam.  Pada uji sitotoksik dengan inkubasi 24 jam yang dideteksi dengan pewarnaan MTT didapatkan IC50sebesar 3.862 μg/mL, sedangkan uji antiproliferasi dengan dosis 550 dan 275 μg/mL mampu menghambat kecepatan pertumbuhan sel 24 jam pertama dan kedua terhadap kontrol. Uji ANOVA satu jalan menunjukkan terdapat perbedaan bermakna rerata kadar ekstrak purwaceng terhadap variasi waktu (p<0.05). Uji apoptosis yang dideteksi menggunakan fluoresensi etidium bromide-acridine orange mendapatkan nilai EC50sebesar 5.271 μg/mL yang ditandai dengan penurunan ekspresi Rb dan bcl-2 serta peningkatan p53 dan caspase-9. Statistik Chi-Square menunjukkan proporsi efek IC50 pada ekspresi gen berbeda  secara bermakna(p<0.05) terhadap kontrol. Dapat disimpulkan bahwa Pimpinella alpinna mempunyai efek antiproliferasi dan sitotoksik terhadap sel kanker paru A549 dan menginduksi apoptosis melalui perubahan jalur apoptosis berupa penurunan ekspresi Rb dan bcl-2 serta peningkatan p53 dan caspase-9. Kata kunci:  Pimpinella alpinna, kanker paru, antiproliferasi, sitotoksik, apoptosis.

Abstract

ABSTRACTMetabolic syndrome (MetS) is a group of risk factors which increase morbidity and mortality for cardiovascular disease and diabetes. The prevalence of MetS has been on the rise. No previous study has described the prevalence of MetS in Yogyakarta Special Region and its components. The study aim was to determine the prevalence of MetS and it’s components in Yogyakarta Special Region, Indonesia. A total of 766 male and female subjects aged ≥40 were analyzed in this retrospective study based on secondary data from the Indonesian Family Life Survey batch 4 (IFLS 4). MetS was defined by International Diabetes Federation (IDF) criteria with ethnicity-specific values for waist circumference. Prevalence of MetS and characteristic of each component of MetS were expressed as mean or %. The difference of the MetS components was evaluated by t-test and chi-square. Prevalence of MetS in Yogyakarta Special Region was 13.19%. The most common of MetS component was hypertension (60.44%), followed by dyslipidemia (56.27%), central obesity (32.38%), pro-inflammatory state (15.71%) and insulin resistance (0.78%). There was a higher prevalence of MetS in females compared to males (15.88 vs 10.19%), pre-elderly compared to the elderly (13.90 vs 12.19%), Javanese compared to other ethnicities (13.23 vs 10.00%), and urban compared to rural populations (15.06 vs 8.37%). In conclusion,the prevalence of MetS in Yogyakarta Special Region is 13.19% with hypertension and dyslipidemia as the most common component.

Abstract

BACKGROUND: Secretome production by stem cells depends on their culture conditions such as oxygen concentration and the composition of the culture media. In this study, we investigated the secretion of neurotrophic growth factors of human umbilical cord mesenchymal stem cells (hUC-MSCs) in amino acid-rich culture medium and under hypoxic condition.METHODS: hUC-MSCs were cultured in normoxic and various hypoxic (1%, 5%, 10%) conditions in an amino acid-rich culture medium. The end-point parameters (cell proliferation and survival, cell morphology and growth factor secretion) were measured at 3 time-points (48 hours, 72 hours and 96 hours). ELISA-based methods were used for neurotrophic factors detection, including neurotrophic growth factor (NGF), vascular endothelial factor (VEGF), and brain-derived neurotrophic factor (BDNF).RESULTS: NGF secretion was not detectable at any time points both in normoxia and hypoxia. BDNF secretion under normoxia was induced at 48 h time point and reached the highest level at an average of 181.9±13.01 pg/mL at 96 hours, whereas hypoxia exposure to hUC-MSCs only induced the BDNF secretion at low level. VEGF secretion was barely detectable in normoxic condition. However, VEGF secretion reached the highest level at an average of 7707.55±2110.85 pg/mL in 5% hypoxia at 96 hours.CONCLUSION: Combination of amino acid-rich culture medium and hypoxia condition dramatically induced high VEGF secretion by hUC-MSCs, especially at 5% hypoxia, induced mild BDNF secretion and had no effect toward NGF secretion.KEYWORDS: human umbilical cord mesenchymal stem cells, neurotrophic growth factor, amino acid-rich, hypoxia

Abstract

Insulin resistance as a cause of type 2 diabetes mellitus is associated with subclinical inflammatory processes. Insulin resistance with obesity, hypertension, and dyslipidemia contribute to metabolic syndrome that increased risk of cardiovascular disease. High sensitivity C-reactive protein (hs-CRP) is an inflammatory marker that is thought to be associated with both type 2 diabetes mellitus and cardiovascular disease. This study evaluated hs-CRP, HbA1c, and body mass index in a healthy community. This cross-sectional study is an observational analytic study evaluating the association between hs-CRP, HbA1c, and body mass index. The research subjects were all healthy on a community gathering in community service programs, and if there were any signs or symptoms of infection or inflammation, they would be excluded. Measurements of hs-CRP and HbA1c were carried out using the HPLC and ELISA methods, respectively. The measurement results were analyzed to evaluate the characteristics of the subject and assess the relationship between the parameters studied with different mean and correlation tests. In 25 subjects involved, it was found that 96% had an HbA1c value of <6.5% with a normal body mass index of 15 subjects (60%), and the rest were in the category of overweight. The median hs-CRP level was 2.99 mg / L (0.81-13.74 mg / L), with a low heart risk category of only 4% of all study subjects. There was no correlation between hs-CRP with HbA1c (r = 0.35; p = 0.868) and body mass index (r = 0.37; p = 0.069). Only one subject was included in the diabetes diagnostic criteria, but 96% of the study population had hs-CRP, which was included in the medium-risk and high-risk category for heart disease. There was no association between hs-CRP and HbA1c and body mass index in healthy populations in this community.

Abstract

BACKGROUND: Hypoxia adaptation in cancer cells is mediated by hypoxia inducible factor (HIF)-1 that induces the expression of various proteins. Up to date, the analysis of T47D breast cancer cells-secreted protein is still limited. The aim of this study was to compare the protein profile secreted by T47D breast cancer cells under hypoxic and to normoxic conditions and identify the proteins as candidate for hypoxia marker proteins in T47D breast cancer cells.METHODS: T47D breast cancer cells were cultured under standard conditions. Cells were subcultured in normoxic and hypoxic conditions. The normoxic group was incubated with 20% oxygen and the hypoxic groups were incubated in a hypoxic chamber with 0.5% and 5% oxygen for 6, 24, and 48 hours in serum free medium. Proteins in the culture media were isolated and precipitated. Protein concentrations released by the cells were then measured by bicinchoninic assay (BCA). Protein bands were visualized by the sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) method. The bands that had differences between the hypoxic and normoxic groups were further analyzed by liquid chromatography-mass spectrophotometer (LC-MS).RESULTS: Hypoxic groups showed higher secretory protein than normoxic group. Protein bands were found in the 0.5% hypoxic group with a size of 50-75 kDa. Secretory proteins identified by LC-MS were keratin 1, 2, 9, and 10.CONCLUSION: The T47D cell line under 0.5% hypoxic treatment showed higher secretory proteins that identified as keratin 1, 2, 9, and 10.KEYWORDS: hypoxia, secretory protein, liquid chromatography-mass spectrophotometer
