Abstract

Tuberculosis (TB) is one of the infectious diseases with a high mortality rate worldwide, including in Indonesia. Acid-Fast Bacilli (AFB) microscopic examination using the Ziehl-Neelsen method remains a widely used diagnostic method for TB. One of the crucial stages in Ziehl-Neelsen staining is the decolorization process using an acid-alcohol solution. Variations in the pH of the decolorizing solution are suspected to affect the quality of AFB staining results. This study aimed to determine the effect of acid-alcohol pH variations on the quality of AFB staining using the Ziehl-Neelsen method. The study utilized a laboratory experimental analytical design with acid-alcohol pH variations of 0.5, 1.0, 2.0, and 3.0. The research sample consisted of 24 smears, which were observed based on the microscopic quality of the AFB staining results. Data were analyzed using the Kruskal-Wallis test, followed by a post hoc pairwise comparison test using the Bonferroni correction. The Kruskal-Wallis test results showed a significant effect of acid-alcohol pH variation on the quality of AFB staining (p < 0.05). Post hoc test results indicated significant differences between pH 0.5 and pH 2.0 (p = 0.038), pH 0.5 and pH 1.0 (p < 0.001), as well as pH 3.0 and pH 1.0 (p = 0.013). The pH 1.0 group had the highest mean rank of 21.50, indicating the best staining quality in accordance with the AFB examination quality standards set by the Ministry of Health of the Republic of Indonesia. Therefore, variations in acid-alcohol pH affect the quality of Ziehl-Neelsen AFB staining, and pH 1.0 is recommended as the optimal pH for the decolorizing solution

Abstract

Abstract:  Leptospirosis is a zoonotic disease that remains a significant public health problem, particularly in areas with poor environmental sanitation and high-risk community activities. This community service program aimed to improve public knowledge and awareness regarding the risks and prevention of leptospirosis in Sobayan Village, Pedan District, Klaten Regency. The methods employed included interactive health education sessions, basic health screenings (blood glucose and blood pressure), and training on the preparation of natural antiseptic soap as part of promotive and preventive efforts. Program evaluation was conducted using a pre-test and post-test design to assess changes in participants’ knowledge levels. The results demonstrated a significant increase in community knowledge, as indicated by the rise in the average score from 71.21 in the pre-test to 91.89 in the post-test. In addition, the program contributed to enhancing community awareness in adopting clean and healthy living behaviors (PHBS). In conclusion, this community service activity was effective in improving knowledge and promoting behavioral change as part of leptospirosis prevention efforts.Abstrak: Leptospirosis merupakan penyakit zoonosis yang masih menjadi masalah kesehatan masyarakat, terutama pada wilayah dengan sanitasi lingkungan yang kurang memadai dan aktivitas masyarakat yang berisiko tinggi. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan kesadaran masyarakat mengenai bahaya serta upaya pencegahan leptospirosis di Desa Sobayan, Kecamatan Pedan, Kabupaten Klaten. Metode pelaksanaan meliputi penyuluhan kesehatan interaktif, pemeriksaan kesehatan dasar berupa skrining kadar gula darah dan tekanan darah, serta edukasi pembuatan sabun antiseptik alami sebagai upaya promotif dan preventif. Evaluasi dilakukan menggunakan desain pre-test dan post-test untuk mengukur perubahan tingkat pengetahuan peserta. Hasil kegiatan menunjukkan adanya peningkatan pengetahuan yang signifikan, ditunjukkan oleh kenaikan nilai rata-rata dari 71,21 pada pre-test menjadi 91,89 pada post-test. Selain itu, kegiatan ini juga berkontribusi dalam meningkatkan kesadaran masyarakat terhadap penerapan perilaku hidup bersih dan sehat (PHBS). Dengan demikian, kegiatan pengabdian ini efektif dalam meningkatkan pengetahuan dan mendorong perubahan perilaku masyarakat dalam upaya pencegahan leptospirosis
