Abstract

In vitro regeneration of buds and shoots via organogenesis in two genotypes of snake plants (Sansevieria trifasciata) were established. Leaf segments (1 × 1cm) of ‘Hahnii’ and’Lorentii’ were cultured on Murashige and Skoog (MS) basal medium containing 2,4- dichlorophenoxyacetic acid (2,4-D) for 1 week, transferred into MS medium without plant growth regulator (MS0) for 1 week, and then cultured on MS medium containing different concentrations of N6-benzyladenine (BA) at 0, 0.1, 0.25, 0.5, 1.0 and 2.0 mg/l in three times of serial passages with 4 week intervals. The results showed that regeneration of S. trifasciata was established via indirect organogenesis. ‘Lorentii’ was more responsive on shoot regeneration than ‘Hahni’. The most suitable BA concentration for shoot bud and shoot regeneration for both cultivars was 2 mg/l. Explants of ‘Lorentii’ cultured on MS medium containing 2.0 mg/l BA produced 8.6 shoot buds and 3.4 shoots per explant after 14 weeks. Shoots derived from both cultivars were successfully ex vitro rooted and acclimatized in the greenhouse with high survival rate, i.e., from 96 -100%. Abstract

Penyakit bilur kacang tanah yang disebabkan oleh peanut stripe virus (PStV)merupakan salah satu penyakit utama pada pertanaman kacang tanah (Arachishypogaea L.). Penggunaan varietas tahan merupakan alternatif paling efektif untukmengatasi penyakit tersebut. Rekayasa genetika merupakan metode efektif untukmendapatkan varietas kacang tanah yang resisten PStV. Tujuan penelitian ini adalahuntuk (1) mengetahui respons tanaman kacang tanah transgenik yang membawa gen cpPStV terhadap infeksi PStV dan (2) menguji stabilitas transgen sampai tujuh generasisilang-dalam. Tanaman kacang tanah transgenik cv. Gajah generasi T0, T1, T2, T3, T5,T6, dan T7 diinokulasi secara mekanik dengan PStV. Terdapat tiga jenis respons kacangtanah transgenik terhadap infeksi PStV, yaitu resisten, recovery, dan rentan. Padatanaman resisten gejala tidak muncul. Pada tanaman recovery, gejala chlorotic ringmottle muncul pada satu daun atau lebih, selanjutnya gejala tidak tampak pada daundaunyang tumbuh kemudian. Pada tanaman rentan, gejala severe blotch muncul padasuatu daun, selanjutnya gejala tersebut tetap muncul pada seluruh daun yang tumbuhkemudian. Transgen cp PStV tetap stabil setelah mengalami tujuh generasi silangdalam.Sejumlah galur murni kacang tanah transgenik yang resisten PStV telahdiidentifikasi.

Abstract

This research aimed to study effects of IBA, NAA and their combination on rooting and shoot sprouting in Malay apple cuttings. Cuttings from superior genotype were collected and treated with (in ppm w/w): 0, 2000 IBA, 4000 IBA, 2000 NAA, 4000 NAA, 1000 IBA+1000 NAA, 2000 IBA+2000 NAA. To record the timing and percentage of rooting, cuttings were treated with (in ppm w/w) 1000 IBA+1000 NAA or without auxin as control. The results revealed that application of auxin was significantly enhanced root formation as shown by the significant increases in rooting percentage and number of roots. NAA at 2000 or 4000 ppm was the most effective auxin to promote root formation (100 %, 17.8–25.5 roots per cuttings), followed by NAA+IBA (100 %, 16.8– 9.8 roots per cuttings) and the least effective was IBA alone (79-100 %, 3.2–7.1 roots per cutting). The best treatment for rooting and shoot sprouting were (in ppm) 1000 IBA+1000 NAA, since it produced higher root length, better root morphology and higher shoot sprouting. It was also found that a combination of IBA+NAA each at 1000 ppm not only enhanced root percentage, but also shortened the time for root formation.

Abstract

Jambu air (Syzygium aqueum (Burm. f.) Alston) tergolong tanaman mudah beradaptasi serta dapat tumbuh pada berbagai macam jenis tanah. Salah satu alternatif penyediaan benih vegetatif yaitu dengan cara cangkok. Percobaan ini dilakukan menggunakan Rancangan acak kelompok (RAK) dengan 4 ulangan, perlakuan disusun secara faktorial (2x4) Faktor pertama merupakan varietas jambu air Citra Merah dan Madu Deli Hijau. Faktor kedua merupakan jenis auksin yang meliputi tanpa pemberian auksin, pemberian IAA 2000 ppm, IBA 2000 ppm, NAA 2000 ppm dan setiap perlakuan diulang 3 kali. Pengamatan dilakukan untuk melihat variabel jumlah akar, panjang akar, jumlah tunas, dan panjang tunas. Dilakukan analisis data dengan Uji Beda Nyata Terkecil taraf nyata 5%.Hasil penelitian percobaan menunjukkan bahwa pada jenis jambu Madu Deli Hijau memiliki nilai tertinggi sedangkan jambu Citra Merah memiliki nilai terendah. Pada jenis auksin IAA menunjukan hasil yang lebih baik dibanding NAA dan IBA. Simpulan dari penelitian ini yaitu Varietas jambu air berpengaruh terhadap rata-rata jumlah akar pada cangkok Madu Deli Hijau 8.67 helai lebih besar dari pada jambu Citra Merah 5.17 helai. Namun rata-rata bobot segar akar dan bobot kering akar lebih besar Citra Merah 4.364 gram dan 0.982 gram dari pada Madu Deli Hijau 3.115 gram dan 0.680 gram. Pada 10 MSP (Minggu Setelah Pencangkokan) tanpa pemberian auksin hanya 50% yang berakar sedangkan dengan aplikasi auksin NAA, IBA dan IAA semuanya berakar 100%.
