Abstract

In vitro regeneration of buds and shoots via organogenesis in two genotypes of snake plants (Sansevieria trifasciata) were established. Leaf segments (1 × 1cm) of ‘Hahnii’ and’Lorentii’ were cultured on Murashige and Skoog (MS) basal medium containing 2,4- dichlorophenoxyacetic acid (2,4-D) for 1 week, transferred into MS medium without plant growth regulator (MS0) for 1 week, and then cultured on MS medium containing different concentrations of N6-benzyladenine (BA) at 0, 0.1, 0.25, 0.5, 1.0 and 2.0 mg/l in three times of serial passages with 4 week intervals. The results showed that regeneration of S. trifasciata was established via indirect organogenesis. ‘Lorentii’ was more responsive on shoot regeneration than ‘Hahni’. The most suitable BA concentration for shoot bud and shoot regeneration for both cultivars was 2 mg/l. Explants of ‘Lorentii’ cultured on MS medium containing 2.0 mg/l BA produced 8.6 shoot buds and 3.4 shoots per explant after 14 weeks. Shoots derived from both cultivars were successfully ex vitro rooted and acclimatized in the greenhouse with high survival rate, i.e., from 96 -100%. Abstract

Attempts to identify somaclonal variants of peanut with resistance to Sclerotium stem rot disease due to infection of S. rolfsii were conducted. The objectives of this study were to develop in vitro selection method using culture filtrates of S. rolfsii, identify culture filtrate-insensitive somatic embryo (SE) of peanut after in vitro selection and regenerate peanut R0 lines originated from culture filtrate-insensitive SE. To achieve these objectives, peanut embryogenic tissues were cultured on selective medium containing various concentrations of S. rolfsii culture filtrates and sublethal concentration of the filtrates. Medium containing sublethal level of S. rolfsii culture filtrates was used to identify culture filtrate-insensitive SE of peanut. Subsequently, the selected SEs were germinated, plantlets were regenerated and preliminary tested against S. rolfsii. Results of the experiments showed that addition of S. rolfsii culture filtrates into medium for inducing peanut somatic embryos drastically reduced their growth and proliferation. S. rolfsii culture filtrates at 10% concentration has significantly reduced the number of proliferated SE per explant. However, sublethal level was achieved at 30% of culture filtrates concentration. Responses of five peanut cultivars against 30% of culture filtrates were similar, indicating they were similar in their susceptibility against S. rolfsii. A number of culture filtrate-insensitive SE were identified after culturing 1500 clumps of embryogenic tissue of peanut cv. Kelinci for three consecutive passages on medium containing 30% of culture filtrates. Germination of selected SE and regeneration of plantlet from culture filtrate-insensitive SE resulted in 50 peanut R0 lines. These lines have been grown in the plastic house and produced normal seeds for further evaluation. Results of S. rolfsii inoculation indicated the existence of chimera for insensitivity against S. rolfsii.

Abstract

Tingginya AKI tidak terlepas dari kejadian perdarahan, eklamsi dan infeksi. Disisi lain penyebabnya adalah masih banyak ditemukan masalah kehamilan yang tidak ideal (terlalu banyak anak, usia ibu terlalu muda, usia ibu terlalu tua, dan terlalu dekat jarak kehamilan) yang sangat membahayakan bagi keselamatan ibu yang lebih dikenal dengan 4T. Kehamilan yang berisiko tinggi mengundang masalah sampai keselamatan jiwa saat melahirkan. Penelitian ini dilakukan untuk melihat hubungan pengetahuan dan sikap ibu hamil dengan kehamilan risiko tinggi (4T) di BPM Desita, S.SiT Desa Pulo Ara Kecamatan Kota Juang Kabupaten Bireuen.Desain penelitian yang digunakan dalam penelitian ini yaitu analitik cross sectional. Populasi dalam penelitian ini adalah seluruh ibu hamil yang datang ke BPM Desita, S.SiT Desa Pulo Ara Kecamatan Kota Juang Kabupaten Bireuen. Teknik pengambilan sampel dalam penelitian ini yaitu dengan cara accidental sampling sebanyak 42 orang.Dari hasil penelitian yang dilakukan terhadap 42 responden yang dilakukan pada Februari 2021 menunjukkan bahwa ada hubungan antara pengetahuan dengan kehamilan risiko tinggi (4T) di BPM Desita, S.SiT Desa Pulo Ara Kecamatan Kabupaten Bireuen. Ada hubungan antara sikap dengan kehamilan risiko tinggi (4T) di BPM Desita, S.SiT Desa Pulo Ara Kecamatan Kabupaten Bireuen.         Diharapkan kepada responden untuk lebih maksimal lagimen dalam konsep tentang risiko tinggi 4T pada masa kehamilan, sehingga ibu tidak lagi merencanakan kehamilan jika berisiko terjadi masalah selama hamil ataupun efek pada persalinannantinya. Kata Kunci                  : Pengetahuan, Sikap, Risiko Tinggi 4T

Abstract

Fisherman are a group of people who are highly dependent on the fisheries resources in their efforts to fulfill their basic needs and to be a prosper. The outbreak of the Covid-19 not only threatens people’s lives and safety, but also has a significant impact on economic development both of which are  macro and micro to be weakened. This research aims to determine the impacts of the Covid-19 Pandemic on Fishing activities, incomes, and marketing activities of purse seiner. The research was conducted on October 2020 in the Banabungi Village, Kadatua Subdistrict, South Buton Regency, Southeast Sulawesi Province. The results show that the impact of the Covid-19 has affected purse seiner’s fishing activities where production costs have increased by 4,62%, The of crews (ABK) have increased by 19,69%, and marketing at channel I (Producer-Wholesale)stopped. Consequently, where the change in fish price at fishemen level by 79,75%. The margin of marketing at channel II at wholesale level is IDR 5.556/kg. Meanwhile, at retail level is IDR 8.267/kg. Production was decreased by 79,93%, revenue decreased by 64,06% and profit decreased by 68,26% with value of R/C ratio 3.70 >1. The fishing location of fishermen has not change as in the previous year. Marketing volume during the peak fishing season is 14.809kg/month.

Abstract

Jambu air (Syzygium aqueum (Burm. f.) Alston) tergolong tanaman mudah beradaptasi serta dapat tumbuh pada berbagai macam jenis tanah. Salah satu alternatif penyediaan benih vegetatif yaitu dengan cara cangkok. Percobaan ini dilakukan menggunakan Rancangan acak kelompok (RAK) dengan 4 ulangan, perlakuan disusun secara faktorial (2x4) Faktor pertama merupakan varietas jambu air Citra Merah dan Madu Deli Hijau. Faktor kedua merupakan jenis auksin yang meliputi tanpa pemberian auksin, pemberian IAA 2000 ppm, IBA 2000 ppm, NAA 2000 ppm dan setiap perlakuan diulang 3 kali. Pengamatan dilakukan untuk melihat variabel jumlah akar, panjang akar, jumlah tunas, dan panjang tunas. Dilakukan analisis data dengan Uji Beda Nyata Terkecil taraf nyata 5%.Hasil penelitian percobaan menunjukkan bahwa pada jenis jambu Madu Deli Hijau memiliki nilai tertinggi sedangkan jambu Citra Merah memiliki nilai terendah. Pada jenis auksin IAA menunjukan hasil yang lebih baik dibanding NAA dan IBA. Simpulan dari penelitian ini yaitu Varietas jambu air berpengaruh terhadap rata-rata jumlah akar pada cangkok Madu Deli Hijau 8.67 helai lebih besar dari pada jambu Citra Merah 5.17 helai. Namun rata-rata bobot segar akar dan bobot kering akar lebih besar Citra Merah 4.364 gram dan 0.982 gram dari pada Madu Deli Hijau 3.115 gram dan 0.680 gram. Pada 10 MSP (Minggu Setelah Pencangkokan) tanpa pemberian auksin hanya 50% yang berakar sedangkan dengan aplikasi auksin NAA, IBA dan IAA semuanya berakar 100%.
