Abstract

The aim of this study is to construct the hGAD65 gene and to identify the hGAD65 clone by using PCR & RFLP. The samples were derived from normal person & DM patient’s blood. Blood DNA was isolated by salting out method and then amplified by PCR with a pair of specific primer, GAD65-F-BamH1-807 & GAD65-R-Xho1-945. The PCR-product was cloned into vector pET-28a and the pET28a-hGAD65-clone was transformed into E.coli BL21 competent cells. The pET28a-hGAD65-clone was confirmed by PCR and RFLP by BamH1 & XhoI. The PCR product of pET28a-hGAD65-clone was one band of 159bp and has two bands 5.3 kb and 159 bp by RFLPwith both restriction enzymes. The GAD65 protein is expressed in 65kD of pET28a-hGAD65-clone. PET28a-hGAD65-clone was able to recognize by gold standard monoclonal antibody specifically. These results indicated that the hGAD65 gene inserted into pET28a properly and provided the GAD65 protein expression.Key words: hGAD65, PCR, pET-28a, RFLP

Abstract

Diabetes mellitus is one of metabolic disorder with high level of blood sugar glucose and reduces pancreatic-insulin levels that are altered by unhealthy food and lifestyle. To manage the blood glucose and insulin level, we need the diabetes dietary management related with insulin gene cascade. The purpose of this study was to investigate the influence glucomannan fiber to mRNA level of the insulin gene and genes cascade expression of target cells of type 1 diabetic rat. Three-month-old male (Rattus norvegicus strain Wistar) were fed with fiber of glucomannan of Amorphophallus muelleri Blume with dosage 0,06g/kg BW and 0.12g/kg BW every day during one month. RNA dot blotting with specific cDNA probe was performed mRNA level of insulin gene. The result of this study showed that glucomannan could reduce of body weight and blood glucose level significantly. The mRNA level of insulin gene of diabetic rats-treated 0.12g/kg BW glucomannan fiber dosage was the highest significance level among of all groups. In histopathological analysis, glucomannan was increasing significantly in IRS-1 level expression proliferate properly and consistently increasing of the PI3-K expression level in treated diabetic rats. This study indicates that the 0.12g/kg BW glucomannan was optimum dosage that effectively as role for alternative high-fiber therapy of type 1 diabetic through insulin receptor tyrosine kinase pathway.

Abstract

Sebagian besar masyarakat masih belum mengetahui manfaat yang terkandung pada susu kambing, masyarakat masih menganggap bahwa susu sapi merupakan satu-satunya sumber nutrisi hewani yang memiliki kandungan protein, gizi, dan manfaat yang sangat besar bagi kesehatan. Untuk itu perlu dilakukan sosialisasi tentang pemanfaatan susu kambing karena kurangnya pengetahuan masyarakat sehingga menyebabkan rendahnya daya beli masyarakat. Tujuan dari kegiatan ini adalah untuk menjalin rintisan kerjasama dengan sekolah sebagai salah satu tempat pemasaran produk susu kambing serta orang tua siswa-siswi dalam meningkatkan daya konsumen masyarakat terhadap susu kambing dan memberikan informasi yang berlandaskan data ilmiah tentang manfaat susu kambing. Sosialisasi dilaksanakan melalui pemaparan pemateri tentang manfaat dan pentingnya mengonsumsi susu kambing dalam kehidupan sehari-hari serta pemanfaatan susu kambing menjadi produk nutrisi sehat (susu kambing pasteurisasi). Hasil dari kegiatan sosialisasi adalah para guru dan siswa-siswi bertambah pengetahuannya tentang pentingnya mengonsumsi susu kambing sebagai pangan sehat bernutrisi dan antusias dalam membuat produk-produk sehat berbahan dasar susu kambing.
