Abstract

The aim of this study is to construct the hGAD65 gene and to identify the hGAD65 clone by using PCR & RFLP. The samples were derived from normal person & DM patient’s blood. Blood DNA was isolated by salting out method and then amplified by PCR with a pair of specific primer, GAD65-F-BamH1-807 & GAD65-R-Xho1-945. The PCR-product was cloned into vector pET-28a and the pET28a-hGAD65-clone was transformed into E.coli BL21 competent cells. The pET28a-hGAD65-clone was confirmed by PCR and RFLP by BamH1 & XhoI. The PCR product of pET28a-hGAD65-clone was one band of 159bp and has two bands 5.3 kb and 159 bp by RFLPwith both restriction enzymes. The GAD65 protein is expressed in 65kD of pET28a-hGAD65-clone. PET28a-hGAD65-clone was able to recognize by gold standard monoclonal antibody specifically. These results indicated that the hGAD65 gene inserted into pET28a properly and provided the GAD65 protein expression.Key words: hGAD65, PCR, pET-28a, RFLP

Abstract

The objective this research is to study the isolation and characterization of inhibin from goat granulosa cells.  Sample of ovary collected from abbatoir of Sukun in Malang. Minimum diameter of follicle that can be used in this research is 3 mm. Granulosa cells Oocytes collection was done by aspiration method, and granulosa cells separate from oosit mechanically. Biochemistry characterization of inhibin was done by technique of SDS PAGE, biuret and PAS. The result of research confirmed that inhibin has molecular 32 kDa. Composition of protein, carbohydrate, and glikoprotein inhibin was 3.636±0,002; 2.569±0,002; and 6.205±0.016 mg/mL, respectively. Abstract

Bakterial selulose merupakan selulosa yang dihasilkan dari fermentasi gula dalam suasana asam oleh Acetobacter xylinum. Gugus fungsi dari bacterial elulosa dapat dimodifikasi  dengan berbagai cara yaitu esterifikasi,eterifikasi dan alkilasi. Esterifikasi dapat dilakukan secara enzimatis. Tujuan penelitian ini adalah menentukan waktu reaksi optimum dan rasio reaktan optimum esterifikasi bakterial selulosa secara enzimatis.Bakterial selulosa dipreparasi dengan fermentasi air kelapa pada pH 4 dan kadar sukrosa 12% oleh Acetobacter xylinum selama 7 hari. Selulosa yang dihasilkan dicuci sampai bebas asam asetat , digerus dalam keadaan basah, dikeringkan pada suhu 50°C. selanjutnya selulosa kering digerus sehingga dihasilkan serbuk selulosa. Esterifikasi dilakukan pada berbagai variasi waktu reaksi ( 6-30jam)  dan rasio berat selulosa: asam sitrat (1:1-1:5) , 10mL t-butanol  dan 0,1 g lipase amobil pada suhu 50°C. Sisa asam yang tidak bereaksi ditentukan dengan titrasi alkalimetri. Ester selulosa yang dihasilkan diidentifikasi dengan Spektrofotometer FTIR  dan dikarakterisasi  untuk Indeks Swelling( IS) dan  Derajat Substitusi (DS) . Hasil penelitian menunjukkan bahwa waktu reaksi optimum esterifikasi diperoleh 24 jam dan ratio selulosa asam sitrat (1:4)  menghasilkan sel;ulosa sitrat dengan DS 0,34 dan IS 33,90% . Spektra FTIR  selulosa sitrat menunjukkan puncak pada bilangan gelombang pada 1743, 1141 -1176 Cm-1   Katakunci:  asam sitrat , lipase amobil , Mucor miehei, selulosa

Abstract

Differences in the accumulation of lead heavy metal among plant organs (leaves, roots and rhizomes) were examined in the seagrass Thalassia hemprichii. Samples were taken from three coastal waters in Ambon Island, with total of nine sampling sites encompassing by seagrass distribution and also based on the land use. Lead heavy metal content was analyzed on the seagrass organs, water and sediment by atomic absoption spectrophotometer. It appeared that leaves, roots and rhizomes of T. hemprichii were able to uptake lead from water and sediment. Lead concentration was significantly higher in sediment and roots than water, leave and rhizome. Therefore, roots can be used to determine heavy metal of Pb distribution in coastal waters.

Abstract

There is an increasing amount of evidence that oxidative stress related to hypertension can damage the function of diverse structures such as aorta. It is a well-established fact that chlorogenic acid and anthocyanine found in purple sweet potato generates bioactive compound with antihypertensive and antioxidant activities. The present study sought to investigate antioxidant activity of extract ethanol of purple sweet potato (EP) in deoxycorticosterone acetate (DOCA–salt)–induced hypertensive rats (Rattus norvegicus). The rats were orally administrated a 95% ethanol extract of purple sweet potato (var. Ayamurasaki) (EP) in a daily dose of 200 and 400 mg/kg body weight for 4 weeks. Aorta total malondialdehyde (MDA) and histopathology of aorta abdominal were examined. Aorta injury was observed in DOCA-salt hypertensive group rats compared to normotensive group rats, as aorta MDA significantly increased  (P<0.05). In contrast, treatment of DOCA-salt hypertensive rats with different dose of EP significantly reduced the total aorta MDA, as well as repair kidney damage, suppressed smooth muscle cell proliferation and lessen aorta wall thickening compared to controls. This is the first report that demonstrated blood pressure lowering and antioxidative effects of an ethanol extract of purple sweet potato, containing chlorogenic acid, in a DOCA–salt model of hypertension.

Abstract

The roses known to contain anthocyanins pigments and as a bioactive compound  which  can act as free radical catcher material. When harvest season arrives in number will be very  abundant.  Flowers sorting which can decrease the selling price can be processed in the form  of natural dyes for food, cosmetics and medicines / herbs. The purpose of this study is to know  applications rose to the quality of some industrial products. The experiment was conducted in  (fresh, 2 and 4 days), that performed using randomized block design. Phase II is the application of pigment products of red roses on some industrial products (ie food products, herbal and  cosmetics), using the design in accordance with their respective target. The results showed that  ucts such as fruit juice, yoghurt, jelly, carbonated beverage (food) and body lotion (cosmetic),  and can contribute to the natural color at a time serves as a source of bioactive in effervescent tablets.

Abstract

ABSTRAKKonsumsi tinggi lemak berperan dalam resistensi insulin dan peningkatan risiko penyakit kardiovaskuler. Teh hijau klon GMB 4 merupakan hasil pengembangan teknologi yang dihasilkan oleh Lembaga Penelitian Teh dan Kina Gambung, dan telah  dilakukan    isolasi  dan  purifikasi  Epigallocatechin  Gallate.  Tujuan  penelitian  ini  untuk  membuktikan  pengaruh Epigallocatechin Gallate dalam menghambat resistensi insulin, penurunan ekspresi p110 PI3K dan peningkatan p38 MAPK pada aorta tikus yang diberi diet tinggi lemak. Hewan coba tikus jantan dibagi menjadi 5 kelompok perlakuan yaitu diet normal,  diet  tinggi  lemak,  diet  tinggi  lemak  dengan  penambahan  EGCG  1  mg/kgbb/hari,  2  mg/kgbb/hari  dan  8 mg/kgbb/hari  yang  diberikan  dalam  60  hari.  Parameter  yang  digunakan  adalah  resistensi  insulin  (metode  HOMA), ekspresi p110 PI3K dan p38 MAPK dari aorta tikus yang diukur dengan metode ELISA. Hasil penelitian menunjukkan bahwa isolat  EGCG  menghambat  resistensi  insulin  pada  dosis  8  mg/kgbb/hari.  EGCG  yang  diberikan  bersamaan  dengan  diet tinggi lemak dapat menghambat penurunan kadar p110 PI3K pada dosis 2 mg/kgbb/hari dan dosis 8 mg/kgbb/hari dan menghambat peningkatan aktivitas p38 MAPK pada semua dosis perlakuan yaitu 1 mg/kgbb/hari, 2 mg/kgbb/hari dan 8 mg/kgbb/hari.  Pemberian  EGCG  dosis  8  mg/kgbb/hari  yang  diberikan  bersamaan  dengan  diet  tinggi  lemak  dapat mengambat terjadinya resistensi insulin yang ditunjukkan dengan penghambatan peningkatan p38 MAPK dan penurunan p110 PI3K dan nilai HOMA dalam range normal, superoxide dismutase.Kata Kunci : Diet tinggi lemak, EGCG, p110 PI3K, p38 MAPK, resistensi insulin

Abstract

Autoimmune thyroiditis (AITD) merupakan penyakit autoimun pada kelenjar tiroid. Tujuan penelitian ini adalah mengetahui profil regulasi hormon tiroid, profil siklus estrus dan folikulogenesis pada tikus model hypothyroidism hasil induksi capra thyroglobulin (cTg). Pembuatan tikus model hipotiroid dilakukan dengan injeksi dengan protein cTg yang sudah diemulsi complete freund's adjuvant (CFA)  dengan dosis 200µg/µL melalui subkutan, selanjutnya dilakukan booster injeksi protein cTg yang sudah diemulsi incomplete freund's adjuvant (IFA) dengan dosis 200µg/µL melalui subkutan sebanyak dua kali setiap 12 hari. Serum tikus diambil untuk mengetahui tikus sudah mengalami kondisi hipotiroiditis. Tikus yang sudah mengalami hipotiroid, diamati siklus estrusnya sebanyak tiga kali siklus estrus dan setiap satu kali pengulangan siklus estrus diambil serum darah untuk diukur kadar hormon tiroksin (T4) dan thyroid stimulating hormone (TSH). Pada akhir tahapan penelitian, tikus dibedah dan dikoleksi organ ovarium untuk pembuatan preparat histologi. Hasil penelitian menunjukkan bahwa induksi cTg pada tikus menyebabkan tiroiditis dan selanjutnya mengakibatkan kondisi hipotiroid yang ditandai dengan penurunan kadar hormon tiroksin dan peningkatan thyroid stimulating hormone (TSH) selama tiga kali siklus estrus. Tikus hipotiroid mengalami periode siklus estrus yang lebih panjang dan penurunan jumlah folikel de graff dibandingkan dengan tikus normal.

Abstract

Diabetes mellitus(DM) merupakan penyakit gangguan metabolisme yang ditandai dengan meningkatnya kadar glukosa darah  (hiperglikemia)  akibat  gangguan  sekresi  insulin  dan  atau  meningkatnya  resistensi  insulin.  T erapi  ekstrak  metanol  biji mahoni (Swetenia  mahagoni Jacq)  merupakan  salah  satu  pengobatan  alternatif  penyakit  DM. Penelitian  ini  dilakukan untuk membuktikan pengaruh pemberian terapi ekstrak metanol biji mahoni terhadap kadar insulin,  ekspresi TNF-α  dan perbaikan  jaringan  pankreas  pada  tikus  hasil  induksi  Multiple  Low  Dose-Streptozotocin  (MLD-STZ)  dosis  20  mg/kgBB selama 5 hari berturut-turut. Tikus diukur kadar glukosa darah dengan menggunakan glukometer digital dan dinyatakan DM bila kadar glukosa darahnya ≥ 300 mg/dl. Penelitian ini menggunakan 25 ekor tikus putih (Rattus novergicus) jantan galur wistar yang dibagi menjadi 5 kelompok yaitu kelompok 1 kontrol negatif ,  kelompok 2    kontrol positif ,  kelompok 3, 4 dan 5 tikus hasil induksi MLD-STZ serta masing-masing diberikan terapi ekstrak metanol biji mahoni  dosis 100; 250 dan 400 mg/kgBB selama 7 hari. Hasil penelitian menunjukkan bahwa pemberian terapi ekstrak metanol biji mahoni dosis 100;  250  dan  400  mg/kgBB  pada  tikus  hasil  induksi  MLD-STZ  menunjukkan  penurunan  kadar  glukosa  darah  berturut-turut sebesar  55,47%;  81,01%  dan  73,63%,  peningkatan  kadar  insulin  sebesar  78,38%;  275,68%  dan  145,95%,  penurunan ekspresi  TNF-α  sebesar  30,34%;  67,28%  dan  49,91%,  serta  perbaikan  kerusakan  jaringan  pankreas  pada  penurunan derajat  insulitis  (p&lt;0,05).

Abstract

Shigella  flexneri is the most common causal agent of shigellosis. Its pili are composed of pili protein subunits. Adhesion molecules can be found on the pili and outer membrane proteins (Omp). A hemagglutination reaction can be used for screening of adhesion molecules. Objectives: The purpose of this study was to determine the molecular weight of the pili protein subunits and outer membrane proteins of S. flexneri that act as hemagglutinin proteins, and to prove whether there is a cross-reaction between antibodies against hemagglutinin pili protein subunits and outer membrane proteins of S. flexneri. Methods: Pili protein subunits were isolated using pili bacteria cutters, and the outer membrane proteins were solubilized and obtained using sodium dodecyl sulfate 0.05% as detergent for Omp isolation.  The hemagglutination reaction used mice erythrocytes. The cross reactions between subunit pili proteins were conducted by Western blot and Dot blot. Results:. Antibodies against hemagglutinin sub unit pili protein 18 kDa responded to pili protein subunits 18 kDa; 23 kDa; 34 kDa; and 53 kDa; and Omp 23 kDa and 27 kDa. Omp and subunit pili proteins S. flexneri consists of several identical epitopes  that were responsible for the similarity of the response profile in the cross-reactions of antibodies.
