Abstract

Hepatitis B virus (HBV) can be classified into 8 genotypes, genotype A to H. Genotype of HBV is important for clinical and etiological investigations. Research for HBV genotyping, HBV transmission study using nested PCR and HBV genotyping based on RFLP using restriction enzymes have been reported. However, both of those methods have been not applied for HBV genotyping study among hepatitis B patients in endemic area, like Indonesia yet. Molecular genotyping of HBV will describe epidemiology, pathogenesis and clinical implication of HBV. Combination of nested PCR and RFLP (nested PCR-RFLP) method to determine HBV genotype in Indonesia is still less information. The objectives of study were to develop a system for HBV genotyping by nested PCR combined with RFLP (nested PCR-RFLP) method based on nucleotide sequence of surface protein encoding</div><div>gene (S gene) in HBV genome and to confirm HBV genotypes which predominantly found among hepatitis B patients in Daerah Istimewa Yogyakarta Province and surrounding area. Total of 149 sera from chronic hepatitis B patients from Daerah Istimewa Yogyakarta and surrounding areas were collected for in this work. Viral DNA were extracted from sera of hepatitis B patients and used as template for first round nested PCR amplification using outer primers set. Amplicons of first round PCR were used as template for second round amplification using inner primers set. Then, amplicons of second round nested PCR were restriction digested by Sty I and Bsr I enzymes. For HBV genotyping then the restriction products were analyzed by RFLP based on restriction pattern. Results showed that the first round nested PCR amplification generated DNA fragments of whole S gene in length 1.233 bp, and in second round nested PCR amplification using inner primer set generated DNA fragments 585 bp in length. Genotype analysis for all samples using nested PCR-RFLP methods by restriction digested of Sty I and Bsr I enzymes found only 2 HBV genotypes among hepatitis B patients, namely genotype B and C. Quantification</div><div>data showed that most of hepatitis B patients found infected by HBV genotype B (92,8%), genotype C (3,6%) and unidentified genotype (3,6%). Nested PCR-RFLP methods for HBV genotyping is simple and inexpensive for clinical diagnostic in large scale.

Abstract

Hypertriglyceridemia can be caused by primary (genetic) or secondary etiology. One of the primary causes is hyperlipoproteinemia type I or known as familial chylomicronemia syndrome. Familial chylomicronemia syndrome is a rare autosomal recessive disease that occurs in 1-2 per 1,000,000 people, with specific characteristic signs, namely severe increment of fasting plasma triglyceride up to 100 times the normal value (about 1500-15,000 mg/dl) caused by lipoprotein lipase (LPL) mutation.

Abstract

Abstrak: Bojonegoro memiliki berbagai macam kebudayaan yang menjadi kebanggaan untuk daerah penghasil migas yang tergolong besar. Pada masa sekarang kebudayaan dan kesenian mengalami penurunan peminat dan menyebabkan kepunahan seperti halnya pada Tari Thengul, Tari Thengul dihidupkan kembali dengan menyesuaikan perkembangan zaman. Terciptanya Tari Thengul berawal dari keberadaan Wayang Thengul yang hampir punah, sehingga Joko Santoso dan Ibnu Sutowo seniman di Bojonegoro memiliki insiatif untuk tetap menjaga kesenian Wayang tersebut dengan cara mengangkat kembali unsur-unsur yang ada pada Wayang Thengul menjadi sebuah tarian. Tari Thengul juga dijadikan sebagai sarana belajar untuk kegiatan ekstrakurikuler Tari di sekolah hal ini dilakukan sebagai bentuk pengenalan kearifan budaya lokal dan nilai-nilai budaya local. Selain untuk kegiatan ekstrakurikuler Tari Thengul juga menjadi bagian dari pementasan yang diselenggarakan pemerintah Kabupaten Bojonegoro dan dijadikan ikon Bojonegoro sebagai tarian penyambutan tamu di Kabupaten Bojonegoro. Meskipun Tari Thengul dihadapkan dengan perkembangan globalisasi, Tari Thengul tetap menjadi bagian dari budaya dan menjadi ikon Bojonegoro dan Jawa Timur pada umumnya. Hal ini menunjukan bahwa tarian ini masih eksis dan sangat dihargai dengan baik di tingkat daerah ataupun di tingkat nasional. Tarian ini memiliki nilai-nilai budaya dan kearifan local melalui unsur gerakan dan melalui kostum yang khas, tarian ini menggambarkan keunikan budaya setempat dan sikap menghormati warisan leluhur. Kata kunci: Eksistensi, Tari Thengul, Seniman   Abstract: Bojonegoro has a variety of cultures that are a source of pride for the region, especially in the field of traditional arts and culture. However, in recent times, there has been a decline in interest in cultural and artistic activities, leading to the extinction of certain cultural practices like the Thengul Dance. To revive the Thengul Dance, adjustments have been made to adapt to the modern era. The creation of the Thengul Dance originated from the endangered Wayang Thengul tradition. Artists in Bojonegoro, such as Joko Santoso and Ibnu Sutowo, took the initiative to preserve the Wayang art form by transforming its elements into a dance performance. The Thengul Dance is not only used as a learning tool for extracurricular dance activities in schools but also serves as a means to introduce local cultural wisdom and values. Apart from being part of extracurricular activities, the Thengul Dance is featured in government-organized performances in Bojonegoro and is considered an iconic welcoming dance for visitors to the region. Despite the challenges posed by globalization, the Thengul Dance remains an integral part of the local culture and a symbol of Bojonegoro and East Java as a whole. This demonstrates the enduring significance and appreciation of this dance at both the regional and national levels. The dance embodies cultural values and local wisdom through its unique movements and distinctive costumes, portraying the local cultural heritage and a deep respect for ancestral traditions. Abstract

Background: Abdominal tuberculosis (TB) is a form of extrapulmonary TB that can present with or without involvement of the lungs. The diagnosis is difficult to establish, which may lead to diagnostic delays. Effective management of adolescent TB requires a holistic approach from various medical disciplines and interventions. This case presented a rare case 13-year-old girl diagnosed with abdominal TB Case: A 13-year-old girl presented with seven-months history of subfebrile fever, lymph node enlargement, abdominal distention, pallor, and severe weight loss. She was diagnosed with abdominal TB. The diagnosis was further complicated by portal hypertension, pulmonary TB, and severe acute malnutrition. To address these challenges, a multidisciplinary treatment plan was implemented and closely monitored for a period of 12 months Discussion: Multiple factors are significantly contributing to the successful outcome of the treatment for abdominal tuberculosis, including good adherence to the prescribed anti-tuberculous medications, absence of side effects from the drugs, the patient's positive knowledge, attitude and health behaviours, and housing and environmental health. Conclusion: This case highlights the importance of factors influencing disease outcomes of abdominal TB. Proper management of the factors would lead to significant clinical and nutritional status improvement, reduce TB transmission, and improved the overall quality of life.
