Abstract

This research purposes to identify the pathogen causing leaf spot in cempaka seedling and its control technique. Identify cause of disease is necessary to know precise technique control. Identification of disease was conducted in macroscopic appearances and microscopically to determine pathogens. Identification is done by using the determination key of fungi. The results showed that the early signs of leaf spot disease on cempaka seedlings in the nursery are streaks or spots on the leaf surface with distinct boundaries. Leaf spots are formed generally brown surrounded by darker boundaries. The result shows leaf spot disease on cempaka seedling was caused by fungal pathogen Colletotrichum sp. Techniques for controlling the disease can be done by isolating the infected seeds, reducing the intensity of canopy, and using the appropriate fungicides.

Abstract

Tangale Nature reserve is a conservation area located in Gorontalo that have the biodiversity typical of Wallacea bioregion including macroscopic fungi. The purpose of this study was to identify the fungal species diversity found in the Tangale Nature reserve as well as its potential use, considering that the information on species diversity of fungi in the Wallacea region is very limited. This research was conducted using the cruise method by identifying macroscopic fungi along the hiking trail of Tangale Nature Reserve. The macroscopic fungi were observed for its morphological characteristic include the colour, diameter, surface of the veil, the shape of the stem, the length and diameter of the stem, the lamella, including ring and pore, type of lamella and the type of volva. The research recorded the substrates and the location where the fungus was found. Data were analyzed descriptively qualitative. The results of the research identified 28 species of macroscopic fungi that included to the division Ascomycota and Basidiomycota. It belongs to 16 families and 8 orders  are Pezizales, Agaricales, Auriculariales, Boletales, Cantharellales, Gomphales, Polyporales, dan Russulales. Based on place to grow as much as 57,14% were found growing on decaying wood and 42,86% found growing in the soil/litter. Based on the potential of use, it has identified four species of mushrooms potentially as edible mushroom are Pleurotus ostreatus,  Auricularia auricula, Ramaria formosa, and Polyporus arcularius. 11 species of mushrooms potentially as medicinal mushroom are Calvatia craniiformis, Scleroderma citrinum, Lenzites betulina, Microporus flabelliformis, Coriolus versicolor, Microporus xanthopus, and Albatrellus confluens.

Abstract

Karet (Hevea brasiliensis (Willd. ex A.Juss.) Müll.Arg.) adalah tanaman perkebunan yang masuk dalam famili Euphorbiaceae dan berasal dari Brazil. Karet merupakan salah satu komoditas penting bagi sektor perkebunan di Indonesia. Sebagai negara yang memiliki luas lahan karet terluas di dunia, produktivitas perkebunan karet di Indonesia masih rendah apabila dibandingkan dengan Thailand. Penyakit gugur daun merupakan salah satu masalah dalam budidaya tanaman karet di Indonesia karena mengakibatkan kerugian hasil yang tinggi. Patogen penyebab penyakit gugur daun juga mengganggu produksi perkebunan karet swasta di Kabupaten Kutai Timur. Penelitian ini bertujuan untuk melakukan eksplorasi bakteri agens pengendali hayati dan menguji daya antagonisnya terhadap jamur patogen penyebab penyakit gugur daun. Pengambilan sampel dilakukan di Kabupaten Kutai Timur, Provinsi Kalimantan Timur sementara percobaan laboratorium dilaksanakan di Gedung Genomik dan Gedung InaCC – BRIN, Cibinong pada bulan September 2023 hingga Juni 2024. Hasil uji daya antagonis memperoleh tiga isolat bakteri— F1.4, E1.3, dan E5.2—yang berpotensi sebagai agens pengendali hayati. Tiga isolat bakteri tersebut merupakan bakteri Gram negatif. Isolat bakteri F1.4 dan E1.3 mampu menghasilkan enzim kitinase secara kualitatif. Hasil uji hemolisis dan hipersensitivitas menunjukkan gejala negatif untuk ketiga isolat bakteri tersebut. Penelitan ini diharapkan mampu memberikan gambaran mengenai potensi agens pengendali hayati dalam mendukung pertanian berkelanjutan untuk tanaman karet.
