Abstract

Daun salam (Syzygium polyanthum) adalah salah satu tanaman di Indonesia yang dipergunakan sebagai tanaman obat alternatif seperti obat antidiare, antiinflamasi, dan antidiabetes. Penelitian ini bertujuan untuk mengetahui toksisitas akut ekstrak etanol daun salam (EEDS) terhadap mencit galur DDY yang dilihat dari kematian mencit, perubahan perilaku, perubahan berat badan, dan nilai indeks organ. Penelitian ini dilakukan dengan metode eksperimental menggunakan 24 mencit putih jantan dan 24 mencit betina (Mus musculus) galur DDY yang dibagi menjadi 4 kelompok di setiap jenis dengan variasi dosis 1250 mg/kgBB, 2500 mg/kgBB, 5000 mg/kgBB, dan kelompok kontrol negatif. Pengamatan yang dilakukan pada penelitian ini yaitu kematian mencit, perubahan perilaku, perubahan berat badan, dan nilai indeks organ. Indeks organ yang dinilai adalah jantung, paru-paru, hati, limpa, ginjal, testis, vesica seminalis pada mencit jantan, serta ditambah organ ovarium, uterus pada mencit betina. Data dianalisis secara statistik menggunakan uji one way ANOVA. Hasil penelitian menunjukkan bahwa daun salam tidak menyebabkan kematian mencit, perubahan perilaku, perubahan berat badan, dan perubahan indeks organ mencit. Berdasarkan hasil tersebut dapat disimpulkan bahwa daun salam termasuk kategori praktis tidak toksik menurut klasifikasi dari BPOM tahun 2014. DOI : 10.35990/mk.v4n4.p355-368

Abstract

Kopi Lampung (Coffea canephora var. Robusta) digunakan sebagai alternatif dalam mengobati penyakit karena memiliki efek sebagai antioksidan, antiinflamasi, dan antidiabetes hingga menurunkan risiko kanker. Uji keamanan terhadap obat herbal dilakukan untuk mengetahui dosis aman untuk dikonsumsi. Penelitian ini bertujuan untuk mengetahui efek toksisitas akut ekstrak etanol biji kopi Lampung (EBKL) terhadap tikus wistar berdasarkan jumlah kematian, perubahan berat badan, dan indeks organ relatif. Penelitian inimenggunakan metode eksperimental pada 25 ekor tikus Wistar jantan yang dibagi 5 kelompok, yaitu kelompok kontrol, dan kelompok perlakuan EBKL dosis 625 mg/KgBB, EBKL dosis 1250 mg/KgBB, EBKL dosis 2500 mg/KgBB, dan EBKL dosis 5000 mg/kgBB. Pengamatan penelitian berupa jumlah kematian, perubahan berat badan, dan indeks organ tikus. Hasil penelitian menunjukkan bahwa ekstrak etanol biji kopi Lampung menyebabkan kematian pada hewan coba, yaitu pada EBKL dosis 2500 mg/Kg BB dan 5000 mg/Kg BB dannilai LD50 pada dosis ≥2500 mg/kgBB (p<0.05). Dari hasil penelitian terdapat perubahan berat badan yang signfikan pada dosis 5000 mg/kgBB (p<0.05), dan perbedaan signifikan indeks organ relatif hati pada dosis 5000 mg/kgBB (p<0.05) serta indeks organ relatif jantung pada dosis 2500 mg/kgBB dan 5000 mg/kgBB (p<0.05). Ekstrak etanol biji kopi lampung memiliki manfaat untuk kesehatan karena kandungan antioksidannya. Namun demikian, keamanan untuk digunakan sebagai terapi tetaplah harus memperhatikan dosis efektif dan batas keamanannya karena terdapat perubahan indeks organ relatif pada hati dan jantung setelah pemberian EBKL. Pada penelitian ini dapat disimpulkan bahwa ekstrak etanolbiji kopi Lampung tergolong dalam kategori sedikit toksis. Kata kunci: antioksidan, kopi, tikus, uji toksisitas

Abstract

Inflammation is a natural defense response of the body when an infection occurs. However, excessive inflammatory reactions can be triggered by lipopolysaccharides (LPS), which are key immunogenic components of bacterial capsules or cell walls. These reactions can lead to harmful effects on the body. Black rice (Oryza sativa L. indica) is a natural source rich in phenols, flavonoids, and antioxidant anthocyanins, which act as immunomodulators. This study aimed to investigate the effect of black rice ethanol extract as an anti-inflammatory agent on the proliferation of polymorphonuclear cells (PMN) in the spleen of rats induced by lipopolysaccharides. The study involved 27 mice, which were given a 400 mg/kg BW dose of black rice ethanol extract for 14 days. Afterward, the rats were induced with 0.3 mg/kg BW LPS intraperitoneally 24 hours before euthanasia using CO2. The spleens were then collected to measure the size of the germinal center diameter. The results showed that the treatment group had a germinal center diameter of 1.311 ± 0.226 μm (p<0.05) when compared to the positive control group. The LPS administration in the control group resulted in the disorganization of the white pulp and a reduction in lymphocyte cells and PMN in the germinal center. In conclusion, black rice ethanol extract at a dose of 400 mg/kg BW has an effect on PMN proliferation, as indicated by changes in the morphology of the white pulp in the germinal center of the spleen in rats.
