Abstract

Background: Sodium hypochloride (NaOCl), which is the gold standard for irrigation solutions, has low bactericidal activity against Enterococcus faecalis, the main cause of root canal treatments failure. This resistance due to its proton pump, which can also be found in Escherichia coli. This research was conducted using a combination of NaOCl with lansoprazole, the latest generation proton pump inhibitor. Objective: To determine the antibacterial effect of the combination of NaOCl with various concentrations of lansoprazole against E. faecalis and E. coli. Method: Laboratory experimental research in vitro with the diffusion method which was carried out in the MiCORE laboratory, Faculty of Dentistry, Universitas Trisakti. The independent variables were 5.25% NaOCl combination of 4% and 8.5% lansoprazole, 8.5% lansoprazole, 5.25% NaOCl combination of omeprazole 8.5%, 5.25% NaOCl and aquadest. Results: There was no inhibition zone of E. faecalis and E. coli in 8.5% lansoprazole, 5.25% NaOCl combination of omeprazole 8.5%, and aquadest. Inhibition zones of E. faecalis and E. coli were formed in 5.25% NaOCl combination of 4% lansoprazole, 5.25% NaOCl combination of 8.5% lansoprazole, and 5.25% NaOCl. The mean value of the inhibition zone of E. faecalis were 14,4±2,10 mm, 10,89±0,68 mm, and 25,9±1,46 mm respectively. Meanwhile, the mean value of the inhibition zone of E. coli were 19,48±2,48 mm, 10,89±0,76 mm, dan 30,22±1,10 mm. Conclusion: The antibacterial effect of 5.25% NaOCl combination of 4% and 8.5% lansoprazole was less effective than the antibacterial effect of 5.25% NaOCl.

Abstract

Background: Periapical abscess caused by tooth necrosis which are not treated. Enterococcus faecalis and Staphylococcus aureus area found in periapical abscess. Cefadroxil is one of antibiotics that choices to treat this infection. Several studies proved that C. sinensis leaves extract contains metabolite compounds such as catechins that can inhibit bacterial growth. To determine the antibacterial effect of C. sinensis leaves extract and cefadroxil combination against E. faecalis dan S. aureus growth. Methods: Laboratory experimental research with diffusion method. The research design is post test only control group design. Treatment was carried out with C. sinensis leaves extract 125 mg/mL, combination of C. sinensis leaves extract 15,625 mg/mL, 31,25 mg/mL, 62,5 mg/mL and 125 mg/mL with cefadroxil 8 μg/mL , cefadroxil 16 μg/mL as a positive control, and NaOH 0.1 N as a negative control. Results: There was none of inhibition zone in C. sinensis leaves extract 125 mg/mL and NaOH 0.1 N. The combination of C. sinensis leaves extract 15,625 mg/mL, 31,25 mg/mL, 62,5 mg/mL and 125 mg/mL and cefadroxil 8 µg/mL had an antibacterial effect. Based on Post-hoc Tukey’s HSD test, there was no significant difference between the combination and positive control. Conclusion: C. sinensis leaves extract and cefadroxil has potentially antibacterial effect on E. faecalis and S. aureus.

Abstract

Background: Cefadroxil, betalactam antibiotic belonging to cephalosporin class, is a drug of choice for treating periodontal infection. Previous studies reported that addition of green tea (Camellia sinensis) extract to betalactam antibiotics has shown synergistic antibacterial effect. Objective: To examine the antibacterial effect of GTE combined with cefadroxil on A. actinomycetemcomitans and P. gingivalis growth. Methods: In vitro experimental with post-test-only control design, conducted at MiCORE Laboratory, Trisakti University Faculty of Dentistry. There were 8 treatments: combination of cefadroxil 8 µg/mL with GTE 3,125 mg/mL, 6,25 mg/mL, and 12,5 mg/mL, GTE 3,125 mg/mL, 6,25 mg/mL, and 12,5 mg/mL, cefadroxil 16 µg/mL, and NaOH 0,1 N. Results: In treatment of solely GTE and NaOH 0,1, no inhibition zones were found. Meanwhile, combination of cefadroxil 8 µg/mL with GTE 3,125 mg/mL, 6,25 mg/mL, and 12,5 mg/mL, and cefadroxil 16 µg/mL inhibited both bacteria’s growth. Treatment of cefadroxil 8 µg/mL combined with GTE 3,125 mg/mL resulted in the largest inhibition zone. One-way ANOVA test showed significant difference, then post hoc test showed that combination of cefadroxil 8 µg/mL with GTE 3,125 mg/mL and 6,25 mg/mL were not significantly different from the positive control. Conclusion: Potentiation effect occurred on combination of cefadroxil 8 µg/mL with GTE 3,125 mg/mL and 6,25 mg/mL against A. actinomycetemcomitans and P. gingivalis growth. Green tea extract could increase cefadroxil antibacterial effect up to twice larger.
