Abstract

Alalak River is a river that has high industrial activity, so that the lead metal content due to industry in this Alalak river is very high, which is 0.405 mg/Kg in papuyu fish that live in the river. There needs to be an effort in reducing metals, one of which is with compounds that have adsorption properties such as gelam wood plants. Gelam wood is a plant that is widely found, especially in the South Kalimantan area, this wood has a cellulose value of 76.8% and the ability to absorb iodine of 791.59 mg/g. The purpose of this study was to test the characteristics of the ability of gelam wood activated carbon in reducing lead (pb) levels in papuyu fish in the Barito river and see the effect of time variance in reducing lead (pb) levels. This research method is a true experiment to determine the effect of lead (pb) reduction activity in different exposure times. This study used a control group and a test group with exposure in several variations of contact time with gelam wood activated carbon. The results showed that the characteristics of gelam wood activated carbon were in accordance with the provisions of the Indonesian National Standard (SNI). While the significance value of the decrease in lead (pb) levels with exposure to activated carbon in gelam wood is <0.05 so it can be concluded that there is a significant effect on the decrease in pb levels.

Abstract

Background: Diarrhea remains a major global health problem requiring effective and safe therapeutic interventions. The utilization of natural activated carbon presents a promising alternative for antidiarrheal therapy. Objective: This study aimed to develop an activated carbon tablet formulation derived from Kayu Gelam (Melaleuca sp.) and to evaluate the physical and chemical quality of the resulting tablets. Methods: Activated carbon was obtained through carbonization and chemical activation processes, then formulated into three tablet formulas with varying concentrations of polyvinylpyrrolidone (PVP) as a binder: 2% (Formula 1), 3% (Formula 2), and 4% (Formula 3). Tablet quality evaluation included pH measurement, weight uniformity, hardness, friability, and disintegration time. Data were analyzed using one-way ANOVA. Results: The pH values of Formulas 1, 2, and 3 were 6.15, 6.65, and 6.85, respectively, all within the neutral range (5.8–7.4) and safe for biological systems. The average tablet weights were 157.0 mg, 153.5 mg, and 158.5 mg, respectively, meeting weight uniformity requirements. Hardness values were 6.47 kg, 6.83 kg, and 7.67 kg, indicating adequate mechanical strength. Friability values were 0.664%, 0.763%, and 0.502%, all below the acceptable limit of 1%. Disintegration times were 13.01 min, 13.65 min, and 13.85 min, respectively, satisfying the requirement of less than 15 minutes. One-way ANOVA revealed significant differences (p < 0.05) in pH, hardness, friability, and disintegration time among the three formulas, while weight uniformity showed no significant difference (p > 0.05). Conclusion: All formulated Kayu Gelam activated carbon tablets met the required physical and chemical quality parameters, demonstrating their potential for further development as an alternative antidiarrheal tablet preparation.

Abstract

Background: Acne vulgaris is a chronic inflammatory skin condition where prolonged use of synthetic antibacterials can lead to bacterial resistance, necessitating natural alternatives. Centella asiatica extract, rich in triterpenoid saponins and flavonoids, exhibits antibacterial activity against acne-causing bacteria. Objective: This study aimed to formulate and evaluate the physical properties and in vitro penetration of an acne patch containing C. asiatica extract using a combination of polyvinylpyrrolidone (PVP) K30 and ethyl cellulose polymers. Methods: The extract was obtained by 96% ethanol maceration and formulated into three patch formulas with varying polymer ratios (PVP K30:ethyl cellulose = 100:300, 200:200, and 300:100 w/w). All patches were prepared using the solvent casting method. Evaluations included organoleptic properties, homogeneity, thickness, folding endurance, moisture content, weight uniformity, and in vitro penetration using a Franz diffusion cell. Results: All formulations produced homogeneous, transparent patches with uniform thickness (approx. 1 mm). However, all formulas exhibited poor mechanical properties, with folding endurance values (<30 folds) far below the required standard (>300 folds). Moisture content (21.7-23.3%) exceeded the recommended limit (<10%), indicating potential instability. Furthermore, the cumulative in vitro penetration of the active compound through rat skin was very low (0.001-0.004 µg/cm²) over 180 minutes. Conclusion: The C. asiatica extract acne patches demonstrated acceptable basic physical characteristics (homogeneity, thickness). However, they require significant optimization to improve mechanical strength, reduce moisture content, and enhance drug penetration before being considered a viable anti-acne preparation.

Abstract

Background: Bangang tree latex (Tabernaemontana macrocarpa Jack.) is a natural material empirically used by the community as traditional medicine. However, scientific data on its secondary metabolite content, particularly alkaloids, remain limited. The genus Tabernaemontana contains alkaloids with potential pharmacological activity, warranting quantitative analysis of total alkaloids in Bangang tree latex. Objective: This study aimed to identify alkaloid compounds in Bangang tree latex through phytochemical screening and determine the total alkaloid content using UV-Vis spectrophotometry. Methods: This research was a quantitative descriptive study. Fresh latex was dried, reduced in size, and extracted by maceration using 96% ethanol. Alkaloid identification was performed using Dragendorff, Mayer, and Wagner reagents. Total alkaloid content was determined using caffeine standard, BCG reagent, phosphate buffer pH 4.7, and chloroform at a maximum wavelength of 407 nm with an Operating time of 41 minutes. Results: The extract yield was 4.08%. Qualitative tests showed positive alkaloid results with all three reagents. The caffeine calibration curve produced the regression equation y = 0.003x + 0.0131 with R² = 0.9961 and r = 0.9980. The average sample absorbance of 0.036 produced a concentration of 7.633 ppm and a total alkaloid content of 76.33 mg CAE/g extract. Method validation showed %RSD of 1.74%, recovery of 99.05%, LOD of 0.730 ppm, and LOQ of 2.200 ppm. Conclusion: Bangang tree latex was positive for alkaloids, and its total alkaloid content could be determined using UV-Vis spectrophotometry.
