Abstract

ELISA merupakan teknik uji berbasis plat yang diciptakan untuk mendeteksi dan mengukur hormon, peptida, protein, dan antibodi maupun antigen. Pada metode ELISA, antigen atau antibodi di imobilisasikan ke permukaan padat dan kemudian di komplekskan dengan antibodi atau antigen yang terkait dengan enzim. Deteksi dilakukan dengan mengukur aktifitas enzim yang terkonjugasi melalui inkubasi dengan substrat yang menghasilkan produk yang dapat diukur. Tujuan dari melakukan pencucian adalah untuk menghilangkan sisa antigen-antibodi, substrat dan bahan lainnya yang tidak berikatan pada reaksi enzimatik dalam pengujiannya. Teknik pencucian dalam pemeriksaan metode ELISA dapat dilakukan dengan cara manual dan automatik. Jenis penelitian ini adalah observasional analitik dengan melakukan penelitian terhadap 6 sampel yang di duplo. Hasil yang diperoleh yaitu 24 hasil kadar hCG dengan 12 hasil pengukuran teknik pencucian manual dan 12 hasil pengukuran teknik pencucian automatik dengan rata-rata kadar hCG teknik pencucian manual yaitu 357.8135 IU/L dan rata-rata kadar hCG automatik yaitu 308.432 IU/L. Hasil uji statistik independent T-test data diperoleh p value = 0,469 (>0,05) yang menunjukkan tidak terdapat perbedaan yang dignifikan antara pemeriksaan metode ELISA teknik pencucian manual dengan teknik pencucian automatik. Kesimpulan dari penelitian ini yaitu adanya perbedaan kadar hCG dengan metode ELISA teknik pencucian manual lebih tinggi dari pada teknik pencucian automatik namun tidak berbeda signifikan.

Abstract

Quail eggs are a source of protein that many people consume. Eggs have a short shelf life. The longer eggs are stored, the quality and freshness of eggs decreases. In order to extend the shelf life of eggs, they are treated with a lime solution. The pupose of this study was to determine the protein profil of quail eggs by immersing in lime solution on variations in storagetime based on SDS-PAGE. The results showed that the highest bands were in samples T3 (B), T4 (A and B), T1 (A and B), T2 (A and B) respectively, namely 3 major bands and 6 minor bands, 9 minor bands, 1 major band and 8 minor bands, 3 major bands and 5 minor bands, 2 major bands and 6 minor bands, 3 major bands and 5 minor bands, 1 major band and 7 minor bands. While the lowest band results are in sample T3 (A), namely 1 major band and 6 minor bands. Based on these results, it can be concluded that there are differences in the protein profile of quail eggs between the control sample and the treated sample.

Abstract

Red snapper (Lutjanus bitaeniatus) is a source of animal protein with a high-water content, making it prone to spoilage. Spoilage can be prevented through preservation. Garlic can be used as a natural preservative for fish because it contains antibacterial substances, including allicin, which plays a role in inhibiting and killing spoilage bacteria, as well as other antimicrobial compounds such as alkaloids, flavonoids, saponins, and tannins. This study aims to determine the effect of garlic extract as a natural preservative on the protein profile of red snapper. The research design was divided into five parts: fresh red snapper stored for 24 hours without soaking, and three parts soaked in garlic extract at concentrations of 5%, 10%, and 20%, respectively. This type of research was experimental. The research method was the preparation of garlic extract at concentrations of 5%, 10%, and 20%. Protein concentration was calculated using the Bradford method, followed by SDS-PAGE to separate proteins based on their molecular weight. The SDS-PAGE results were analyzed using the GelAnalyzer 19.1 application to calculate the molecular weight of the proteins and the protein profiles of the treatments compared to fresh fish. The results showed that the highest protein concentration was found in the 5% garlic extract treatment compared to the 10% and 15% concentrations. The protein profiles of fresh snapper and the 5% garlic extract treatment were not significantly different from fresh red snapper, with 13 protein bands (9 major bands and four minor bands). In contrast, fresh red snapper had 16 protein bands (13 major and three minor bands). This study concludes that garlic extract immersion treatment can be used as a natural preservative, with the optimal concentration being 5% garlic extract.
