Abstract

This study was motivated by the lack of mathematical problem solving ability of students, especially on the material linear equations. The causes of low due to lack of student interest in mathematics and students are less active in the classroom and the learning method are still monotonous (teacher centered). The purpose of this study was to determine the effect of discovery learning method on students mathematical problem solving ability. The study was a quasi-experiment. The population in this study were all students of class X SMA N 5 Batam. The sample in this research were class XA as an experimental class using the discovery learning method and class XB as a control using conventional learning method. Instruments in this study was an essay test. The instruments analysis used validity, reliability, level of difficulty of questions, and different power problems. Data were analyzed using one sample t-test.. Based on the research results, the level of mathematical problem solving ability of students classified as good with an average of 77.04. With μ = 75 then obtained t = 12.686, so it can be concluded that the discovery learning method was effective against the ability of students mathematical problem solving and discovery learning method was better than the conventional method of the mathematical problem solving ability of students grade X SMAN 5 Batam.Key words: discovery learning method and problem solving ability

Abstract

This research aims to improve safe and legal food management in small industries by providing assistance through training and the provision of simple tools. The method used is a qualitative approach with case studies, involving in-depth interviews, observations, and document analysis regarding food safety regulations. Data is collected from small businesses in the food sector to identify the challenges they face in implementing food safety standards. The results of the study show that many business actors do not fully understand the applicable regulations, and access to adequate training and resources is very limited. However, after the implementation of the assistance program, there has been a significant improvement in food safety understanding and practices, as well as a reduction in errors in data recording. The recommendations of this study include the need for government support in providing affordable training and access to resources, as well as the development of online platforms to facilitate business actors in food safety management. With these measures, it is hoped that small industries can adapt to changes in the market and regulations, thereby improving product quality and competitiveness in the local market

Abstract

The spike protein of SARS-CoV-2 is crucial for initiating infections by binding to host cells and mediating membrane fusion. In this study, HEK-293T cells were transfected with plasmids encoding three structural proteins of SARS-CoV-2, i.e., Spike (S), Membrane (M), and Envelope (E). This transfection enabled the formation of SARS-CoV-2 Virus-Like Particles (VLPs), which allows for safer studies of the virus and its proteins. To serve as a marker for expression, an enhanced green fluorescent protein (EGFP) was fused to the spike protein, resulting in a Spike-EGFP (S-EGFP) fusion protein. The characteristics of SARS-CoV-2 spike protein expression in the transfected HEK-293T cells were then investigated using polymerase chain reaction (PCR) and flow cytometry. The PCR analysis revealed non-specific DNA band smearing, which did not provide conclusive confirmation of spike protein expression. However, flow cytometry analysis demonstrated that approximately 30% of the transfected cells exhibited green fluorescence, indicating the expression of the Spike-EGFP fusion protein. These findings, obtained through flow cytometry, confirmed the successful spike protein expression in transfected HEK-293T cells.
