Abstract

Plant regeneration through invitro culture is strongly influenced by the use of growth regulators. BAP is a class of cytokinins that affect cell division which is very well used to stimulate plant growth. The aim of the study was to determine the effect of BAP concentration on the regeneration of various in vitro mutant taro. The study was carried out at the Plant Tissue Culture Laboratory,  Faculty of Agriculture, Hasanuddin University, Makassar. The method used was factorial completely randomized design with 2 factors as the environmental design. The first factor was the BAP concentrations of 0.0 mg/L, 1.0 mg/L, 1.5 mg/L, 2.0 mg/L and 2.5 mg/L. The second factor was mutant plants consisting of control plants (diploid), mutant 1 (Putative-Tetraploid), mutant 2 (Mixoploid), mutant 3 (Mixoploid), and mutant 4 (Putative-Tetraploid). The results showed that the best regeneration was obtained by BAP concentration of 0 mg/L with control plants on the parameters of root length (2.41 cm), number of roots (11.66 pieces) and plantlet formation rate (13 days). The BAP concentration of 1 mg/L was shown by mutant plant 1 on the parameters of the number of shoots (26 pieces) and number of leaves (16 pieces). BAP concentration of 0 mg/L in mutant 4 had a shoot height of 1.83 cm. The results of ploidy flow cytometry analysis of mutant plants with Putative-Tetraploid parental traits after regeneration resulted in detectable diploid chromosomes that were different from the parent types in all BAP concentration treatments

Abstract

This study aims to determine how the interaction occurs between the best concentration of EMS with gibberellin in inducing aglaonema variegata and regeneration of aglaonema Red Majesty in vitro. The research was conducted at the Tissue Culture Laboratory of the Faculty of Agriculture, Hasanuddin University and took place from January to June 2022. The study used the method of Separate Plots Design in a Completely Randomized Design as the environmental design. The results showed that the best mutant genetic diversity and in vitro regeneration at 0.3 percent EMS concentration with 15 ppm gibberellin (variegata), and 45 ppm (chimera) and 0.6 percent EMS concentration with 0 ppm gibberellin (variegata) had different characters compared to the control treatment and other treatments

Abstract

widuri adalah tanaman hias dengan daya tarik utama terletak pada keindahan daunnya dan termasuk tanaman hias yang secara komersial sangat prospektif dikembangkan. Akan tetapi, tanaman aglaonema ini memiliki kendala terkait pertumbuhan yang lambat. Oleh sebab itu, dilakukan perbanyakan melalui bioteknologi seperti kultur jaringan yang dipadukan dengan teknik mutasi agar mendapatkan tanaman yang tumbuh cepat, seragam dan bernilai tinggi. Untuk itu dikombinasikan bahan EMS karena terbukti dapat menyebabkan terjadinya mutasi titik dan bahan GA3 yang dapat meregenerasi tunas secara in vitro sehingga diperoleh kombinasi dengan konsentrasi terbaik dalam menginduksi variegata dan mengetahui heritabilitas keragaman dan pertumbuhan tanaman variegata aglaonema widuri. Metode yang digunakan adalah Rancangan Acak Lengkap 2 Faktorial dengan faktor pertama yaitu EMS dengan konsentrasi (0%, 0.3%, 0.6%, 0,9) dan Faktor kedua yaitu GA3 dengan konsentrasi (0 ppm, 15 ppm, 30 ppm, 45 ppm) sehingga di peroleh hasil bahwa perlakuan yang memberikan data kecepatan tumbuh tunas, panjang tunas, banyak tunas yang paling baik adalah perlakuan E2G2 meliputi perlakuan EMS (E2) 0,6 % dan Konsentrasi GA3 (G2) 25 ppm. Selain itu, pada tahap analisis regresi semua variabel memiliki pengaruh terhadap variabel lain yang diamati, dan hasil analisis korelasi mengarah ke positif. Kemudian untuk hasil analisis nilai heritabilitas setiap variabel memiliki nilai heritabilitas yang tinggi, hal ini menunjukkan bahwa perbedaan pertumbuhan dan arah variegata dari setiap karakter, lebih banyak dipengaruhi oleh faktor genetik dibandingkan dengan faktor lingkungan. Olehnya, terdapat interaksi yang nyata berbagai konsentrasi perlakuan terhadap parameter yang di amati.
