Abstract

Spektrofotometer UV-Vis Fluostar Omega Tipe Mikroplate di laboratorium Kimia FMIPA UNNES telah dipakai untuk praktikum, penelitian dan layanan pengujian sampel. Untuk menjamin hasil pengukuran yang valid, kalibrasi spektrofotometer tersebut perlu dilakukan secara berkala. Pada penelitian ini, kalibrasi dan verifikasi kinerja instrumen spektrofotometer UV- Vis Fluostar Omega Tipe Mikroplate dilakukan untuk penjaminan mutu pengujian kadar sampel. Sampel yang dianalisis adalah vitamin C dengan metode kurva kalibrasi. Kalibrasi yang dilakukan meliputi kalibrasi fotometri, dan kalibrasi panjang gelombang. Selanjutnya, validasi metode dan pengujian kadar vitamin C dalam sampel juga dilakukan. Akurasi fotometri pada daerah ultraviolet dan visibel serta akurasi panjang gelombang menunjukkan Persentase RSD < 2%. Nilai ini membuktikan bahwa pembacaan absorbansi dan panjang gelombang oleh alat sangat teliti. Uji lineritas standar vitamin C didapatkan persamaan linear y = 0,0164x + 0,1128 dengan nilai koefesien korelasi(R2) sebesar 0.9982. Validasi metode pengujian vitamin C didapatkan rata-rata persen galat, persen recovery, dan persen akurasi masing-masing sebesar 5,6784%, 94,3216%, dan 94,3216%, dengan kategori sangat teliti. Pengujian kadar sampel tablet vitamin C 500 mg didapatkan hasil yang sangat teliti dengan persen galat, persen recovery, dan persen akurasi masing-masing sebesar 8,2565%, 91,7435%, dan 91,7435%.

Abstract

Purple sweet potato is one of the potential sources of anthocyanins. Anthocyanins can be utilized in various fields, such as sensors. Chitosan biopolymer allows formulation with anthocyanins because it possesses biodegradable and antimicrobial properties, and it does not inhibit color changes from anthocyanins. This study aims to prepare anthocyanins from purple sweet potatoes, formulate chitosan-anthocyanin biopolymer (CAB), and test the effect of time and formalin concentration on color changes in CAB. The research results show: 1) anthocyanin extract (TAC) from purple sweet potatoes is 852 mg/L with anthocyanin monomer (cyanidin) of 443.03 mg/500 g or 88.61 mg/100g. Anthocyanin extract at pH 1-12 has colors ranging from red to brownish-yellow; 2) CAB was successfully formulated with anthocyanin extract from purple sweet potatoes, chitosan, and glycerin. BKA-A was obtained with a content of 10 mL anthocyanin extract and CAB-B with 15 mL anthocyanin extract; 3) the produced biopolymers have average swelling values of 444% (CAB-A) and 339.5% (CAB-B). The storage time of CAB-B is longer than CAB-A. FTIR analysis showed a broad peak at ~3336 cm-1 indicating hydroxyl groups from anthocyanin extract; and 4) ANOVA test results showed that interaction time and formalin concentration affect color changes in CAB-A partially, while color changes in CAB-B are only influenced by interaction time with formalin.

Abstract

This article has been retracted. Reason: This article has been retracted following a mutual agreement between the authors and the journal that the manuscript should not proceed to publication. The retraction is issued to ensure the integrity of the academic record Basic Health Research (Riskesdas) in 2018 showed an increase in the prevalence of hypertension in Indonesia. Captopril is one of the antihypertensive drugs that is often prescribed because it is affordable and effective in lowering blood pressure, but captopril also has several side effects. Therefore, the development of sensitive, simple, rapid and cost-effective analytical techniques for the determination of captopril is urgently needed. This research aims to develop a colorimetric detection of captopril using silver nanoparticles and analyze its performance in measuring the captopril in tablet samples. The parameters studied include linearity, sensitivity, accuracy and precision. The results of the research show that silver nanoparticles can be used for qualitative detection of captopril which is characterized by a color change and absorbance decrease of the sample solution. The results of the performance test of the developed method showed good results. The linearity test showed a correlation coefficient (r) of 0.9915 with the regression equation obtained was y = 0.263x + 129.26. The limit of detection (LOD) value is 0.0206 ppm and the limit of quantitation (LOQ) is 0.0687 ppm. In the accuracy test results, the recovery percentage (%recovery) was 95.3% and the precision test with the relative standard deviation (%RSD) obtained was 0.85%. The captopril content obtained in the 50 mg captopril tablet sample preparation was 98.0%.
