Abstract

Salah satu upaya peningkatan produktivitas padi sawah irigasi adalah dengan merakit Varietas Unggul Baru (VUB) yang memiliki daya hasil tinggi serta sesuai dengan kondisi agroekosistem, sehingga perlu adanya peningkatan stabilitas suatu galur agar tetap berproduksi tinggi. Tujuan penelitian ini untuk mengevaluasi keragaan agronomi hasil 31 galur padi (Oryza sativa L.) inbrida sawah irigasi dengan potensi hasil tinggi pada dataran rendah Sukamandi. Penelitian dilaksanakan pada bulan September 2021 – Januari 2022 (Musim Tanam (MT) 2-2021) di Kebun Percobaan Balai Besar Penelitian Tanaman Padi Sukamandi. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) dengan 39 perlakuan yang terdiri dari 31 galur dan 8 varietas pembanding yaitu Inpari 30, Inpari 32, Inpari 42, Cakrabuana, Inpari Digdaya, Inpari Gemah, Inpari 47, dan Inpari 48 yang diulang sebanyak 3 kali. Hasil penelitian menunjukkan bahwa terdapat galur-galur yang diuji memiliki perbedaan yang nyata pada seluruh karakter keragaan agronomi. Sebanyak delapan galur harapan nyata mempunyai hasil panen mendekati varietas pambanding terbaik Inpari 42 (5,5 t/ha) dengan kisaran 4,25-5,34 t/ha. Galur-galur tersebut yaitu BP 4126f-Kn-1-WBC-2-3-4, BP 30546D-SKI-19-3-2, B14667E-MR-18-5-Kn-1, B14928D-MR-27-2-5-3-PN-2, IR 102860-8:42-B-B, B1398E-KA-6-3, PR40786-16-2-0-SBY-0-CRB-0, dan BP18322-3-2-JK-0-IDN-2-SKI-6-PWK-2.

Abstract

The continual of the same high yield variety (HYV) is not recommended because it can cause resistance breakage against pest disease. One of the Indonesian Center for Rice Research (ICRR) main tasks is to establish new rice varieties with high yield potential and other better characteristics than the previous HYVs. Yield Test of promising lines aimed to select the yield potential, resistance to major pests, and rice quality to be further tested in a breeding processes. The research was carried out in the ICRR Experimental station, located in Sukamandijaya Village, Subang Regency, West Java. This study used a randomized complete block design with 4 replications consisting of 14 advanced generation lines, namely G1 (BP 30533D-SKI-4-3-1), G2 (BP 19210e-6-2-1- 3), G3 (BP 30531D-SKI-24-3), G4 (BP 30531D-SKI-1-2-3), G5 (BP 30534D-SKI-6-1), G6 (BP 30533D-SKI-4- 3-2), G7 (BP 30485E-SKI-9-5-2-0), G8 (BP 30533D-SKI-29-1-1-Ski), G9 (BP 31507D-SKI-1-1-1) , G10 (BP 30475c-SKI-6-4-1-5), G11 (Koshi CSSL [IR64] SL2129), G12 (BP 30533D-SKI-29-1-1-Skb), G13 (BP 31456D-SKI- 7-1), G14 (BP 31523D-SKI-2-1-0-0), and 2 checks (A (INPARI 33) and B (INPARI 32)). The results of the analysis of variance and the post hoc test used was DMRT test The data were analyzed by analysis of variance on yield did not show a significant difference between the lines tested and the best checks INPARI 33 ( 6.18 t/ha MPD). The G1 and G11 lines showed a moderately resistant response to bacterial leaf blight (HDB) IV, both lines were recommended for further testing

Abstract

Forage is a very important element for livestock development as they need it to survive, produce and reproduce. The forages that is often used is panic grass (Panicum maximum) which is a feed grass from the African plains, widely used in various regions of Indonesia. This grass is used for their high nutritional value, the ability to adapt various types of soil and weather, and usage as an ideal or high quality “cut and carry” grass for beef and dairy cattle. Forage cultivation by considering plant genetics can improve the quality of all types of forage inherited from the parent hence increase the productivity of feed crops and support livestock development. This can be done with molecular marker, which is a technology that can confirm target genes effectively, efficiently and accurately and are not influenced by environmental factors. Among the markers or molecular markers currently available, microsatellites or Simple Sequence Repeats (SSR) have become a marker system that is commonly used because of its various advantages, namely that it can be amplified by Polymerase Chain Reaction using primers that combine sequences in the target DNA. In this research, genes that carry leaf-rolling, plant height, and fertility-restoring properties that have previously been identified in rice plants will be tested on panic grass using RM20A, RM241, RM315. After being visualized using electrophoresis gel, it can be concluded that RM241 had the best result as it has clear amplification result and polymorphic bands with effectiveness and polymorphism levels of 75% and 100% respectively.
