Abstract

Penyakit busuk pelepah merupakan salah satu penyakit penting pada tanaman padi yang dilaporkan semakin meluas dan berdampak signifikan terhadap penurunan hasil. Selain Sarocladium oryzae, beberapa spesies Fusarium, termasuk Fusarium longipes, diketahui berasosiasi dengan penyakit ini dan berpotensi memperparah kerusakan tanaman. Penelitian ini bertujuan untuk mengevaluasi tingkat ketahanan beberapa varietas padi terhadap penyakit busuk pelepah akibat infeksi F. longipes berdasarkan keparahan penyakit dan parameter epidemiologi. Penelitian dilaksanakan di rumah kaca menggunakan sepuluh varietas padi, yaitu Ciherang, Sunggal, Inpari 32, Inpari 42, Inpari 43, Memberamo, Situ Bagendit, IR64, Padjadjaran, dan SRP08, yang disusun dalam rancangan acak kelompok dengan tiga ulangan. Inokulasi dilakukan pada umur 60 hari setelah tanam menggunakan bulir beras yang telah dikoloni F. longipes. Pengamatan keparahan penyakit dilakukan pada 3, 6, 9, 12, dan 15 hari setelah inokulasi. Hasil penelitian menunjukkan bahwa keparahan penyakit meningkat seiring waktu pengamatan dan berbeda antarvarietas. Pada akhir pengamatan, seluruh varietas menunjukkan keparahan penyakit yang tinggi dan diklasifikasikan sebagai sangat rentan berdasarkan kriteria IRRI. Nilai laju infeksi dan Area Under Disease Progress Curve (AUDPC) menunjukkan adanya perbedaan dinamika perkembangan penyakit antarvarietas, meskipun tidak ditemukan varietas dengan tingkat ketahanan efektif terhadap infeksi F. longipes. Hasil penelitian ini menegaskan pentingnya evaluasi ketahanan varietas sebagai dasar pengelolaan penyakit busuk pelepah dan pengembangan varietas padi yang lebih adaptif.

Abstract

Abstrak: Penggunaan pestisida kimia sintetis secara berlebihan di kalangan petani menimbulkan dampak negatif terhadap kesehatan dan lingkungan, sekaligus meningkatkan biaya produksi pertanian. Desa Ngargoloka, Kecamatan Gladagsari, Boyolali, Jawa Tengah, menghadapi permasalahan tersebut di tengah melimpahnya limbah tembakau yang belum dimanfaatkan secara optimal. Kegiatan pengabdian ini bertujuan untuk meningkatkan pengetahuan dan keterampilan petani dalam membuat pestisida nabati berbasis limbah tembakau sebagai alternatif pengendalian hama yang ramah lingkungan dan ekonomis. Metode yang digunakan meliputi tiga tahapan, yaitu edukasi dampak pestisida kimia, pelatihan pembuatan dan formulasi pestisida nabati, serta evaluasi menggunakan instrumen pre-test dan post-test. Kegiatan melibatkan 17 petani anggota kelompok tani aktif. Hasil evaluasi menunjukkan peningkatan pengetahuan rata-rata sebesar 67,06%, dari 31,76% menjadi 98,82%. Peningkatan tertinggi terjadi pada aspek pemahaman potensi limbah tembakau sebagai pestisida nabati dan keyakinan tentang kemampuannya mengurangi ketergantungan pada pestisida kimia, masing-masing sebesar 94,12%. Selain itu, seluruh peserta memberikan penilaian positif terhadap materi, narasumber, dan sarana pelatihan, serta 64,71% menyatakan berminat untuk mempraktikkan pembuatan pestisida nabati secara mandiri.Abstract: The excessive use of synthetic chemical pesticides among farmers has caused negative impacts on health and the environment, while also increasing agricultural production costs. Ngargoloka Village, Gladagsari District, Boyolali, Central Java, faces these challenges amid an abundance of tobacco waste that has not been optimally utilized. This community service activity aimed to improve farmers' knowledge and skills in producing plant-based pesticides from tobacco waste as an environmentally friendly and cost-effective alternative to pest control. The method consisted of three stages: education on the impacts of chemical pesticides, training in the production and formulation of botanical pesticides, and evaluation using pre-test and post-test instruments. A total of 17 active farmer group members participated. The evaluation results showed an average knowledge improvement of 67.06%, from 31.76% to 98.82%. The highest increase occurred in understanding the potential of tobacco waste as a botanical pesticide and in participants' confidence in its ability to reduce dependence on chemical pesticides, each rising by 94.12%. Furthermore, all participants gave positive assessments of the training materials, presenters, and facilities, with 64.71% expressing interest in independently practicing the production of botanical pesticides.

Abstract

Yellow oyster mushroom (Pleurotus citrinopileatus) is one of the edible mushroom commodities that has the potential to be developed as an alternative source of nutritious food. Optimization of the growing media is an important factor in supporting mycelial growth because the substrate must provide lignocellulosic compounds as the main carbon source. This study aimed to analyze the response of mycelial growth rate of yellow oyster mushroom to different baglog media formulations and to determine the best growing media for yellow oyster mushroom spawn development. The research was conducted from June to December 2025 using a randomized block design (RBD) with a single factor, namely media formulation consisting of seven treatments with three replications. The media treatments consisted of combinations of sengon sawdust, cocopeat, corn, and millet. The observed parameter was mycelial growth rate measured on days 7, 14, 21, and 30. Data were analyzed using ANOVA at a 5% significance level followed by Tukey’s test (HSD). The results showed that media formulation had a highly significant effect on mycelial growth rate. Treatment P1 (100% sengon sawdust) produced the highest mycelial growth rate of 0.526 cm/day and was significantly different from other treatments, while treatments P2–P7 produced growth rates ranging from 0.252–0.341 cm/day. Pure sengon sawdust media resulted in faster mycelial growth and earlier baglog colonization due to its high lignocellulose content and porous substrate structure, which supports aeration and optimal mycelial growth.

Abstract

This study aimed to evaluate the effects of growth medium formulation, storage temperature, and preservation period on the spore viability of the white oyster mushroom (Pleurotus ostreatus). The study employed an experimental method using a split-plot design involving three types of preservation media: Potato Dextrose Broth (PDB), PDB supplemented with 15% paraffin, and corn decoction medium. The treatments were evaluated under two storage temperature conditions, namely 4°C and room temperature at 26°C, across four preservation periods: 2, 4, 6, and 8 weeks. The observed parameters included the time of colony emergence, colony number, and microscopic hyphal morphology. The results showed that all media stored at 4°C were able to maintain spore viability for up to 8 weeks. PDB produced the highest colony numbers, ranging from 224 to 593 colonies, and showed the fastest colony emergence, occurring on days 1 to 2. PDB supplemented with 15% paraffin produced 128 to 248 colonies, with colony emergence on day 2, whereas the corn decoction medium produced 89 to 142 colonies, also with colony emergence on day 2. All treatments stored at room temperature at 26°C became contaminated and therefore failed to maintain spore viability. Microscopic observations indicated that hyphal structures remained normal in all treatments stored at 4°C. Based on colony number and growth rate, PDB stored at 4°C demonstrated better preservation performance than the other media in maintaining the spore viability of Pleurotus ostreatus for up to eight weeks of storage.

Abstract

This study was conducted to evaluate the diversity of insect communities associated with sorghum plants of the genotypes Watar Hammu Witting Walla and Watar Hammu Manippa Tadda. Sampling was carried out using a Z-shaped transect method on both genotypes. Each genotype was represented by five replications, with every replication consisting of five plots, resulting in a total of 25 plots per genotype or 50 plots in total. Within each plot, six Yellow Sticky Traps were placed at representative points following the Z transect pattern to ensure systematic and comprehensive sampling. The traps were installed in the morning at approximately 07:00 a.m. and left in the field until 05:00 p.m. on the same day. After collection, insects adhered to the traps were gathered and stored in clear plastic bags for subsequent identification in the laboratory. The results of the analysis showed that the diversity index of the WH Witting Walla genotype was 1.54, while that of the WH Manippa Tadda genotype was slightly higher at 1.62. The dominance index values were 0.29 and 0.28, respectively, both of which are categorized as moderate. Across both genotypes, 13 insect genera were identified, including Musca, Bactrocera, Phaenicia sericata, Condylostylus longicornis, Bemisia, Bathrogia, Nezara, Camponotus, Abispa, Coleomegilla, Coccinella, Pieris rapae, Spodoptera, and Oxya. Among these, Musca sp. (housefly) and Bemisia sp. (whitefly) were the most dominant species, followed by Spodoptera sp. and Oxya sp. These findings suggest that insect communities on both sorghum genotypes exhibit moderate diversity and balanced dominance, indicating a relatively stable ecosystem.

Abstract

Crocidolomia pavonana is a major pest that damages Brassicaceae plants such as cabbage, mustard greens, and broccoli. Continuous use of synthetic pesticides can cause pest resistance. This study aims to evaluate the acute toxicity and potential of Jahe Emprit (Zingiber officinale var. amarum) extract which is rich in gingerol as a botanical biopesticide against the third instar larvae of Crocidolomia pavonana, which is one of the main pests of Brassicaceae plants. This study was conducted using an experimental method with a Randomized Block Design (RBD) using four extract concentration treatments, namely 2%, 4%, 6%, and 8%, and used a negative control in the form of distilled water and a positive control pesticide Fenite 150 OD. Analysis of the active compound gingerol in the extract was carried out using High-Performance Liquid Chromatography (HPLC). The extract mortality test was carried out to determine Lethal Time 50 and Lethal Concentration 50 through the Topical Application method. The results of HPLC analysis of the bioactive gingerol compound showed a high level of 8.082 µg/mL. Mortality tests showed a significant increase in larval mortality along with increasing concentration and duration of exposure. The (LC50) value was recorded at 4.52%, indicating that the extract has a fairly high potential for acute toxicity. However, based on the (LT50) value of 60.26 hours (±2.5 days), this extract is included in the category of insecticides with a slow-acting mechanism, which is a typical characteristic of plant secondary metabolites. The most efficient extract concentration, namely 8%, was able to produce mortality of up to 70% and efficacy of 66.70% after 72 hours of treatment. The novelty of this research is the use of standardized ginger extract with verification of gingerol content through HPLC as a basis for biopesticide development., providing an alternative for sustainable pest control, and supporting environmentally friendly agricultural practices.

Abstract

Spodoptera litura is a polyphagous pest that causes serious damage to various agricultural commodities. Control methods that still rely on synthetic pesticides negatively influence the environment and health, necessitating more environmentally friendly control alternatives. This study aims to determine the efficacy of ginger extract (Zingiber officinale var. amarum), which contains shogaol compounds, on the mortality of third instar S. litura larvae. The study was conducted experimentally using a one-factor completely randomized design (CRD) with six treatment levels: a positive control (fenite 150 OD), a negative control (Aquades), and ginger extract concentrations of 2%, 4%, 6%, and 8%, each with four replicates. Mortality tests were conducted using the topical application method and observed for 72 h. Data were analyzed using ANOVA, Tukey's test, and probit analysis to determine the LC₅₀, LC₉₀, LT₅₀, and LT₉₀ values. The results showed that ginger extract treatment significantly influenced the mortality of S. litura larvae (Sig. = 0.032). The 8% concentration resulted in the highest mortality of 82.5% at 72 h and the highest efficacy value of 82.5%, approaching that of the positive control. The LT₅₀ and LT₉₀ values were 37.15 hours and 64.81 hours, respectively, while the LC₅₀ and LC₉₀ values were 0.815% and 23.134%. The extract treatment also caused morphological changes in the larvae, including changes in body color, shrinkage, and tissue damage. Emprit ginger extract containing 6.8% shogaol, particularly at an 8% extract concentration, has the potential to serve as an effective environmentally friendly biopesticide in controlling S. litura.

Abstract

This study aimed to evaluate the antifungal activity of emprit ginger (Zingiber officinale var. amarum) extract containing 6-gingerol against Fusarium oxysporum under in vitro conditions and to determine the concentration with the highest inhibitory effect. The experiment was conducted using a completely randomized design with six treatments consisting of a negative control, emprit ginger extract at concentrations of 0.5%, 1.5%, 2.5%, and 5%, and a synthetic fungicide as the positive control, with five replications for each treatment. The 6-gingerol content was quantified using High-Performance Liquid Chromatography (HPLC), while antifungal activity was assessed using the poisoned food technique on Potato Dextrose Agar (PDA) medium. Data were analyzed using analysis of variance (ANOVA), followed by the Honestly Significant Difference (HSD) test at a 5% significance level. The results revealed that emprit ginger extract contained 6-gingerol at 8.082 µg/mL. Growth inhibition of F. oxysporum increased with increasing extract concentrations, reaching the highest inhibition (56.49%) at 5% extract concentration. Under the tested in vitro conditions, this treatment showed higher inhibition than the positive control (43.26%). The extract also induced morphological alterations, including reduced mycelial density, pigmentation changes, and restricted colony expansion. These findings indicate that emprit ginger extract containing 6-gingerol has potential as a botanical biofungicide candidate for suppressing F. oxysporum. Further validation through in vivo and field-scale studies is required before practical application.
