Abstract

Exploration of microorganisms containing secondary metabolite compounds with antimicrobial properties, especially in marine fish pathogenic bacteria, is urgently required. This study aims to test the antibacterial effectiveness of endophytic fungal extracts from the mangrove plant Rhizopora apiculata in the Petengoran Mangrove Forest against pathogenic bacteria in marine fish such as V. alginolyticus and P. damselae spp. The endophytic fungal isolate found was a fungus from the genus Aspergillus sp after observing its morphological form macroscopically and microscopically. Ethyl acetate extract of mangrove endophytic fungi with a concentration of 500 µg/disc and 1000 µg/disc was dripped onto a 6mm paper disc and then placed on the pathogenic bacterial isolate. Measurement of the diameter of the clear zone produced around the paper disc after 24 hours concluded that the antibacterial effectiveness of the endophytic fungal extract against V. alginolyticus was 27.27% (500 µg/disc) and 32.32% (1000 µg/disc). Meanwhile, its effectiveness against P. damselae spp is 27.27% (500 µg/disc) and 34.82% (1000 µg/disc). The isolate extract from the endophytic fungus Aspergillus in this study has the potential to produce antibacterial compounds in marine fish pathogens.

Abstract

Penerapan probiotik cair Lactobacillus rhamnosus dalam pemeliharaan ikan badut (Premnas epigramma) bertujuan untuk menentukan dosis efektif pemberian probiotik terhadap pertumbuhan dan kualitas air media pemeliharaan. Ikan badut dipelihara dalam akuarium 60 L dengan kepadatan 2-3 ekor per liter selama 42 hari pemeliharaan. Perlakuan diferensiasi dengan dosis probiotik cair 0 mL; 5 mL; 10 mL per 50 liter air laut. Hasil penelitian menunjukkan bahwa pertambahan berat ikan pada dosis probiotik 0 mL, 5 mL, dan 10 mL adalah 0,23 g, 0,33 g, dan 0,32 g, secara berturut-turut. Terdapat efek positif pemberian probiotik terhadap perubahan warna kulit ikan yang menjadi lebih cerah. Parameter kualitas air pemeliharaan Premnas epigramma seperti suhu, salinitas, pH, dan DO yang diamati selama percobaan masih sesuai untuk budidaya ikan laut. Konsentrasi nitrit dalam air menurun pada kedua perlakuan dengan penambahan probiotik, tetapi amonia menurun menjadi 0,1 mg/L hanya pada dosis probiotik 5 mL per 50 mL air laut. Pemberian probiotik selama pemeliharaan ikan badut belum terlalu efektif menekan populasi bakteri Vibrio pigmen hijau. Akan tetapi dapat menekan populasi Vibrio pigmen kuning yang cenderung patogen.
