Abstract

Background: Pancreatic β-cells compensate for the increased insulin demand due to increasing glucose levels during pregnancy or in gestational diabetes mellitus (GDM). The enzyme that regulates the rate of gluconeogenesis production is PEPCK. Circulatory and metabolic disturbances in the brain caused by hyperglycemia could affect brain-derived neurotrophic factor (BDNF) expression as an agent for neurodevelopment. Objective:  The purpose of this work is to ascertain the impact of combining nanoemulsions of Rosmarinus officinalis and Centella asiatica on the expression of BDNF and PEPCK. Methods: This study was a true experimental laboratory using zebrafish larvae. The larvae were exposed to 3% glucose and a combination of nanoemulsion extracts. The study sample of zebrafish larvae at 72 hpf (hour post fertilization) was divided into control, glucose, treatment 1, 2, and 3, with the combination of Rosmarinus officinalis and Centella asiatica nanoemulsions at 2.5 μg/ml, 5 μg/ml, and 10 μg/ml, respectively. PEPCK and BDNF expression were measured by the real-time PCR method. Results: The expression of PEPCK increased significantly in the glucose group (3.05 ± 0.27). The combination of Rosmarinus officinalis and Centella asiatica nanoemulsions significantly decreased PEPCK (p-value 0,001 < 0.05). The BDNF level did not significantly differ (p-value 0.253 >0.05) among groups, but the highest level (1.06 ± 0.60) was gained in the T2 group, which received an embryonic medium (EM), 3% glucose, and the combination of Rosmarinus officinalis and Centella asiatica nanoemulsions at 5μg/ml. Conclusion: The combination of Rosmarinus officinalis and Centella asiatica nanoemulsions significantly reduces PEPCK expression but does not affect BDNF expression.

Abstract

Gestational Diabetes Mellitus (GDM) is diabetes that occurs during pregnancy. GDM is characterized by hyperglycemia. Hyperglycemia causes an inflammatory response characterized by the excretion of pro-inflammatory cytokines, one of which is IL-6. Centella asiatica and Rosmarinus officinalis are known as anti-diabetic and anti-inflammatory. This study aims to study the effect of the combination of nanoemulsion Centella asiatica and Rosmarinus officinalis on the expression of Interleukin-6 in Zebrafish larvae induced by 3% glucose. Hyperglycemia is known from increased levels of Phosphoenolpyruvate carboxykinase in Zebrafish larvae. Phosphoenolpyruvate carboxykinase and Interleukin-6 expression were measured by conventional PCR method. The combination of Rosmarinus officinalis nanoemulsion and Centella asiatica was given with three different doses, namely at a concentration of 2.5, 5 and 10 µg/mL. The statistical analysis used was One Way ANOVA to measure Interleukin-6 expression levels. The results of this study prove that the combination of Rosmarinus officinalis and Centella asiatica nanoemulsions reduces the expression of Interleukin-6. The dose closest to the negative control was at a concentration of 5 μg/mL, although the final result was statistically significant (p<0.05). In conclusion, combining nanoemulsion Centella asiatica and Rosmarinus officinalis with the right dose reduced levels of Interleukin 6 in zebrafish larvae induced by high glucose.

Abstract

Diabetes gestasional adalah jenis diabetes yang terjadi selama masa kehamilan. GDM ditegakkan dengan kondisi hiperglikemi dalam darah. Hiperglikemi memicu respon inflamasi yang ditandai dengan ekskresi sitokin pro inflamasi, salah satunya adalah TNF-α. Centella asiatica dan Rosmarinus officinalis dikenal sebagai anti-diabetes dan anti-inflamasi. Penelitian ini bertujuan untuk mempelajari efek kombinasi nanoemulsi Centella asiatica dan Rosmarinus officinalis terhadap ekspresi TNF-α di larva Zebrafish yang diinduksi glukosa 3%. Hiperglikemi ditandai dengan terjadinya peningkatan kadar Phosphoenolpyruvate carboxykinase di larva Zebrafish. Phosphoenolpyruvate carboxykinase dan TNF-α akan diukur menggunakan metode PCR konvensional. Kombinasi nanoemulsi Rosmarinus officinalis dan Centella asiatica diberikan dengan tiga dosis yang berbeda yaitu dengan konsentrasi 2.5, 5 dan 10 πg/mL. Analisis statistic yang digunakan adalah One Way ANOVA untuk mengukur kadar ekspresi TNF-α. Hasil dari penelitian ini menunjukkan bahwa kombinasi nanoemulsi Rosmarinus officinalis dan Centella asiatica tidak signifikan mempengaruhi kadar ekspresi TNF-α. Dosis yang menurunkan kadar ekspresi TNF-α adalah kelompok dengan konsentrasi 2,5 πg/mL, meskipun hasil akhir memiliki nilai tidak signifikansi . Kesimpulan dari penelitian ini adalah mengkombinasikan nanoemulsi Rosmarinus officinalis dan Centella asiatica dengan dosis 2,5 πg/mL; 5 πg/mL; dan 10 πg/mL tidak signifikan mempengaruhi kadar TNF-α di larva Zebrafish yang diinduksi glukosa tinggi (3%).
