Abstract

Serum merupakan sediaan kosmetik dengan konsentrasi tinggi dan memiliki kemampuan lebih dalam mengangkut bahan aktif ke kulit sehingga lebih efektif dalam mengatasi permasalah kulit akibat radikal bebas. Daun binahong merah (Anredera cordifolia) diketetahui mengandung metabolit sekunder yang berpotensi sebagai antioksidan yaitu flavonoid, fenol, tanin, dan vitamin C. Serum dengan kandungan antioksidan dapat dibuat dalam bentuk nanopartikel karena memiliki tetesan kecil dan luas permukaan yang besar. Tujuan dari penelitian ini adalah untuk mengetahui karakteristik dan aktivitas antioksidan serum nanopartikel ekstrak daun binahong merah (Anredera cordifolia). Penelitian ini menggunakan jenis penelitian kuantitatif dengan model penelitian true experimental laboratory dan desain rancangan acak lengkap. Pembuatan NLC dilakukan dengan metode emulsifikasi-sonikasi, karakteristisasi serum meliputi ukuran partikel NLC dengan metode PSA dan efisiensi penjerapan NLC dengan spektrofotometer UV-Viss, kemudian serum di evaluasi dengan parameter uji organoleptis, uji homogenitas, uji pH, uji daya sebar, uji iritasi, uji kelembapan, dan uji aktivitas antioksidan dengan metode DPPH. Data yang dihasilkan dianalisis menggunakan ANOVA one way. Karakteristik NLC belum memenuhi ukuran partikel dengan rata-rata 3,811 µm dan efisiensi penjerapan 99,96%. Parameter evaluasi sediaan sudah memenuhi standar sediaan topikal pada uji organoleptis menghasilkan warna hijau muda, bau khas dan tekstur cair; hasil homogen; pH 5; dan daya sebar 5,5-7,5 cm. Serum tidak mengiritasi kulit dan memenuhi standar kelembapan kulit dengan rentang 46%-55%. Nilai IC50 F0, F1, F2, F3 berturut-turut 834,12 ppm, 149,71 ppm, 130,24 ppm, 99,44 ppm. Serum memiliki karakteristik, sifat fisik dan kimia sesuai dengan sediaan topikal dan aktivitas antioksidan dalam kategori kuat pada F3.

Abstract

Antioxidants are compounds that have the ability to againts the harmful effects of free radicals. Antioxidant activity was influenced by active compounds, growing environment, cultivation systems, techniques and handling of raw materials. Hydroponics is a plant cultivation system that does not use soil as media. Differences in hydroponic and conventional systems in soil use can affect antioxidant activity. This study aims to conduct a qualitative test of antioxidants through phytochemical screening and analysis of thin layer chromatography (TLC) and to test the antioxidant activity of hydroponic and conventional red spinach leaf extract. Red spinach leaves were extracted by maceration method for 3 days with stirring every 1x24 hours. The red spinach leaf extract was then organoleptically tested for color, shape, and odor. Phytochemical screening was carried out to determine the content of flavonoids and saponins using color and foam reactions. The screening results were confirmed by thin layer chromatography (TLC) analysis with hexane:ethyl acetate as the mobile phase in a ratio of 5:5. Antioxidant activity test with DPPH Method. The results of the calculation of the yield on hydroponic red spinach leaf extract was 11.5% and conventional was 15.1%. The results of phytochemical screening and TLC analysis showed that both hydroponic and conventional red spinach leaf extracts were positive for flavonoid and saponin compounds. The IC50 value of hydroponic red spinach leaf extract was 142.31 while conventional was 73.73 ppm. The antioxidant activity of hydroponic red spinach leaf extract was in the medium category and conventional in the strong category.
