Abstract

Penelitian ini bertujuan untuk menentukan keberadaan proteolitik, waktu optimum produksi danaktivitas protease dari bakteri Bacillus subtilis. Metode yang digunakan adalah metode eksperimental, data dianalisa secara deskriptif kuantitatif.  Bahan yang digunakan adalah bakteri Bacillussubtilis terseleksi yang berasal dari saluran pencernaan ikan Nila (Oreochromis niloticus). Hasil penelitian menunjukkan bahwa bakteri Bacillus subtilis mempunyai aktivitas proteolitik. Diameter zone hambat yang terbentuk menunjukkan secara kualitatif tingginya kemampuan proteolitik enzim protease yang dihasilkan atau juga tingginya jumlah enzim yang diproduksi dan dilepas keluar. Keberadaan enzim protease ekstraseluler ini sangat penting bagi kehidupan bakteri karena menyediakan kebutuhan senyawa bernitrogen yang dapat diangkut ke dalam sel. Jenis-jenis bakteri yang mempunyai kemampuan mensekresikan enzim protease ini memiliki potensi yang besar untuk digunakan sebagai agensia pembersih bahan pencemar yang bersifat protein. Jenis medium sangat berpengaruh terhadap produksi enzim protease bakteri Bacillus subtilis. Pola waktu optimum produksinya sejalan dengan pola kurva pertumbuhan bakteri secara umum. Hasilpenelitian menunjukkan bahwapada jam ke-46,bakteri menghasilkanaktivitasproteaseyangpalingtinggi,yaitusebesar 2,163U/mL.

Abstract

Bacillus subtilis strain VITNJ1 merupakan bakteri potensial yang berasal dari saluran pencernaan ikan nila (Oreochromis niloticus). Penelitian ini bertujuan untuk menganalisis aktivitas enzim protease dari bakteri Bacillus subtilis strain VITNJ1. Metode yang digunakan adalah eksperimental, data dianalisa secara deskriptif kuantitatif. Hasil penelitian menunjukkan B. subtilis strain VITNJ1 memiliki kemampuan produksi enzim protease yang diukur dari indeks proteolitiknya yakni sebesar 26 mm. Waktu inkubasi optimum enzim protease bakteri B. subtilis strain VITNJ1 dicapai pada jam ke-54 dengan aktivitas enzim protease sebesar 9,10 U/mL; kadar protein 110,64 mg/mL; dan aktivitas spesifik sebesar 0,082 U/mg; dan diharapkan dapat dijadikan sebagai probiotik untuk budidaya ikan nila.Bacillus subtilis strain VITNJ1 is a potential bacterium derived from nile tilapia’s (Oreochromis niloticus) digestive tract and known to have proteolytic activity. This research aimed to determine the protease activity of B. subtilis strain VITNJ1 bacterium. The method used were experimental design, the data were analyzed in quantitative descriptive. The research result confirms that B. subtilis strain VITNJ1 can produce protease. The proteolytic activity was 26 mm. The optimum incubation period of B. subtilis strain VITNJ1 was obtained at hour-54 with the proteolytic enzyme activity of 9.10 U/mL, the protein concentration of 110.64 mg/mL, and the specific activity of 0.082 U/mg. This research suggests that the bacteria strain could be used as an alternative probiotic bacterium in nile tilapia farming.

Abstract

This study aimed to determine fishers' perceptions of IFM as a cold chain tool in Padang City. The data used in this study were from interviews/questionnaires with IFM managers and anglers around the City of Padang. We also employed fish caught data in 2019 and ice flake sales from 2 (two) IFM providers in Padang City from January to August 2019. The statistical analysis used was linear regression. Most anglers did not know the IFM machine's availability and how to treat the fishery products using ice (cold chain systems). The lack of socialization from the provider to fishers and fisheries business parties in the surrounding area is believed to cause the decreased demand for ice flakes, resulting in the inactive operation of IFM’s in Padang City.

Abstract

ABSTRACT Training on making paving blocks from plastic waste (colored and or clear bags) for cleaning services of campus 3 Universitas Bung has been conducted. The Community Service (PKM) aims to transfer knowledge and skills in the manufacture of paving blocks which have a higher selling value than plastic bag waste. The long-term goal of this program is to support the Padang City Government program to reduce the amount of waste to the Final Disposal Site and become a community business opportunity to increase sources of income. The methods used in this PKM are: 1) preparation of socialization activities, and 2) counseling and training. The campus cleaning service is enthusiastic about this activity and considers that the training adds to their insight in the utilization of plastic waste into paving blocks, which they never knew about that.

Abstract

The remainder of human activity in the form of solids, whether deliberately disposed of or not into the marine environment is called marine debris. This study aims to determine the weight, composition and density of marine debris in Purus, Padang City. Sampling was carried out from February to April 2021. Determination of the location by purposive sampling using line transect sampling method. Waste is separated based on macro and meso sizes and then the weight and amount of each type of waste is calculated. From the research results found 9 thypes of waste, namely types of plastic waste, plastic foam, cloth, glass and ceramics, metal, paper and cardboard, rubber and wood and another materials. There were 9805 pieces of waste with a total weight of 27170.74 gr, consisting of 21554.7 gr of macro waste and 5616.4 gr of meso waste. Based on the weight of waste, the most dominant types found were cloth (9311.6 gr) for macro waste and 5616.04 gr for meso waste. The highest density is found in the type of wood with WD02 code as much as 50.92 pieces/m2 for macro waste and WD06 as many as 226.8 pieces/m2 for meso waste.

Abstract

Marine debris is all forms of solid objects that are disposed of intentionally or unintentionally into the marine environment. Research on the type, composition, amount, and density of marine debris that washed up on Air Manis Beach and Pasir Jambak Beach has never been done. The purpose of this study was to determine the source, amount, type and density of marine debris found at Air Manis Beach and Pasir Jambak Beach, Padang City. The research was conducted from March to July 2022. The method used to collect waste was using the line transect method, the location determination was carried out intentionally according to research needs (purposive sampling). All waste obtained during the study was grouped by type, amount and size. Based on the research, it is known that the garbage stranded on the two beaches comes from the river, the activities of residents around the location, fishing activities and visiting tourists. It was found that there were 36232.24 grams of waste consisting of 36195.4 grams of macro waste and 36.84 grams of meso waste. Macro waste with the highest percentage was found in plastic waste (86.84%), and for meso-sized waste was found in metallic waste (72.27%). The density of marine debris at Air Sweet Beach and Pasir Jambak Beach is 23.36 pieces/m2 for macro waste and 4.2 pieces/m2 for meso waste.

Abstract

From several studies it was revealed that there are still many people who use formalin and borax in food because these ingredients are easy to obtain. Nagari Sasak is one of the areas in the Sasak Ranah Pasisia District, West Pasaman Regency, located on the coast where most of its residents produce salted fish for their livelihood. The procedure used by fishermen in this area when processing salted fish is still traditional.  This study aims to analyze the presence of formalin and borax in salted fish processed by conventional fishermen in Nagari Sasak, Sasak Ranah Pasisia district, West Pasaman regency. The samples used in this study included seven types of dried salted fish obtained from three processors in KanagarianSasak. For more details, the types of salted fish tested consist of three kinds of anchovy (Stolephorus sp.), giant trevally (Caranx sp.), yellow jack (Caranx sp.), barracuda (Sphyraena jello), and tobi (Sardinella fimbriata). The research sample was obtained by purposive sampling way. In addition, the method used in this research is the descriptive method, while the data analysis is gained through qualitative descriptive methods. The test parameters of this study included formalin and borax qualitative tests and organoleptic tests of dried salted fish. The results showed that five out of seven salty fish samples, as much as 71.43%, were detected to contain formalin.

Abstract

Penggaraman dan pengeringan merupakan metode yang sering digunakan oleh nelayan pengolah untuk memperpanjang mutu ikan. Tujuan penelitian ini adalah: mengidentifikasi keberadaan formalin pada produk olahan ikan asin, dan menganalisis kandungan proksimat ikan asin. Bahan  yang digunakan dalam penelitian ini adalah ikan Talang-talang (Chorinemus tala) dan Gulamah (Pseudocienna amovensis) asin yang berasal dari beberapa pengolah di daerah Gasan Gadang, Kabupaten Padang Pariaman, Sumatera Barat. Hasil penelitian menunjukkan bahwa tidak ditemukan penambahan formalin pada seluruh sampel ikan asin. Rata-rata kadar protein ikan asin adalah 26,5589%, kadar lemak 5,7499%, kadar air 33,4258%, kadar abu 18,5629%, karbohidrat 15,7023%, dan garam 15,5116%.

Abstract

Pempek ikan merupakan produk olahan hasil perikanan yang memiliki gizi yang baik bagi tubuh manusia, tetapi dikhawatirkan para pengolah pempek ikan menggunakan bahan tambahan non-pangan berbahaya seperti boraks untuk meraup keuntungan. Penelitian ini bertujuan untuk menentukan ada atau tidaknya keberadaan boraks pada olahan pempek ikan di beberapa warung di Kota Padang. Metode yang dipakai adalah metode deskriptif dengan populasinya adalah beberapa warung pempek ikan yang berada di Kota Padang. Pengambilan sampel menggunakan teknik purposive sampling. Sampel diuji dengan menggunakan test kit boraks dan selanjutnya di uji secara organoleptik. Hasil penelitian pada uji test kit boraks tidak ditemukannya kandungan boraks pada sampel A, B, C, D dan E yang berarti pada semua warung yang diuji negatif menggunakan boraks. Pada uji organoleptik nilai rata-rata kenampakan 3,77, aroma 3,66, rasa 3,78, warna 3,69 dan tekstur 3,78. Faktor yang mempengaruhi perbedaan dari setiap parameter adalah komposisi, bahan, bumbu, dan keterampilan pengolah setiap warung berbeda.

Abstract

Ikan tongkol asap yang diproduksi oleh masyarakat daerah Dusun Berkat di oleh secara tradisional dengan pengasapan panas secara terbuka. Penelitian ini bertujuan mengetahui karakteristik sensori, kimia dan mikrobiologi ikan tongkol asap yang diolah 3 pengolah rumah tangga ikan asap di Dusun Berkat Desa Tuapejat Kabupaten Kepualaun Mentawai. Metode yang digunakan dalam penelitian ini adalah metode uji sensori analisis proksimat, uji ALT dan analis kapang yang akan dianalisis secara deskriptif kuantitaif. Sedangkan analisis pengolahan ikan asap akan di analisis secara deskriptif kualitatif. Berdasarkan penelitian ini di ketahui uji sensori ikan tongkol asap (kenampakan, bau, rasa dan tekstur) memiliki nilai yang sesuai dengan SNI 2725:2013. Kadar air ikan tongkol asap pada penelitian ini tertinggi 59.29 % dan terendah 55.59 %. Kadar protein ikan asap tongkol tertinggi adalah 30.04 % dan terendah adalah 27.18 %. Hasil Total bakteri dan kapang ikan tongkol asap daerah Dusun Berkat melebihi batas syarat dari SNI 2725:2013. Pengolahan ikan asap pada penelitian ini masih bersifat tradisional.
