Abstract

Free radicals are reactive compounds that can cause damage to cells, enzymes, lipids, and even DNA, thus triggering various degenerative diseases. Antioxidants neutralize free radicals and prevent oxidative damage. Chinese betel herb (Peperomia pellucida L.) is known to contain phenol and flavonoid secondary metabolite compounds that act as natural antioxidants. This study aims to test the antioxidant activity of ethanol extract of Chinese betel herb with DPPH (2,2-diphenyl-1-picrylhydrazyl) method. Extraction was done by maceration method using 80% ethanol solvent. Antioxidant activity was tested by reacting the extract against DPPH solution and measuring the absorbance using UV-Vis spectrophotometry. The results showed an IC₅₀ value of 32.97 ppm, which is classified as very strong antioxidant activity. These results indicate that ethanol extract of Chinese betel herb has great potential as a source of natural antioxidants.

Abstract

Oxidative stress is a condition resulting from an imbalance between free radicals and the body's antioxidant system, which can damage cells and trigger degenerative diseases such as diabetes, cancer, atherosclerosis and heart disease. Antioxidants play an important role in neutralizing free radicals to prevent cell damage. Papaya leaves (Carica papaya L.) are known to contain bioactive compounds that have potential as a source of natural antioxidants. This study aims to test the antioxidant activity of papaya leaf ethanol extract using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, with quercetin as a comparator. Extraction was done by maceration using 80% ethanol. Antioxidant activity was measured based on the decrease in absorbance of DPPH solution at a wavelength of 517 nm using UV-Vis spectrophotometer. The results showed that papaya leaf extract had strong antioxidant activity with IC₅₀ value of 21.35 ppm, while quercetin was 4.84 ppm. Thus, the ethanol extract of papaya leaves showed high potential as an effective natural antioxidant.

Abstract

Diabetes mellitus (DM) is a growing public health concern worldwide, particularly in Indonesia, where its prevalence continues to rise, causing significant health and economic burdens. The need for effective, accessible, and safe therapies is critical, as current treatments often come with side effects and limitations. Alternative natural treatments, such as herbal remedies, are being explored for their potential to offer safer and more sustainable options for managing DM. Objective: This study aims to evaluate the antidiabetic effectiveness of ethanol extract of sweet mango leaves (Mangifera indica L. var. sweet mango) on reducing blood glucose levels in streptozotocin (STZ)-induced diabetic mice. Methodology: A true experimental post-test only control group design was employed using mice (Mus musculus) divided into six groups: normal control, negative control (STZ), positive control (glibenclamide), and three treatment groups of mango leaf extract with doses of 50, 100, and 150 mg/kgBW. The extraction was carried out using the maceration method with 70% ethanol, and the antidiabetic activity was tested over a 15-day period. Results: The results showed that administration of ethanol extract of sweet mango leaves significantly reduced blood glucose levels (p < 0.001) compared to the negative control. A dose-response pattern was observed, with a dose of 150 mg/kgBW providing the most significant blood glucose lowering effect, closely approaching the effectiveness of glibenclamide. Discussion: The antidiabetic activity of the extract is thought to be related to its bioactive compounds, such as flavonoids, phenolics, and mangiferin, which are believed to enhance insulin sensitivity, inhibit gluconeogenesis, and protect pancreatic β cells from oxidative stress. Originality and Contribution: This study provides new insights into the antidiabetic potential of the ethanol extract of sweet mango leaves (Mangifera indica var. Arum Manis), a variety not extensively studied for its effects on diabetes. The research contributes to the growing body of knowledge on natural-based therapies for DM, offering an alternative to conventional treatments with potentially fewer side effects and wider accessibility. The findings support the further development of mango leaf extract as a natural candidate for diabetes management.

Abstract

Bacterial infection in chronic wounds, particularly diabetic foot ulcers in patients with diabetes mellitus, remains a serious health problem due to their susceptibility to pathogenic bacteria such as Pseudomonas aeruginosa. Soursop leaves (Annona muricata L.) are rich in bioactive compounds with potential antibacterial properties, and several previous studies have reported their antibacterial activity. Therefore, this study aimed to evaluate the antibacterial activity of soursop leaf extract against these two bacteria in vitro using the disc diffusion method, with inhibition zone diameter as an indicator. This experimental study focused on assessing the interaction of 96% ethanolic soursop leaf extract obtained by maceration against Pseudomonas aeruginosa at various concentrations. The results demonstrated concentration-dependent antibacterial activity, in which increasing extract concentration led to larger inhibition zones. However, the antibacterial effect of the extract was lower than that of the positive control (gentamicin), which may be attributed to the use of crude secondary metabolites and the limitation of the disc diffusion method as a preliminary screening tool with restricted quantitative accuracy. Keywords: soursop leaves (annona muricata L.), antibacterial activity, pseudomonas aeruginosa, disc diffusion method, ethanol extract   Abstrak Infeksi bakteri pada luka kronik, terutama ulkus diabetikum pada penderita diabetes melitus, tetap menjadi masalah kesehatan serius karena kerentanan terhadap bakteri patogen seperti Pseudomonas aeruginosa. Daun sirsak (Annona muricata L.) kaya akan senyawa bioaktif yang berpotensi sebagai antibakteri alami, dan beberapa studi sebelumnya telah menunjukkan aktivitasnya. Oleh karena itu, penelitian ini bertujuan menguji aktivitas antibakteri ekstrak daun sirsak terhadap kedua bakteri tersebut secara in vitro menggunakan metode difusi cakram, dengan mengukur zona hambat sebagai indikator. Penelitian ini bersifat eksperimental, fokus pada pengujian interaksi ekstrak etanol 96% daun sirsak yang diperoleh melalui maserasi terhadap Pseudomonas aeruginosa dengan variasi konsentrasi. Hasilnya menunjukkan aktivitas antibakteri yang bergantung pada konsentrasi, di mana peningkatan konsentrasi ekstrak meningkatkan diameter zona hambat. Namun, daya hambat ekstrak ini masih lebih rendah dibandingkan kontrol positif gentamisin, hal ini dikarenakan ekstrak daun sirsak (Annona Muricata L.) masi merupakan metabolit sekunder, dan metode difusi cakram hanya cocok sebagai skrining awal dengan keterbatasan dalam akurasi.
