Abstract

We cloned and sequenced cDNA encoding p48 subunit of the chicken CAF-1, chCAF-1p48, and histone acetyltransferase-1, chHAT-1 from chicken DT40 cell lines. We showed that the p48 subunit of CAF-1 tightly binds to two regions of chicken histone deacetylase 2, chHDAC-2, located between amino acid residues 82-180 and 245-314, respectively. We also established that two N-terminal, two C-terminal, or one N-terminal and one C-terminal WD repeat motif of chCAF-1p48 are required for this interaction. The GST pulldown assay, involving truncated and missense mutants of chCAF-1p48, revealed not only that a region containing the seventh WD dipeptide motif of chCAF-1p48, comprising amino acids 376-405, binds to chHAT-1 in vitro, but also that mutation of the motif has no influence on the in vitro interaction. We also established that the region, which is located between amino acids 380-408 of chHAT-1 and contains a leucine zipper motif, is required for its in vitro interaction with chCAF-1p48. Mutation on each of four Leu residues in the leucine zipper motif of chHAT-1 causes the disappearance of the interaction with chCAF-1p48. These results should be useful information for understanding the participation of chCAF-1p48 protein as histones chaperone in DNA-utilizing processes, such as replication, recombination, repair and gene expression in DT40 chicken B cell.

Abstract

The ability of tetrakis-thiomethyl-C-4-methoxyphenylcalix[4]resorcinarene for trapping Pb(II) and Cd(II) from aqueous solution at different pH, contact times, and initial metal concentration was investigated. Concentration of metal ions before and after interaction was analyzed by Atomic Absorption Spectrophotometer (AAS). The result indicated that tetrakis-thiomethyl-C-4-methoxyphenylcalix[4]resorcinarene is a good host for metal ions, where Cd(II) sorption capacity was higher than that of Pb(II). Effect of pH on batch experiments for the mentioned ions indicated that the optimum pH for metal binding were 5 for both of lead(II) and cadmium(II). In order to investigate the mechanism of adsorption, the 1st-order, pseudo 1st-order, and pseudo 2nd-order kinetic models were used. The adsorption model of metal ions on the resorcinarene followed pseudo second order of Ho & McKay expression. The equilibrium adsorption isotherm has been analyzed by Langmuir and Freundlich equations. Langmuir model had the higher correlation coefficient than that of Freundlich model.

Abstract

This paper describes 3-hydroxyurs-12-en-28-oic acid and its structural isomer 3-hydroxyolean-12-en-28-oic acid isolated from Fragraea fragrans fruits and their biological activities; anti-tumor, anti-inflammation, anti-microbial, and anti-fungal included their ultra violet photo-protective effect after exposed under sunlight radiation. They are useful for cosmetic ingredient. The above triterpenes are very promoting compounds for leukemia L1210 anti-tumor due to limited reports dealing with this type triterpenoid anti-tumor test. They significantly gave IC50 value of 5.78 µg/mL against leukemia L1210 cells, relatively closed to IC50 value of drugs, 4.0 µg/mL [12-13,17].

Abstract

A sensitive and selective absorptive stripping voltammetric (AdSV) method to determine Cd(II), Cu(II), Pb(II) and Zn(II) in sea water is proposed. The aim of this study was to get optimum condition for the determination of Cd(II), Cu(II), Pb(II) and Zn(II). Absorptive stripping voltammetry has been used for ultra trace determination of Cd(II), Cu(II), Pb(II) and Zn(II) using calcon as a complexing agent (ligand). In this case, the optimum conditions were reached at 0.1 M KCl supporting electrolyte, concentration of 0.5 mM calcon for Cd(II), Cu(II) 0.3 mM while 0.7 mM for Pb(II) and Zn(II), pH 6 for Cu(II) and pH 7 for Cd(II), Pb(II) and Zn(II), accumulation potential -0.5 V for Cu(II) and Pb(II) and -0.6 V for Cd(II) and Zn(II) and accumulation time 70 sec for Cd(II), 90 sec for Cu(II) and Pb (II) while 50 s for Zn(II). At the optimum condition the relative standard deviations were 7.80%, 4.25%, 8.70% and 0.86% for Cd(II), Cu(II), Pb(II) and Zn(II) respectively for eight replicates (n = 8) measurements of 10 μg/L Cd(II), Cu(II), Pb(II) and Zn(II). The method was applied for the direct determination of Cd(II), Cu(II), Pb(II) and Zn(II) in sea water around Bungus, Padang City. Concentration Cd(II), Cu(II), Pb(II) and Zn(II) in sample were 13.200 μg /L for Cd(II), 17.200 μg/L for Cu(II), 0.089 μg/L for Pb(II) and 62.000 μg/L for Zn(II) with recovery of 98.68%, 97.99%, 96.17% and 99.96% for Cd(II), Cu(II), Pb(II) and Zn(II), respectively.

Abstract

Cryogenic transmission electron microscopy (Cryo-TEM) was used to investigate the aggregates morphology and properties of candle tree (Aleurites moluccana) endosperm, sesame (Sesamum indicum L. syn.) seeds, and coconut (Cocos nucifera) endosperm phospholipids in dilute aqueous system. The micrographs showed that candle tree phospholipids formed planar bilayer and cluster of vesicles with lipid droplets, while coconut and sesame phospholipids formed well-defined unilamellar vesicles. The vesicles size could be as small as 50 nm in diameter. Coconut phospholipids also showed a good bending ability. Formation of clusters of vesicles was also found in coconut phospholipids dispersion, but this cluster was easily broken by extrusion through a small pore membrane.

Abstract

Binuclear complex, [N(n-C4H9)4][MnIIFeIII(ox)3] (ox=oxalate), has been synthesized and characterized. Binuclear complex with formula of [N(n-C4H9)4][MnIIFeIII(ox)3] was synthesized by forming precursor K3[Fe(ox)3] which was followed by adding manganese chloride and tetrabutyl ammonium chloride. Characterization of the complex was made by microanalysis, FT-IR spectroscopy, x-ray diffraction and Magnetic Susceptibility Balance. Magnetic moment of binuclear complex [N(n-C4H9)4][MnIIFeIII(ox)3] is 7.81 BM. The compound has higher magnetic moment than the previously reported compound with different organic cation, [N(n-C5H11)4][MnIIFeIII(ox)3]. This behavior might be associated with the decreasing range among layers on polymeric network.

Abstract

A research has been conducted to investigate the capability of a series of novel calix[6]arenes-based polymers: poly-monoallyloxycalix[6]arene (2a), poly-monoallyloxypenta-estercalix[6]arene (2b) and poly-monoallyloxypenta-acidcalix[6]arene (2c) for trapping of heavy metal cations such as Cd(II), Cu(II) and Cr(III). The existence of active hydroxy group (-OH) and with a tunnel-like structure of the polymers, caused the polymers can be used as adsorbents for heavy metals. The adsorption process was carried out in batch method in the variation of acidity (pH), contact time and initial concentration of metal ions. The results showed that the amount of adsorbed metal increased with the increasing of the pH of metal solution. For these three polymers, the amount of metal ions Cd(II), Cu(II) and Cr(III) adsorbed was optimum at pH 7, 6 and 5 respectively. The optimum contact time for Cd(II) and Cu(II) was 120 min, while that for Cr(III) was 60 min. Study of the adsorption kinetics showed that the adsorption of Cd(II), Cu(II) and Cr(III) using polymer 2a followed kinetics model of Ho. For adsorbent 2b and 2c, the adsorption kinetics of Cd(II) and Cr(III) also followed kinetics model of Ho while for the Cu(II) followed Lagergren kinetic models. Isothermal studies showed that the adsorption of metal ions on all adsorbents tend to follow the Langmuir isotherm. The adsorption energies of the three adsorbents were higher than 23 kJ/mole and polymer 2c has the largest adsorption capacity for Cr(III).

Abstract

Mahkota dewa (Phaleria macrocarpa (Scheff.) Boerl.) which is belong to family of Thymelaeaceae is one of Indonesian traditional medicines. Chemical constituent has been isolated from bark of mahkota dewa. Sample was extracted with methanol. Concentrated methanol extract was extracted by n-hexane, chloroform and ethyl acetate. A Compound that separated and purified by column chromatography from ethyl acetate extract is a red spherical crystal (m.p. 94-95 °C). Its spot gave yellow fluorescence at TLC plate (UV366) and has optical rotation of -9.3°(c. 2 mg/mL, methanol). Structure elucidation by UV, IR, 1H-NMR, 13C-NMR and NMR 2 dimension (HMQC, COSY, HMBC and DEPT-135) spectroscopy show that the compound gives a name macronone. Computational chemistry calculation using Hyperchem on the level of semiempirical method PM3 was confirmed the conformation of macronone. DPPH method shows that macronone has lower antioxidant activity compare to the ethyl acetate extract.

Abstract

A new method for determination of stoichiometry of colorless complexes by using atomic absorption spectrophotometric technique in continuous variation method and slope ratio method was described here. This method can be used in same manner as that of mole ratio method and slope ratio method. In this method atomic absorption spectroscopy was used instead of UV-Vis spectrophotometry. Atomic absorption spectrophotometric technique is superior to UV-Vis spectrophotometry as it can be applied to colorless soluble complexes. Pb(II) and n-benzoyl-n-phenyl hydroxylamine react to form colorless complex at pH 6.5, which can be easily determined by this method. It was found that Pb(II) forms 1:2 complex with n-benzoyl-n-phenyl hydroxylamine and is quantitatively extracted back to aqueous solution for AAS analysis.

Abstract

Chemical stability, reusability, and adsorption selectivity of Cd2+ ionic imprinted Nannochloropsis sp with silica matrix (Cd(II)-IIP) from precursor tetraethyl orthosilicate (TEOS) have been studied through adsorption experiment series with batch method. Nannochloropsis sp (Cd(II)-IIP) material was characterized with an infrared spectrophotometer (IR) to identify the functional groups in this material and identification of metal ion concentration was analyzed with an atomic absorption spectrophotometer (AAS). Chemical stability was determined in solution media of acid, neutral, and base. Adsorption selectivity was obtained with determination of selectivity coefficient (α) of Cd2+ ion toward its ionic pair such as Ag+, Zn2+, Cu2+, and Ni2+ ions. Nannochloropsis sp Cd(II)-IIP material is very stable in acid media and lack stable in base media as well as it can be reused for extraction 4 cycles with adsorption capacity value > 95% using eluent of 0.1 M Na2EDTA. Selectivity of Cd(II)-IIP material upon Cd2+ ion is higher than non imprinted polymer (NIP) and it increases with these orders; Cd2+/Ag+ < Cd2+/Zn2+ < Cd2+/Cu2+ < Cd2+/Ni2+ for each α at metal ionic ratio of 1:1; 0.887; 20.180; 28.053; 33.417, respectively.
