Abstract

Tuber of purple yam (Dioscorea alata L.) has been used as an alternative food in some areas in Indonesia. The tuber contains high carbohydrate, has low glycemic index and is gluten free. Therefore, study on genetic improvement of this species needs to be carried out in order to increase its productivity and to find out new cultivars which can be cultivated in marginal lands. This research was aimed to investigate the effect of NaCl concentration on growth and proline content of purple yam grown in vitro. Shoot tips were cultured on MS (Murashige and Skoog) medium supplemented with NaCl at concentrations of 25; 50; 100; 200 and 250 mM. After six weeks in culture, height of shoots, number of nodes, number of leaves, as well as proline content were recorded. The results showed that shoots grown on MS medium supplemented with NaCl at 25 and 50 mM had better growth compared to control. The best medium for its growth was MS containing 50 mM of NaCl. An increase in NaCl level resulted in decrease of growth. The LD50 value was obtained at 183 mM of NaCl. The highest proline concentration was achieved by shoots grown on the medium supplemented with 100 mM of NaCl. This result indicated that purple yam was tolerant to the increase of NaCl concentration up to 100 mM on MS medium without addition of plant growth regulators. Keywords: purple yam, Discorea alata L., NaCl, in vitro growth, proline content.

Abstract

Polycyclic Aromatic Hydrocarbons (PAHs) as well as crude oil are widespread environmental pollutants. The contamination of air, soil, freshwater (surface water and groundwater), and marine environments by PAHs as well as crude oil has been reported. Of concern to public health is the fact that many PAHs or their metabolites are mutagenic, carcinogenic, or both. North Java coastal area such as Jakarta Bay is the polluted marine area in Indonesia as a result from anthropogenic wastes and the oil spill. Although evaporation and photo-oxidation play an important role in oil detoxification, ultimate and complete degradation is accomplished mainly by marine micro flora, and being dominant in this function. Certain bacteria are well-known could consume and degrade the PAHs as well as crude oil. Therefore investigating the potential PAH and oil degrading marine bacteria is important. In this study, we collected sample from oil polluted area in Marina Port Ancol, Jakarta Bay and isolated four PAH substrates and Arabian crude oil degrading marine bacteria using enrichment method and direct isolation method. As result, 223 strains could degrade PAHs, among these strains, 94 strains could degrade phenanthrene, 23 strains degrade fluoranthene, 92 strains could degrade dibenzotiophen, 14 strains could degrade phenotiazin and 106 isolates degrade crude oil.Key words: polycyclic aromatic hydrocarbons, crude oil, degrading bacteria, bioremediation.

Abstract

Trichoderma is a well -known candidate to be promoted as cellulase producer  for the hidrolysis of lignocelluloses that contain  in  woody  biomass. The  number of trichodermal  isolates  in our  laboratory collected  from diverse ecosystem types in Indonesia increases significantly during the last 5 years. It is our aim to assess the cultures for its ability in producing cellulase. Sixty-six trichodermal isolates used in this experiment are obtained from Biotechnology Culture Collection (BTCC), Research Center for Biotechnology, Indonesian Institute of Sciences(LIPI)  The 31 isolates were isolated from District of Liwa (South Sumatra, Indonesia) and the 35 isolates from District of Maros (South Sulawesi, Indonesia). The  isolates were screened qualitatively,  7  isolates  from  Liwa and 12  isolates from Maros showed cellulolytic activity. From the results of quantitative test, two strains  (ID08-T004 and ID08-T63) showed the higher cellulolytic activity among the selected strains, 133.5 and 133.5 U/ml, respectively.  These  extracellular  enzymes  were  characterized  their  temperature  and  pH  optimum.  The temperature  optimum  for  both  enzymes  was  the  same,  50C,  with  activity  213.6  U/ml  for  enzyme  extracted from ID08-T004 and 197.3 U/ml for enzyme from ID08-T0063. The pH optimum was pH 5 of ID08-T004 with activity137.7 U/ml and pH 6 for ID08-T063 with activity 75.0 U/ml. The enzymes from ID08-T004 and ID08-T063 were stable in their temperature and pH optimal condition even after 90 minutes incubation with activity 179.0 U/ml and 86.7 U/ml, respectively. The enzyme stability was approximately 150 minutes for both enzymes in the temperature and pH optimum.   Key Words: Trichoderma, cellulase, Culture Collection
