Abstract

Rice stem borer (Scirpophaga sp.) is considered as one of the primary pests causing yield decrease in Indonesia. Genetic engineering is considered as one of the most effective way to improve the resistance of rice plants to yellow stem borer since no resistance gene has been found in rice and its wild relatives. A double T-DNA binary vector p2TDNAcryIAb wich carry two independent T-DNA was used to transform Indonesian elite rice cv Ciherang using Agrobacterium-mediated transformation to generate transgenic rice that free from selectable marker genes. One T-DNA containing a selectable marker hygromycin phosphotransferase (hpt) gene and the other carrying the cry1Ab gene. As many as 37 putative transgenic plants from 14 embryos were generated. PCR analysis demonstrated that 22 out of 37 putative transgenic rice contain cry1Ab gene. All Ciherang rice carrying the cry1Ab gene expressing Cry1Ab protein as shown by the immunostrip assay. Further work will be carried out to determine the transgenes copy number, to select marker free transgenic rice, and to evaluate the resistance of transgenic rice against neonates larvae of yellow stem borer.Keyword: double T-DNA, transgenic rice, Cry1Ab, transformation

Abstract

Cationic lipid is one of transfection agents which show high efficiency and low cytotoxicity. The transfection efficiencies may depend upon the type or amount of cationic lipids, the cell line or DNA plasmid being used for transfection. The purpose of this study was to find optimal condition for transfection of CHO-K1 and CHO-S cells with pJ-EPO plasmid (contain human epo gene) compared with pEGFP-c1 plasmid (contain gfp gene) by cationic lipid Lipofectamin 2000TM to generate stable transfectant expressing recombinant human erythropoietin (rhEPO). Optimization was carried out regarding the amount of lipofectamin, DNA concentration, and concentration of antibiotic Geneticin (G418) for selection of stable transfectants. Using standard amount of lipofectamin (10µl/well) in 6-well plate, highest expression level of green fluorescent protein (GFP) was shown after transfection of CHO-K1 cells with 4µg/well pEGFP-c1 while highest expression level of rhEPO was observed after transfection of CHO-K1 cells with 6, 8, and 10µg/well pJ-EPO plasmids. The data also indicated that optimal transfection conditions of CHO-K1 and CHO-S cells with pJ-EPO were shown with the use of 4µg/well DNA and 15µl lipofectamin, respectively. Concentration of G418 affected the expression where strongest rhEPO expression was shown at 1,000 ng/µl G418 concentration.

Abstract

The aims of this study were to identify the effects polymorphism of β-lactoglobulin gene  on milk yield and percentage of milk protein  and  fat  in  local sheep. A total number of 83  heads of  lactating ewes  raised under an extensive management at  Jonggol Animal Science Teaching and Research Unit (JASTRU) of  the Faculty of Anima Science, Bogor Agricultural  University were  studied.  Research  activities  were  carried  out  through  some  step involving blood collection. DNA isolation, DNA amplification and separation of DNA fragments by electrophoresis with silver staining method. By using polymerase chain reaction (P  R) then genotyped by single strand  conformation polymorphism technique (SSCP), it was success fully amplified a fragment length at 420 bp in β-lactoglobulin gene located on  the exon 7. The electrophoresis  pattern  revealed 5 types of β-lactoglobulin gene and designated as AB. C, D and E types. The proportion for  those respective 5 type from the  highest to  the lowest were for A (27.7%)C (16.87%), D (12.05%), (10.84%) and B (9.64%)  respectively. There were no  significant effect  polymorphism β-lactoglobulin gene on both milk yield and quality  in  I  cal  sheep at JASTRU.   Keywords : polymorphism. β-lactoglobulin in gene, PCR-SSCP and local sheep

Abstract

The objective of study was to produce amylase, cellulase and phytase on sargassum and rice bran on solid state fermentation using Aspergillus niger, Neurospora crassa and Rhizophus oryzae. Media for solid state fermentations composed of dried sargassum and rice bran. The effect of particle of sargassum, initial moisture content on phytase, amilase, and cellulase were evaluated. Optimum enzyme activity of phytase, amylase and cellulase were obtained after 4 days fermentation at 30°C, and initial moisture content was adjusted to 60%. The optimum particle size of dried sargassum attaining the highest enzyme activity was 25 mesh. Best formula forenzymes production was at the ratio of 4:6 (w/w) of Sargassum spinosum (SS) and rice bran (RB) respectively. At this formula highest phytase activity was obtained by Aspergillus niger, cellulase by Rhizopus oryzae, and amylase by Neurospora crassa. Media composed of sargassum and rice bran can be used for phytase, amylase and cellulase production.

Abstract

The protocol for  Agrobacterium-mediated transformation of local banana plants cv  “Mas Lampung” (AA) has been established. A selectable marker gene (hpt) has been used to study the transformation using  in vitro corm slices as target tissues.  Banana  in vitro corm slices were co-cultivated with the EHA105 strain of Agrobacterium tumefaciens harbouring binary vector pCAMBIA 1301 containing hygromycin resistance gene (hpt) as a selectable marker and intron-containing β-Glucuronidase (gus-intron) gene as a reporter gene driven by  CaMV 35S promoter.   Polymerase Chain Reaction (PCR) were used to examine the existence of  hpt gene in plants resulted from the transformation.  Using primer pairs specific for hpt gene, our PCR analysis on leaves showed the presence of the  hpt   transgene in banana transgenic plants at first generation (T0) of transformation.  To prove the existence of hpt gene in the fruits of transgenic banana plants, PCR analysis were also carried out.  The data showed that the  hpt  gene could be amplified  from banana fruits of tested samples.   These result demonstrates that the Agrobacterium-mediated transformation method used in this experiment has been successful to transfer gene into banana plants. Thus, the transformation method reported here could be used as a standard protocol to  transfer another useful genes into local banana plants cv. “Mas Lampung”.  Furthermore, the presence of transgene in fruits of banana transgenic plants is important achievement  especially for transgene that is expected to be expressed in the fruit including to introduce vaccine genes into banana fruits for edible vaccine.Key words:  Agrobacterium,  hpt  gene, transgenic banana Mas Lampung, Musa acuminate corm slices, PCR
