Abstract

Morphology and molecular characterization of microalgae isolated from Wakatobi Marine National Park was conducted. An understanding of the characteristics of morphology, molecular, as well as metabolites profile of the microalgae species is potentially useful for its future application. The primary aim of this study was to isolate, identify and characterize the microalgae strain isolated from Wakatobi Marine National Park labeled as LIPI13-WKT066 with the emphasis on the evaluation of amino acid content as a basis for its health application. Morphological observation under the microscope and molecular identification suggested that the microalgae strain of LIPI13-WKT066 belong to the strain under species of Chlorella volutis. Metabolite characterization of the microalgae strain showed that the content of protein (11.9%), lipid (12.4%) and carbohydrate (4.7%) was in the regular range. Further analysis of its amino acid content revealed the potency of the microalgae strain to be used as antihypertensive agent.

Abstract

Pasteurized milks spoiled at refrigerated storage due to growth of psychrotrophic bacteria. Honey which contain antibacterial and aromatic compounds may be used as supplement to inhibit psychrotrophic bacteria activitiy . To know nutritional and flavor compounds of milks with and without honey, effect of honey on bacterial growth0protein degradation. amino acids and volatile compounds of stored milk were detected. Bacterial growth,protein degradation, amino acids and navor compounds were detected by total plate counts. formol titration,HPLC and GCMS, respectively. The results show that bacterial growth and protein degradations in honey milks were lower than that without honey. Bacterial growth (5.2x10^3 - 9,3x10^6 cfu/mL) and protein degradation (2,37-2.59%) in honey whole milks were lower than that (6.2x10^4 - 6.5x10^7 cfu/mL)(2.54-2.88%) in skim milks,respectively (P<0,05). At 10 days after use by date, changing between amino acids' contents in whole milks withand without honey were more significant than that or skim milk (P<0.05);. and volatile compounds' percentages in honey whole milks were higher than that without honey. while that in honey skim milks vice versa. Honey caused decreasing bacterial growth and protein degradation, changing aminoi acid contents and producing volatile compounds of stored milk, and honey whole milk were better than honey skim milks.

Abstract

The genus Penicillium has a potential ability to produce β-glucosidase. The aim of the study was to improve the β-glucosidase activity of Penicillium sp. ID10-T065 with physical (Ultraviolet = UV), chemical (Ethyl Methyl Sulfonate = EMS), and combined mutation (UV-EMS). The spores of Penicillium sp. ID10-T065 were exposed into UV irradiation for 3 minutes with dose of 0.1 J/cm2 and 13 cm of distances. Chemical mutation was done by treated spores into 3% of EMS solution for an hour. Combined mutation of UV and EMS were also performed by UV for 3 minutes (0.1 J/cm2, 15 cm) and continued with soaking into 2-3% of EMS solution. The developed mutants were screened, selected and assayed. Comparison of enzyme activities with the wild- type (1.78 U/ml), mutant UV13 (5.53 U/ml) showed a 3.1 fold increase; mutant EM31 (4.26 U/ml) showed a 2.4 fold increase. Meanwhile, mutant UM23 obtained from the multiple exposures showed a decreased activity (1.75 U/ml). Mutant UV13 showed the best enzyme activity to be considered as a potential strain for β-glucosidase producer. This result needs to be further elaborated especially on its genetic stability studies in order for the ascertained as a stable mutant.

Abstract

In this research,  construction  of expression  vector  for thermostable α -L-arabinofuranosidase  in Saccharomyces cerevisiae was conducted. BJ1824 was conducted The  Escherichia coli/S. cerevisiae  shuttle vector, pYES2 was  used  as  parental  vector  in  construction.  The  abfA  gene  encoding  α-L- arabinofuranosidase  from Geobacillus  thermoleovorans  IT-08  was  amplified  by  PCR,  in  which  the  plasmid  pTP510 was  used  as  a template. The amplification product was treated with  SacI and XhoI and then subcloned to the pYES2 vector, which was previously digested with  SacI and  XhoI. The recombinant plasmid was designated as pY-Af and propagated  first  in  E.  coli  Top 10,  and  then  transformed  into  S.  cerevisiae  BJ1824.  For  α- Larabinofuranosidase (AbfA) production, the yeast transformants were grown in YNBG selective medium and YPG rich medium, using galactose as an inducer. The AbfA activity was assayed by measuring the amount of p-nitrophenol (pNP) released  from  p-nitrophenyl-α-L-arabinofuranoside  (pNPA) substrate at pH 6.0 and 70 C  for  30  min.  The  recombinant  AbfA  activity  was  detected  in  either  of  culture  medium  (0.98%),  cellassociated (14.17%) and intracellular (84.85%) when recombinant yeast was grown in YPG rich medium.Key words: α-L-arabinofuranosidase; Saccharomyces cerevisiae; expression vector
