Abstract

A  cry1B synthetic gene of Bacillus thuringiensis has been used  for  the transformation of the javanica rice plants  cv.  Rojolele  confer resistance  to  an  important  pest,  yellow  stem  borer  (Scirpophaga  incertulas). Embryogenic callus were co-cultivated with  the EHA 105 strain of Agrobacterium tumefaciens harbouring binary vector CAMB  IA 130 I containing cry  / B gene under  the control of wound inducible gene promoter  (mpi), hygromycin resistance gene (hpt)  as a selectable marker and intron-containing β-glucuronidase (gus - intron) gene al) a reporter gene  driven  by  CaMV35S  promoter.  Previously.  our  histochemical  assay  and  P  R  analyses.  had  proved  th integration of  cry1B gene  into  the genome of rice plants at first generation .  However,  the existence of the gene should remain  table  throughout generation.  In  this study,  the presence of the cry1B transgene in  rice  transgenic plants at  second generation was confirmed by Polymerase Chain Reaction (PCR) .  Insertion of  the cry1B gene in the genome of PCR positive plants was  verified by Southern blot analysis is and showed that integration of cry1  B  into the genomic DNA of  javanica rice plants cv. Rojolele.  An effective resistance of transgenic  plants against  stem borer was verified in  bioassays.

Abstract

An ideal therapeutic for cancer would be one that selectively targets to tumor cells, is nontoxic to normal cells, and that could be systemically delivered, thereby reaching metastases as well as primary tumor. Immunoliposomes directed by monoclonal antibody or its fragments are promising vehicles for tumor targeted drug delivery. Transferrin receptors (TfR) levels are elevated in various types of cancer cells and considered to correlate with the aggressive or proliferative ability of tumor cells. Therefore, TfR levels can be elaborated as a prognostic tumor marker, and TfR is a potential target for drug delivery in the therapy of malignant cells. Here, we report the preparation of an anti-TfR single-chain antibody variable (scFv) immunoliposome for tumortargeted delivery vehicle. The cDNA encoding the variable heavy and light chain domains of the anti-TfRscFv antibody fragment was derived from the murine monoclonal antibody Clone E6, which is specific towards transferrin receptor. The gene encoding the anti-TfR scFv fragment was codon optimized for expression inEscherichia coli, subsequently synthesized, and cloned into the expression vector pJexpress404. The His6- tagged anti-TfR scFv fragment was expressed in E. coli and purified by means of immobilized metal-ion  affinity chromatography on TALON™ matrix. SDS-PAGE revealed that the scFv fragment had the size of approximately 27 kDa, which corresponded with the predicted size of the protein based on its amino acid sequence. Liposome containing 5% MPB-DOPE were prepared by ethanol injection method. Afterwards, the anti-TfR scFv fragments were covalently conjugated to the liposome to produce the anti-TfR scFv immunoliposome with the size of around 200 to 300 nm.

Abstract

Gebang is an Indonesian local genotype which has been selected as superior genotype for low amylose  cassava. Prior to induction of new mutants of this genotype, series of research have been conducted to improve regeneration and acclimatization as well as shoot cuttings production of irradiated in vitro shoots. Four dosage treatments of gamma ray irradiation i.e. those of at 0; 0.2; 1.0; 1.5; 2.0 krad were applied to 32 in vitro shoots multiplied from apical shoots of plants in the field. The highest multiplication rate and acclimatized shoots were obtained from shoots irradiated with 0.2 krad multiplied on various level of BAP added on MS medium and resulted that MS supplemented with 1 mg/L BAP was the best medium. The phenotypic variation was observed in shoots irradiated with 1.0; 1.5 and 2.0 krad while the ones irradiated with 0.2 krad performed normal appearances. Subsequent production of propagated young stem cuttings called “ratooning system” decreased after the second cycle of propagation especially in the survival rate of the ones irradiated with 0.2 krad. The findings lead to the opportunity to produce cassava propagules derived from irradiated in vitro culture at a higher amount using Gebang genotype as a model.Keywords: cassava Gebang, irradiated shoot, young stem cutting/ratooning , acclimatization, in vitro.
