Abstract

Isolation and determination of optimum lipase conditions from Avocado seeds germination (Persea americana Mill) have been done. The purpose of this study is to determine the optimum working conditions and lipase specific activity of avocado seeds germination. Lipase was isolated using homogenization and centrifugation techniques. The crude extract of the enzyme containing the lipase produced from avocado seeds germination was used for the determination of total protein concentration and tested its activity on the pH conditions, temperature, and optimum substrate concentration with different with various variations. Determination of total concentration was done by Bradford method, while the determination of optimum working conditions and lipase specific activity was performed by titrimetric method. The results showed that total protein concentration in avocado seeds germination was 458 μg / mL. The optimum working conditions on avocado seed sprouts are the degree of acidity (pH) 6, temperature 35 oC, and 1.5% v/v substrate concentration. The lipase specific activity of avocado seeds germination is 144,1 U / mg.

Abstract

The research about activation of natural zeolite with variation concentration HCl for adsorption reactive yellow 86 from waste liquid woven Samarinda has been conducted. The activation of zeolite was prepared with two way, by mechanicalstirring and ultrasonic assisted. From the X-Ray Diffraction (XRD) result showed that natural zeolite activated with ultrasonic (NZA-U) has 38 % composition of SiO2, while natural zeolite activated with mechanical stirring (NZA-MS) is 21 %. Surface area analysis using methylene blue shows for NZA-U is 25 m2/g and 24 m2/g for NZA-MS. The results on adsorption of reactive yellow 86 from waste liquid woven Samarinda using spectrophotometer visible on wavelength 417 nm shows the optimum condition adsorption on time contact60 minutes and concentration HCl 3 M, with percent of adsorption is 56.14 % for NZA-U and 53.93 % for NZA-MS

Abstract

Research synthesis esteramin with cattapa seed oils (Terminalia cattapa) has been done through a transesterification reaction. The synthesis process was conducted on the extraction, esterification and transesterification. The results showed cattapa seed oil content of 51.9%, the acid value of 7.559 mg KOH / g and ALB levels of 3.4496%. The composition of the oil determined using GC-MS. GC-MS analysis of the composition of the most dominant oil is oleic acid amounted to 34.86%. Esterification process in the formation of methyl esters can be known ALB decreased to 0.7%. Methyl esters are reacted with triethanolamine through a transesterification reaction. The results showed esteramin water surface tension value of 34.947 dyne / cm, the HLB value of 5.25 is included in the water in oil emulsifiers scale and on a TLC plate there are two stains.

Abstract

The screening of amylase enzyme producing thermophilic bacteria from sediment of hot spring in Muara Jawa has been done. Screening has done by using selective media of starch, will show the formation of clear zones around the colonies by dripping the iodine solution. From alot colonies selected 1 colony with widest clear zone. Single colony obtained was identified as Bacillus sp. Amylase produced was tested for amylase activity by DNS method and obtained the amylase activity at pH 6 and temperature at 55°C is 2,8460 U/mL.

Abstract

Studi Docking Molekular senyawa Kuersetin, Kalkon dan turunannya sebagai inhibitor sel kanker payudara MCF-7 (Michigan Cancer Foundation-7) telah dilakukan . Hasil yang didapatkan bahwa energi docking setelah proses penambatan pada mode 1 dari ke empat senyawa uji yaitu Kuersetin -7.4 Kkal/mol, 2-(3,4- dihidroksifenil)-5,7-dihidroksi-3-metoksi-3,4-dihidro-2H-1-benzopyran-4-on -6.8 Kkal/mol, Kalkon -6.9 Kkal/mol dan 3-(2,4-dihidroksifenil)-1-fenilpropan-1-on -6.6 Kkal/mol. Kuersetin membentuk ikatan hidrogen dengan asam amino Asam Aspartat 61, Arginin 65, Arginin 68, Fenilalanin 71 dan Serin 75. 2-(3,4- dihidroksifenil)-5,7-dihidroksi-3-metoksi-3,4-dihidro-2H-1-benzopyran-4-on membentuk ikatan hidrogen dengan asam amino Alanin 59, Aparagin 102 dan Arginin 105. Kalkon membentuk ikatan hidrogen dengan asam amino Asam Glutamat 94. Dan 3-(2,4-dihidroksifenil)-1-fenilpropan-1-on membentuk ikatan hidrogen dengan asam amino Tirosin 67. Kuersetin dinilai lebih berpotensi sebagai kandidat obat antikanker dibandingkan 2-(3,4- dihidroksifenil)-5,7-dihidroksi-3-metoksi-3,4-dihidro-2H-1-benzopyran-4-on, chalcone and 3-(2,4-dihidroksifenil)-1-fenilpropan-1-on. Abstract

This study demonstates the potential of starch tuber ganyong (Canna edulis Kerr.) as a substance for making ethanol by Saccharomyces cerevisiae with microalgae addition of Spirulina sp. as a source of nutrients in the fermentation process of ethanol has been carried out. The study also aims to determine the amount of nutrient concentration and fermentation time was used to generate the optimum ethanol content. Hydrolysis process by enzymatic is done through a liquification phase with α-amylase and saccharification phase with gluko-amylase. Furthermore the process of fermentation stage with the variation of the time (132 hours, 156 hours, 180 hours and 204 hours) using the yeast Saccharomyces cerevisiae with the addition of Spirulina sp. as a nutrient in the variation of Spirulina sp. concentrations (0,6 % (w/v), 1,2 % (w/v) and 1,8 % (w/v)). The result showed the sugar reducing after of hydrolysis was obtained with amount of 0,527 %, increased from the initial state with amount of 0,351 %. The highest ethanol content be found on the increase nutrient with amount of 1,2 % (w/v) by is time of fermentation to180 hours.

Abstract

The research on the use of water hyacinth powder (Eichornia crassipes) activated by adsorption method as the adsorption of cadmium metal ions (Cd) using dye bag system had been done. Water hyacinth powder was activated previously using HNO3 0,3 M then characterized by FT-IR spectroscopy. Infra-red spectrum showed hydroxyl cluster (-OH) from cellulose could function as the active sites of metal adsorption. Adsorption process was done by using variant contiguous time periods of metal ions, variant concentration of metal ions and addition of the metal ions of lead (Pb) ion adsorption on metal cadmium (Cd). From the results of the study Atomic Absorption Spectrophotometer (AAS) had shown the percentage of adsorption by water hyacinth powder adsorbent which was activated at 30 minutes of optimum contiguous time with optimum concentration 100 mg/L of 57,175 mg/L and 56,441% percentage of adsorption. In addition of the metal ions of lead (Pb) ion adsorption on metal cadmium (Cd) decreased to 20,681 mg/L with the percent adsorbed at 20,415% . Keywords: , , , , , AAS.

Abstract

Synthesis of ascorbyl palmitate from methyl palmitate and acetyl ascorbate through interesterification reaction has been done. Modified were done to improve the nature of ascorbic acid that only soluble in water (lipophobic) become soluble in oil or fat (lipophylic). The interesterification reaction conducted with molar ratio 1:4 (methyl palmitate:acetyl ascorbate), lipase from coconut as a catalyst with amount 70 % from its substrat, pH 7, the temperature reaction 40-50 °C and the reaction time is 48 hours so the yield produce is 46,7241 %. Characterization of the functional group were confirmed with FT-IR. Analysis of antioxidant activity using radical reduction method 2,2-diphenyl-1-picrylhidrazyl (DPPH) result the antioxidant activity value of ascorbyl palmitate is 45,41 ppm. The result showing that ascorbyl palmitate synthesized is have a potency as an active antioxidant.

Abstract

Research on the synthesis of activated carbon from jengkol shells (Pithecellobium lobatum) with the help of ultrasonic waves as an electrochemical energy storage material has been done. The activated carbon from jengkol shells is characterized by surface area using blue methylene method and pore morphology with SEM (Scanning Electron Microscope) instrument. The characterization results show that the active carbon with ultrasonic wave exposure has a larger surface area of 181.3693 m2/gram than the activated carbon without exposure to ultarsonic waves of 178.0290 m2/gram. The SEM results in activated carbon with ultasonic wave exposure indicate that the pore distribution is more evenly distributed, the number of pores more, and the pore size tends to be smaller, whereas in the activated carbon without the exposure of larger and slightly larger portions of ultrasound waves. The optimum value of capacitance on activated carbon without and with ultrasonic wave exposure of and 6.97 μF/gram and 5.21 μF/gram.

Abstract

Spent bleaching earth (SBE) as adsorbent of Pb2+ ion, through oil extraction process and continued by acid activation has been studied, this process assisted by ultrasonic wave in 30, 60 and 90 minutes reaction time. Based on the results analysis of X-Ray Flourescence (XRF), the reaction time of 30, 60 and 90 minutes showed an increase of SiO2 were 37.802 %, 42.374 % and 38,793 %. From the surface area with method methylene blue adsorption showed the surface area of DSBE-A 30, 60 and 90 were 34.3057 m2/g, 36.3375 m2/g and 35.8893 m2/g, respectively. XRD analysis showed the type of DSBE-A 60 was rectorite mineral with chemical compound was Na.Al4(Si,Al)8.O20.(OH)4.H2O and has a crystallinity 53.93%. Optimum conditions of DBSE-A 60 to adsorb Pb2+ ion in contact time was at 80 minutes, pH 3 and 20 ppm of Pb2+ ion concentration with adsorbtion efficiency up to 46.04 % and the optimum capacity of DSBE-A 60 to adsorb Pb2+ ion was 0.9209 mg/g.
