Abstract

Amplification of pncA genes from water bacterial post mining in Samarinda has been done. The aim of this research is to isolate chromosomal DNA and amplificated the pncA gene from water bacterial post mining in Samarinda. The research methode consist of chromosomal DNA isolation and amplification of pncA gene with polymerase chain reaction (PCR) technique. The result showed the size of chromosomal DNA is ± 10 kb. By using primer pncA gene, the result of amplification of the pncA gene showed that there were two sizes of amplicon, namely 0.7 kb and 0.2 kb. The conclusion that can be drawn was there is two sizes of amplicon pncA gene. Keywords: chromosomal DNA isolation, amplification of pncA gen, PCR

Abstract

Singkil (Premna cordifolia Roxb.) is a plant that is widely found un Kalimantan. The people of Kalimantan are often used singkil leaf by as traditional medicine are believed to have medicinal properties as a cure for intestinal worms, colds, and antidiabetic. In this research, it used phytochemical screening and inhibition activity amylase test on singkil’s methanol extract. The objective of this study was to determine in-vitro enzyme amilase inhibition activity against sampel by DNS method. The result of this research was phytochemical screening in singkil’s methanol extract contains alkaloids, flavonoids, saponins and phenolics. Singkil leaf methanol extract at a concentration of 10 µg/mL can inhibit the activity of the amylase enzyme with an inhibition value of 60,93 %. Acarbose used as a positive control in this study showed activity against the amylase of 44,79 %. Amylase inhibition activity of methanol extract was higher than acarbose, the singkil leaf methanol extract has the potential as an antidiabetic drug. Abstract

Pucuk Merah (Syzygium myrtifolium Walp.) is an ornamental plant that has benefits as an antioxidant, antibacterial, antiplatelet, antiulcerative, antimalarial, antiviral, antihyperglycemic and anti-inflammatory. This research aims to determine the optimum time for the production of secondary metabolites of pucuk merah endophytic bacteria for amylase and protease inhibition. Production of secondary metabolites was carried out at various times of 24 hours, 48 ​​hours and 72 hours. The method for the amylase inhibition test used the DNS method and the protease inhibition test used the Bradford method. The results showed that the optimum time for the production of secondary metabolites was 48 hours. Keywords: Syzygium myrtifolium Walp., Endophyte Bacteria, Production Time.

Abstract

One of the causes of acne is infection by the bacteria Propionibacterium acnes. Singkil (Premna cordifolia Roxb.) is a kind of herb plant from Kutai Kartanegara which is used as a traditional herb including for acne’s treatment. This study aims to know the content of secondary metabolites compound in singkil’s extract and determine the antibacterial activity against Propionibacterium acnes KCCM 41747 bacteria. In this study, it’s phytochemical screening and antibacterial activity test using the disc diffusion method on methanol extract, n-hexane, ethyl acetate and water-methanol fractions. the result of phytochemical screening revealed that methanol extract of singkil contains alkaloids, flavonoids, steroids, and saponins. The n-hexane fraction contains steroids and the ethyl acetate fraction contains alkaloids, flavonoids, and phenolics while the water-methanol fraction contains alkaloids, flavonoids, and phenolics. The antibacterial activity test extract with a concentration of 10% showed that the diameter of clear zone of the metanol extract, n-hexane fraction, ethyl acetate fraction, and water-methanol fraction were 13 mm, 9 mm, 13,67 mm and 6 mm, respectively. The ethyl acetate fraction had the highest antibacterial activity among the four extracts. A minimum inhibitory concentration value of the ethyl acetate fraction against Propionibacterium acnes KCCM 41747 was 0,625%. The ethyl acetate fraction have the potential as an antibacterial against the bacteria Propionibacterium acnes KCCM 41747. Abstract

Addition of metal ions on lipase activity has been done. The aim of this research is to find out the effect of metal ions on lipase activity from water bacterial post mining in Samarinda. Lipase activity was determined based on the Titrimetric method with olive oil as a substrate. The result showed lipase activity from water bacterial post mining without the addition of metal ions is 1,0 U/mL and with the addition of metal ions Mg2+, K+, Fe3+, Ca2+, dan Ba2+their respective lipase activities is 0,6 U/mL, 0,7 U/mL, 1,9 U/mL, 1,4 U/mL dan 0,8 U/mL. The conclusion that can be drawn that is metal ions K+, Mg2+ and Ba2+ inhibit lipase activity with a relative activity each at 70; 60 dan 80% while Ca2+ increase lipase activity with a relative activity at 140% and Fe3+ causes denatured lipase with a relative activity at 190%. Abstract

Phytochemical test and antibacterial activity of methanol extract of Mentawa leaf (Articarpus anisophyllus Mig.) have been done. The extraction of Mentawa leaf samples was carried out by the maceration method using methanol as a solvent. An antibacterial activity test was carried out using the agar diffusion method. Phytochemical tests results show that methanol extract of Mentawa leaf has secondary metabolites of flavonoid, steroid, triterpenoid, phenolic, and quinone compounds. The antibacterial activity test results obtained MIC values for Salmonella typhi bacteria were 1.25-2.5%, Streptococcus mutans bacteria were 0-0,625%, Streptococcus sobrinus bacteria were 1.25-2.5% and for Propionibacterium acnes bacteria were 0.625-1.25%. Methanol extract of Mentawa leaf has broad-spectrum antibacterial activity.

Abstract

Endophytic bacteria is one of the source of extracellular enzymes producer. Enzymes that produced by endophytic bacteria are more beneficial because they can be cultured in small spaces with big quantities, relatively fast time and cheaper production costs compared to the sources of enzyme from plants and animals. This study was conducted to obtain amylase, lipase and protease from endophytic bacteria of ciplukan leaves (Physalis angulata L.). The research method used is experimental, by isolating endophytic bacteria from ciplukan leaves (Physalis angulata L.) in Nutrient Agar (NA) media and screening tests of extracellular enzyme activity from endophytic bacteria isolates. The isolation results was obtained 24 endophytic bacteria isolates that 4 bacteria isolates produce amylase, 3 bacteria isolates produce lipase and 2 bacteria produce protease.

Abstract

Plants at this time are widely used as medicine because according to some research medicinal materials, plants that have the potential as medicine do not cause too many side effects. One of the plants that are used as medicine is agarwood leaves because it is known to have several medicinal properties, one of them is as an antibacterial drug. This study was conducted to find out the secondary metabolite compounds contained in agarwood leaves and to determine the MIC value of agarwood leaves methanol extract against Streptococcus sobrinus KCCM 11898 and Salmonella typhi ATCC 422, by means of agarwood leaves extracted using methanol, phytochemical screening, and antibacterial activity tests using disc diffusion method. Phytochemical screening of agarwood leaves methanol extract results revealed alkaloids, steroids and phenolics. The MIC value of agarwood leaves methanol extract toward Streptococcus sobrinus KCCM 11898 and Salmonella typhi ATCC 422 is 1.25%.

Abstract

Meniran (Phyllanthus niruri L.) is one of the herb plants used as traditional medicine, such as toothache medicine and diarrhea medicine. The purposed of this study was to determined the type of secondary metabolites in the ethanol extract of meniran herb and to determined the minimum inhibitory concentration (MIC) value of the ethanol extract of meniran (Phyllanthus niruri L.) to Streptococcus sobrinus KCCM 11898 and Salmonella typhi ATCC 422. The ethanol extract of meniran was tested for phytochemicals. The MIC value of the ethanol extract of meniran herb for the test bacteria was obtained by varying the concentration of the ethanol extract of meniran herbs. Phytochemical test results showed that the ethanol extract of meniran herbs contains alkaloids, phenolics and steroids. The minimum inhibitory concentration value of the ethanol extract of meniran herb at the bacteria Streptococcus sobrinus KCCM 11898 and Salmonella typhi ATCC 422 each was 0.625%. Abstract

Endophytic bacteria are microorganisms that throughout or part of their life cycle inside plants and will not appear certain symptoms on the plant itself. Endophytic bacteria can produce enzymes. Enzymes produced by bacterial endophytes are more beneficial because the production time is faster. One of the plants that contain bacteria is Pucuk Merah (Syzygium myrtifolium). This study aims to isolate endophytic bacteria from leaves of Pucuk Merah (Syzygium myrtifolium) and to screen these endophytic bacteria as amylase, lipase and protease producers. The results of this study were obtained there are 24 isolates of endophytic bacteria from Pucuk Merah leaves. A total of 19 bacterial isolates from 24 bacterial isolates produce amylase, 1 isolate produce lipase and 24 isolates produce protease.
