Abstract

Peripheral blood sel mononuclear cell (PBMC) is a white blood cell that can be isolated using the gradient centrifugation method. Ficoll is a reactant which can be used in this method because it produce a high viability i.e. 97.1% 1.0%. This research aims to isolate PBMC from healthy human blood. PBMCs result is used for analysis of dengue virus immune response.Dengue viruses infect PBMC, so going on the secretion of cytokine which is one marker of severity of dengue disease. Expression of cytokines can be measured using quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR).Venous blood from healthy human is takenaseptically using anticoagulants heparin, then PBMC is isolated with ficoll gradient centrifugation method to produce buffy coat that contain PBMC. Viability of PBMC is calculated using hemositometer with the help of a microscope.Based on the results obtained by PBMC isolation per mL of venous blood that is 2, 08x106 up to 2, 61x106 with an average of 2, i.e. 31x106 withviabilityof 95-99%. These results indicate that this method is quite effective in isolating PBMC from healthy human blood.Keyword : blood, ficoll, centrifugasigradien, PBMCs

Abstract

The objective of this study was to analyze the effect of snack intervention on student nutritional and anemia status at Pasanggrahan 2 elementary school. Design study was experimental onegroup pretest-postest design and was conducted at Pasanggrahan 2 elementary school in Purwakarta from August to December 2012. The total number of fifty three students were selected purposively. The primery data were collected namely: household social economi and student characteritics, food habbit, nutritional and anemia status. The result of study showed that the energy and protein contribution to average of students nutrition adequate was 17.9% and 23.6% respectively. While the contribution of snack energy and protein to average of daily total nutrition consumption was 26.7% and 31.3% respectively. There was significant difference of student nutritional and anemia status before and after snack intervention as additional school feeding.

Abstract

Lake Cikaret is the largest lake in the village of Cikaret in district of Cibinong, Bogor district which is also a reserve of water for residents. The Lake is thought to have experienced pollution due to the inclusion of various types of waste from households and activities waste disposal by visitors. Research done by the method of field survey, the research is divided into 3 point i.e. inlet, middle and outlet. The results showed the temperature 25 oC-38oC, pH values in the range 5-7, the value of dissolved oxygen content ranged between 5.9-14.5, BOD values ranged from 5.6-7.5 mg/liter and CO2 values ranging between 11.3-13.5 mg/l due to the pollution of the natural. Net primary productivity of photosynthesis 3.2 mg/l, gross 4.3 mg/l of photosynthesis and respiration 1.1 mg/l. Plankton found Chlorophyceae, Cyanophyceae and composed Bacillariophyceae. belong to the phytoplankton and zooplankton 5 classes, namely Euglenoidae, Dapniidae, Maxillo-poda, crustaceans and Monogononta. The value of diversity index includes low range 0.05-0,149. Equity index value on Lake Cikaret range 0,009-0,064. Abstract

The breadfruit tree Artocarpus altilis species is a species that is often used fruit as a food source , while the stems and roots of this plant can be used as a medicinal plant. Around the area of Sukabumi, west Java people use the bread fruit tree flowers as a insect repellent. The purpose of this study is to determine the potential of breadfruit tree stamens extracts as larvicides against Aedes aegypti mosquito larvae and determine the optimum concentration of extract of stamens breadfruit tree which can be used effectively as Aedes aegypti mosquito larvicides. Determine larvicidal activity of stamens breadfruit tree (Artocarpus altilis) includes : sample preparation and phytochemical test , sextraction , larvicidal test , toxicity testing and data processing . Comparison test was done to compared the activity of the male flower extracts as larvicides with breadfruit tree abate powder against mosquito larvae , as well as the toxicity of the statistical test using the LC50.Phytochemical test results positive breadfruit tree botanicals stamens contain tannins , saponins , and triterpenoids . Water content in the crude drug obtained 12.04 % . Breadfruit tree extract of stamens is not harmful to humans . This study getting the LC50 of 0.25 % at phase n - butanol extract , and 1 % in the water phase extract.Key words : Waste, Breadfruit, Aedes aegypti, Artocarpus altilis

Abstract

Sargassum is one of the genus including in Class Phaeophyceae. Sargassum spp. are found as many as 150 species in zone waters tropic, subtropic and frigid zone. This research to know variation morphology and anatomy Sargassum spp. which found in Bayah Beach. Result of the research will giving knowledge for different Sargassum species based on variation morphology and anatomy. Research methods divided of three work phases that are collect in the field, morphology observation and anatomy observation. Collect alga in the field with execute transek method, morphology observation with differents variation formed from cauloid, branching type, filoid, cryptostomata, midrib (middle vein filoid), vesicle (float) dan receptacle (reproduction organ). Anatomy observation with measure length and wide cells, analize cells shape, counting cells amount (enlargement 160 x), from each parts cortex and medulla by cauloid and filoid Sargassum spp. Based observation results are found 7 species Sargassum that are Sargassum binderi, S. cinereum, S. echinocarpum, S. duplicatum, S. plagyophyllum, S. crassifolium dan Sargassum sp. which have in Bayah Beach, South Banten. Abstract

This event will be based on research by the large number of reports on the benefits of soursop for various diseases especially antihipertensi and antihiperuresemia. Forms need to be developed so that a more practical preparations and ketersediannya continuous, one of which is in the form of effervescen granule juice. This research aims to find out whether the soursop fruit juice can be a formulation in granule material of efervesen who meet the requirements of quality, standards and terms of efervesen granule and making it acceptable in society. In the study of fruit juice made with the soursop fruit flesh filter using Batiste, juice drained using Freezedryer with the addition of maltodekstrin 20%. The powder is then made 3 fruit formula granule effervesent with the difference of acid-catalyzed and the manufacturing process is done by the method of smelting using alcohol 70% without binder. Test results show the BSLT fruit pollen have toxic effects against larvae of shrimp Artemiasalina Leach with LC50 108.914 ppm. The results of the evaluation of the third shows the formula for granule corner quietly and solubility are eligible, cohesive, and the granule flow values test froth above 70%, the test results of licentious formula2 has the most preferred taste. Stability test for efervesen granule 8 weeks the most stable at a temperature of 15 C containing Sodium ion levels 3, 97g/100 ggranul, potassium of 0, 24g/100 ggranul, 2, 19mg SAG polyphenols/ggranul and vitamins C51, 91mg/100ggranul.Key words : granul, efervesen, stabilitas, sirsak

Abstract

In the manufacture of jelly milk drinks are necessary also other additional ingredients such as sugar is used as a sweetener in the manufacture of jelly while working to ease hydrocolloid dispersion. Potassium citrate is used for auxiliary materials, especially the formation of gel texture carrageenan, citric acid is used as a substance other than flavor in soft drinks, used also as an inhibitor of microbial growth in milk jelly drinks.An analysis of milk jelly beverage products, including the organoleptic analysis of where the testing is done against the panelists to determine the level of liking for the product. Viscosity to determine the viscosity of the product before the final product, the analysis is conducted to determine the strength of the gel on which the final product gel strength will be measured by looking at the gel fracture when pressed with a certain weight. PH measurement is necessary to know the level of acidity flavor obtained from addition of citric acid and then measuring brix to see the level of sweetness of the product because of sweetener used is pure sugar. In this study the analysis of protein levels because the products contain milk which is produced by converting nitrogen compounds into ammonium sulfate and then elaborated by NaOH, whereas the liberated ammonia was tied with H3BO4 and then titrated with HClKey words: Gels, hydrocolloid, sugar, potassium citrate, citric acid, organoleptic, viscosity, gelstrength, pH, brix, protein levels

Abstract

Perubahan iklim menjadi kajian yang penting dan sangat menarik pada beberapa tahun terakhir, karena mengakibatkan kenaikan suhu udara dan kenaikan muka air laut. Tujuan makalah ini adalah untuk mengkaji model dan skenario iklim yang sesuai diterapkan sehingga dapat menggambarkan adanya perubahan iklim khususnya wilayah lokal dan mengembangkan analisis dampak kenaikan muka air laut di kawasan pesisir. Dalam Makalah ini pemodelan iklim menggunakan perangkat lunak MAGICC/SCENGEN 5.3(Model sirkulasi global CSIRO, dengan skenario A2-AIM dan B2-AIM) sedangkan kenaikan muka laut dengan memanfaatkan data Data Sea Level Anomaly dari satelit Altimeter Jason 1A. Hasil pemodelan proyeksi perubahan suhu rata-rata dan perubahan presipitasi bulanan menunjukkan pola yang teratur meningkat secara linier dan Berdasarkan trend kenaikan nilai SSHA dari satelit altimetri Jason-1 yang diekstraksi dari hasil interpolasi lintasan satelit diperoleh rata-rata kenaikan muka laut relatif selama 7 tahun (2002-2009) sebesar 11,1 mm/tahun. Skenario yang sesuai dengan kondisi daerah pesisir Kabupaten Tuban adalah skenario B2AIM. Hasil pemodelan MAGICC sesuai dengan skenario B2AIM menunjukkan laju rata-rata kenaikan muka air laut dari tahun 1990-2050 adalah sebesar 0.61 meter. Hasil pemodelan SCENGEN sesuai dengan skenario B2AIM, menunjukkan perubahan curah hujan yang cukup besar pada bulan Juli, Agustus, September dan Oktober, dan perubahan kenaikan temperatur hasil pemodelan SCENGEN yaitu sebesar 0.19?C 1.56?C pada tahun 2050. Abstract

Poksai horse (Garrulax rufifrons rufifrons) is a bird endemic to the island of Java, the bird is only found in some mountainous regions in West Java. Distribution and number of individuals poksai horses in nature can not be known with certainty, there is still a lot of data that is unknown, such as how it behavior, why is it spreading increasingly narrows, reproductive system and others.One of the Institute of conservation in Sukabumi have started doing this to kind of breeding programs namely Cikananga Conservation Breeding Centre (CCBC), but there are still some obstacles that arise during the breeding program is running. In February until March 2014 conducted research at CCBC, against 2 pairs of poksai horses. Observations using scan sampling method, for 7 hours/day divided into two time, namely at 07.00-12.00 and 13.00-15.00 GMT.Based on the results of the daily behavior of the dominant research on bird poksai is the behavior moves (94,725%), ingestive(3.93%), silent behavior (1,295%). The activity observed in the behavior of Pre-breeding activity is approaching, probe, feeding, talking, making nests, dancing, destroying the hive. From poksai 2 pairs of horses were observed only a pair of eggs and incubate successful son. Abstract

This research aims to determine of river water quality status in the province of Java, including Banten, DKI Jakarta, West Java, Central Java, Yogyakarta and East Java. The data used are secondary data obtained from the ministry of environment and forestry in 2015. In this research, Fuzzy Mamdani method will be used to give solution of river water quality status. Parameters measured and observed are the parameters of Total Suspended Solid (TSS), Dissolved Oxygen (DO), Biochemical Oxygen Demand (BOD), and Chemical Oxygen Demand (COD).Top of Form Based on fuzzy mamdany , Status of river water quality status in DKI Jakarta and West Java have been heavily pollution. Banten province was in mild pollution to moderate pollution. Central Java was in light pollution to heavily pollution. DI Yogyakarta was in mild pollution, and East Java was in mild pollution to moderate pollution.Key words: Fuzzy Mamdani, river water pollution, water quality status
