Abstract

IRF-2 is a member of the interferon regulatory transcription factor (IRF) which encodes interferon regulating factors. The study aimed to describe the character of the Interferon Regulatory Factor 2 (IRF-2) gene sequence from maleo and provide an experimental module about molecular genetic methods. This study used descriptive methods. Genetic data were obtained by using the molecular method through three stages of DNA isolation, DNA amplification, and sequencing. Alignment is done using Clustal X in the MEGA6 program. Phylogeny tree constructed by Neighbor-Joining algorithm and Juke-Cantor evolution's model from program MEGA6. The sample consisted of 0,3 ml blood from two individual maleo and species of Megapode group as a comparison that was obtained from GenBank. The results of the analysis from 612 bp of IRF-2 genes identified with the composition of the nucleotide base of 23.1% T, 20.8% C, 35.1% A and 21.0% G. IRF-2 gene sequence of nucleotide composition was rich in adenine (A). Analysis of IRF-2 gene sequences phylogeny tree topology tree produced good enough and has aggregated species from Megapodiidae family. Moreover, the validation suggested that the practical guide book in this study were ‘eligible’ to be used as learning media.

Abstract

This study aims to improve the students' abilities through the application of an environmentally-based guided inquiry learning model in grade IV SDN INPRES Kabonena and the student learning outcomes in science learning through the application of environmentally-based guided inquiry learning models in grade IV SDN INPRES Kabonena. This type of research is a classroom action research, using a spiral model design of Steven Mc. Taggart, which consists of 4 stages, namely planning, implementing, observing, and reflecting. The location of the research was at SDN INPRES Kabonena, Palu City. The research subjects were 20 students, consisting of 11 male students and 9 female students. The data consists of learning data including data on teacher skills and student activities. Data collection techniques are by observation and tests. Data analysis was performed by qualitative data analysis by concluding based on the mean value or standard deviation of observations (percentage of observations). The results showed that the students' ability in learning through the observation of teacher skills in the first cycle obtained a score of 22 with a good category percentage of 61%, and in the second cycle a score of 29 was obtained with a percentage of 81%, very good criteria. The ability of students in learning based on student activities in teaching and learning activities, in the first cycle a score of 21 was obtained, with a percentage of 58% in the sufficient category, and in the second cycle, it was obtained 27.5 with a percentage of 76.4% in the good category. Meanwhile, the student ability data obtained for pre-action, the number of students who completed as many as 5 students, with classical absorption (DSK) 50% and classical learning completeness (KBK) 25%. The data on student ability in the first cycle obtained 6 students with a DSK of 57% and a CBC of 30%, an increase in the second cycle where the number of students who completed 20 students with 88% DSK and 100% CBC While the data on student learning outcomes in the first cycle obtained 8 students with a DSK of 59% and a CBC of 40%, an increase in cycle II where the number of students who completed was 20 students, with a DSK of 87% and CBC 100%. Based on these results, it can be stated that the application of an environment-based guided inquiry learning model can improve problem-solving abilities and learning outcomes of fourth-grade students of SDN INPRES Kabonena.

Abstract

The study aimed to determine whether there is an influence between the principal leadership on the learning outcomes on the science of the fifth-grade students of SD Tojo Una-Una Regency and to improve performance of teachers in improving learning outcomes on the science of the fifth-grade students of SD Tojo Una-Una Regency. This study used a type of quantitative research. The instruments in this study were a questionnaire sheet. The instrument test used Validity Test and Reliability Test. The data analysis technique used the t-test technique which is preceded by an analysis prerequisite test, namely the normality test and linearity test. P-value results of the normality test on the principal's leadership value (0.28)> 0.05 and student learning outcomes (0.17)> 0.05. thus, the data obtained were normally distributed. Therefore, the principal leadership can influence work motivation and performance of teachers, so that student learning outcomes also increase.

Abstract

This study aimed to describe the conception of junior high school students, high school and students about eclipses and the phases of the moon. Respondents involved in this study were 35 junior high school students, 24 high school students and 38 college students. The instrument used to mendeksripsikan conception as much as 10 numbers. At the first level students are expected to choose the right option right or wrong or memilh existing options. At the second level students are asked to give a reason or their reasoning on the answers chosen. Presentation answers and reasons given by the students and the students analyzed and interviewed nine respondents by category of high, medium and low. The results showed the error of misconceptions on the respondents and conception does not depend on the level of education as evidenced by the acquisition of junior high students score higher than high school students and college students. Finally, further advice is based on an error of misconceptions and research development are presented for future research. The results obtained are the respondents students and students assume the time required to rotate months is not the same as the time required months evolved, respondents can not explain well the position and phase of the moon that occurred, respondents have a better conception on the questions about the eclipse compared with Matter for the phase of the month and the level of education of the respondents did not affect the respondents have a higher conception.
