Abstract

The laboratory diagnosis of candidiasis can be done by means of microscopic examination, molecular identification, serological tests and culture systems on specimens. In the method of culture of candidiasis disease is done by isolating the fungus Candida albicans from patient samples on the media. Several previous researchers managed to find a natural medium for the growth of the fungus Candida albicans.This research is experimental using Completely Randomized Design (CRD) with 2 treatment factors. Factor 1 is the type of fungus, namely Candida albicans, factor 2 is the type of natural media, namely media from corn, cassava and yellow sweet potato and PDA media as control. In the observation of 48 hours incubation, the average growth ofcolonies Candida albicans was 4.27 on corn media, 5.87 on cassava media, 5.70 on yellow sweet potato media and 2.48 on PDA media. From the results of the Saphiro-wilk test, it was found that the data were normally distributed (p>0.05). The data from the Levene Test showed that the data were assumed to be the same (p>0.05), and thetest One Way ANOVAfound that (p<0.05) which means that there were differences in the growth of Candida albicans on corn, cassava and yellow sweet potato media.

Abstract

Consumption of coffee in Indonesia has increased every year. This is what makes the trend of coffee drinkers continue to increase. There are caffeine in coffee which are an iron absorption inhibitor where caffeine binds iron so it cannot be absorbed by the body. Iron was required in the synthesis of hemoglobin. Reduced hemoglobin levels can cause anemia. The limit of male hemoglobin levels for diagnosing anemia is when the hemoglobin level was less than 13 g/dl. This study aims to determine the relationship between habits drinking of coffee with hemoglobin levels in visitors to the "Sederhana Kopi" Surakarta Shop. The type of this research was observational analytic with cross sectional approach. The time of the study was carried out in February - June 2021. The sample in this study was 30 samples with a total sampling technique. After conducting research on 30 respondens, the results of the study using the Repeated One Way Anova test obtained a significance value of 0,302 (p <0,05) and a correlation value of  0,302.  From the results, it can be concluded that there is no significant relationship between habits drinking of coffee and hemoglobin levels in visitors to the "Sederhana Kopi" Surakarta shop.

Abstract

Cervical cancer is a cancer that apprears in the cervix caused by Human Papilloma Virus (HPV) infection. According to the World Health Organization (WHO) explains that cervical cancer is the fourth-order cancer that represents 6,6% of all cancers is women. Pap smear is an examination done to detect cervical cancer. Pap smear examination using vaginal swab examinations material that must be fixated before coloring papanicolaou. This study aims to determine the defference in alcohol fixation result 96% for 15 minutes and 30 minutes using papanicolaou coloring conducted at Dyatnitalis Anatomy Pathology Laboratory Palembang. Type of cross sectional research, with research design using intact group comparison. The sample consisted of 8 respondents each performed a fixation of alcohol 96% for 15 minutes and the next 30 minutes colored with the coloring papanicolaou. Research is conducted starting from patient preparations, examination material retrieval, examination material processing, analysis and examination result. Wilcoxon test results are known that the significant value is p = 0,024. The value obtained is p =? 0.05.The results can be concluded that there is a difference in alcohol fixation results 96% for 15 minutes and 30 minutes.

Abstract

Cross Match is an examination by reacting donor serum with patient cells (minor) and reacting patient serum with donor cells (major). Research has been carried out on the effect of time and temperature of storage of blood samples on the results of the Cross Match examination. The method used in this research is the gel test method. Blood samples were stored at refrigerator and room temperature with storage times of 0, 2nd, and 4th days. The results showed that there were differences in the quality of the results of the Cross Match examination on blood samples with storage times of 0, 2nd, and 4th days. The results of the Cross Match examinations at the refrigerator temperature for the 2nd and 4th days were the same as the control samples. The results of the Cross Match examinations at room temperature for the 2nd day were still the same as the control samples, while on the 4th day showed different results, namely there was agglutination accompanied by mixed fields on the results of the Major, Minor and Auto Control. Then the conclusions obtained are the time and temperature of storage of blood samples affect the results of the Cross Match examinations.

Abstract

Hepatitis B occurs in people throughout the world and is one of the world's major health problems. Hepatitis B virus infection can develop into chronic hepatitis, liver cirrhosis, liver cancer and even death. This disease is caused by the hepatitis B virus which is transmitted through exposure to blood, and does not cause specific symptoms so that an infected person can carry out blood donor activities. Blood donors are people who donate blood or components to patients for the purpose of healing diseases and restoring health. Donor blood needs to be screened for screening tests to avoid the risk of disease transmission through blood screening tests for infectious infections through blood transfusion (IMLTD). One of them is a screening test for hepatitis B to detect Hepatitis B surface antigens (HBsAg). Research method: This research is descriptive. The purpose of the study: to find out the description of the results of hepatitis B strain test on blood donors based on the type of donation in the Blood Transfusion Unit of Raden Mattaher Hospital Jambi Province in 2019. Data analysis using bivariate and univariate. Results: 335 blood donors were 6 (1.8%) people with reactive HBsAg results. Conclusion: Based on the type of donation from 83 voluntary donors, there were 2 (2.4%) reactive HBsAg people, while 252 donor replacements were found in 4 (1.6%) reactive HBsAg people. Suggestion: Donors with reactive test results should be notified, so counseling can be done and referred to a confirmation check.

Abstract

Platelet components for transfusion are classified by various techniques or types of blood bags, namely manual centrifugation (Tc-Tr, Tc-Qd, Tc-Bc) and apheresis (Tc-Ap). The effectiveness of the use of these components is assessed through internal quality control. This study aimed to determine the quality of the platelet components with the examination of pH, volume, platelet count, and leukocyte count. Platelet index (MPV, PDW, and P-LCR) was performed as an additional examination. The research data were analyzed based on PMK No. 91, 2015 standard. The difference test was performed between manual centrifugation techniques (Tc-Tr, Tc-Qd, Tc-Bc) and apheresis (Tc-Ap) with p-value < 0,05. The results of the analysis showed that the parameters of pH, volume, and the number of leukocytes were by the standard, except for the number of platelets in the components of Tc-Tr and Tc-Qd. MPV and PDW values tend to be low and P-LCR values are generally normal. This can indicate that the platelet component is generally not in an activated state. The conclusion of this study, the Tc-Ap component is the best platelet component with quality control that is not significantly different from the Tc-Bc component.

Abstract

Hemoglobin is a protein in red blood cells that serves to transport oxygen from the lungs throughout the body. Hemoglobin may increase or decrease. Decreased hemoglobin levels in the blood called anemia. Decreased hemoglobin levels often occur in the elderly because of changes in the elderly body that cause damage to the bone marrow so that a decrease in the production of erythrocyte cells can result in a lack of hemoglobin in the body. Anemia is caused by many factors including bleeding, low nutrition, iron levels, folic acid, low vitamin B12 and problems in digestion and bone disorders. The purpose of this study is to know the picture of hemoglobin levels as screening anemia in elderly people aged 60-74 years in sukasari village in Seluma Regency. In this study using primary data with sampling techniques using purposive sampling. Examination directly using the Cyanmethemoglobin method. The results of the study obtained hemoglobin levels in seniors aged 60-74 years with a total of 30 samples obtained 17 samples with low hemoglobin levels with a percentage value of 56.7% , 12 samples have normal hemoglobin levels with a percentage of 40% and one sample high hemoglobin levels with a percentage of 3.3%.

Abstract

Tofu pulp is a waste that is widely produced along with the increasing need for tofu. The advantages of tofu waste are high nutritional content, cheap and easy to obtain. The by-product of this tofu industry still contains 26.6% protein, 18.3% fat, and 41.3% carbohydrates for every 100 grams. Based on this, tofu pulp has the potential to be used as raw material for Candida spp growth media, replacing the gold standard medium, which is Potato Dextrose Agar. The ability of tofu pulp flour media to support the growth of Candida spp. need to be tested further for the number and morphology of Candida spp. The purpose of this study was to determine whether tofu pulp media could be used as a substitute for Potato Dextrose Agar media for the growth of Candida albicans and Candida spp. This study is an experimental study with a completely randomized design (CRD). The composition of the tofu dregs flour media used includes tofu dregs flour (0, 1, 2, 3, 4, 5gr); dextrose 2gr; agar 1.5gr; and 100mL aquadest.  Candida inoculation on tofu pulp flour and Potato Dextros Agar media was using a streak plate method. Observations included the number of colonies and the morphology of the tested Candida colonies. The number of Candida albicans and Candida sp colonies growing on tofu pulp flour media was much larger than on control media, but the size of the colonies was  small. Colony recovered from  Tofu pulp flour media with a ratio 5gr tofu pulp flour and 2 gr dextrose were 222.5 CFU/10uL for Candida albicans and 148 CFU/10Ul for Candida sp. The test results of Candida growth on gold standard media were 54.5CFU/10uL for Candida albicans and 50 CFU/10uL for Candida sp. Based on the results of this study, tofu dregs flour media was able to support the growth of Candida spp in terms of numbers but was not able to provide large colony sizes as in gold standard media. For further research, different methods can be used in making the media or adding additional materials for nitrogen sources in the media.
