Abstract

Wudhu sebagai aktifitas rutinitas yang dilakukan oleh muslim sebelum melakukan ibadah telah diteliti memiliki manfaat dalam menjaga kesehatan tubuh. Kulit sebagai bagian tubuh manusia yang terpapar langsung dengan perlakuan wudhu, merupakan barier pertama tubuh dari lingkungan luar dan dari masuknya mikroba yang berkolonisasi di permukaan kulit. Adanya proses membasuh kulit dengan air memungkinkan adanya terlepasnya bakteri dari kulit sebagai akibat kontak bakteri kulit dengan air. Penelitian ini bertujuan untuk mengetahui perbedaan jumlah bakteri kulit pada sebelum dan sesudah perlakuan wudhu. Penelitian ini memiliki dua tahapan, tahapan pertama membandingkan jumlah bakteri kulit pada kelompok muslim dan non muslim. Pada tahap kedua, dilakukan penghitungan jumlah bakteri kulit responden muslim pada lima titik waktu pengamatan. Penghitungan bakteri kulit dilakukan dengan mengambil sampel usap dari permukaan kulit pada ventral lengan reponden untuk kemudian dilakukan penghitungan jumlah bakteri dengan seri pengenceran yang sesuai. Perbandingan data kelompok muslim dan non muslim dianalisis secara deskriptif. Sedangkan data pengamatan jumlah bakteri sebelum dan sesudah wudhu pada lima titik waktu kemudian dianalisis statistik menggunakan t-test berpasangan. Hasil penelitian ini menunjukkan adanya pengaruh perlakuan sebelum dan sesudah wudhu terhadap jumlah bakteri kulit (p=0.006). Kata kunci: Wudhu, Bakteri Kulit, Hitungan Cawan.

Abstract

Masyarakat masih banyak yang belum sadar pentingnya menjaga kesehatan demi mencegah penyakit degeneratif dimasa tua. Apalagi pada jaman sekarang begitu banyak makanan siap saji yang berpotensi menimbulkan banyak penyakit, salah satunya adalah diabetes mellitus. Tim pengabdian masyarakat memberikan penyuluhan untuk meningkatkan pemhaman dan wawasan tentang pentingnya pemanfaatan herbal medicine dalam rangka pencegahanpenyakit diabetes mellitus. Tercapainya target dari kegiatan pengabdian kepada masyarakat ini, terbukti dari antusiasnya para peserta mengikuti penyuluhan dari mulai sampai berakhirnya acara serta banyaknya pertanyaan daripara peserta. Sedangkan tercapainya tujuan dan manfaat dari penyuluhan tentang pemanfaatan herbal medicine bagipasien pre diabetes memberikan wawasan bagi masyarakat Desa Gelang Kabupaten Sidoarjo tentang pentingnya perankonsumsi tanaman herbal dalam menunjang penurunan kadar glukosa darah adalah menjadi tolak ukur keberhasilanpenyuluhan ini.

Abstract

The Snakehead fish contains high protein that was mostly used as a treatment during healing process in either invasive or passive surgery. Therefore, it could be developed as well as a mixture of wounds dressing for skin healing. The aim of this study was to know the effect of  combination of concentrations between chitosan and collagen of  Snakehead fish  (Channa striata) skin and scales on fungal total number recovered from the composite. Snakehead fish skin and scales was treated by soaking in 2% HCl solvent for 48 hours to obtain collagen from its filtrate. Filtrate obtained continued to neutralize with 1 M NaOH until soluble collagen  appeared. Collagen obtained in this study then  mixed with 2% chitosan liquid to make wound dressing in various concentrations group. Combination of colagen mixture in this study were chitosan liquid only as control, 25% collagen-75% chitosan (C1), 50% collagen-50% chitosan (C2) and 75% collagen-25% chitosan (C3). The study results showed that on Control, C1 and C2 group there was no fungal contamination, but on C3 group there was contamination of fungal with total counting about 2,43 x 103 CFU. Based on statistically test showed that there was discrepancy for each group with p-value was 0,02 (p<0,05).

Abstract

Wound dressing has a function to protect the wound area from external exposure and provide a moist state around the wound area for accelerating wound healing process. Collagen and chitosan are known as appropriate biomaterials to synthesise of wound dressing because they have anti-bacterial, anti-fungal, biodegradable and biocompatible properties. One of biocompatibility assay for a material is cytotoxicity assay using Brine Shrimp Lethality Test (BSLT) that could be applied before in Vivo assay. The aim of the research was to know the cytotoxicity level of collagen-chitosan wound dressing with variance of concentration such as K0 as control used pure chitosan, K1 used collagen 25% and chitosan 75%, K2 used collagen 50% and chitosan 50%, K3 used collagen 75% and Chitosan 25%. Skin and scales of Gabus fish (Channa striata) were extracted using 2% HCl solvent to obtain collagen and chitosan powder dissolved in 1% acetic acid. Furthermore, wound dressing is made by a combination of collagen-chitosan concentration according to each group (K0, K1, K2 and K3. Citotoxicity assay used Brine Shrimp Level Test (BSLT) method with concentration each sample group were 10, 50, 100, 250, 500, 750 and 1000 ppm. The results showed that each wound dressing group such as K0, K1, K2 and K3 had LC50> 1000ppm that indicated wound dressing was non-toxic.

Abstract

Collagen gives a moist state on the wound area to accelerate the wound healing process. Chitosan is a polymer as known as non-toxic, antibacterial, antifungal, biodegradable, and biocompatible materials. Combination of collagen and chitosan expected to be the best biomaterials as a wound dressing for the healing process. The study aimed to determine cytotoxicity assay on collagen-chitosan wound dressing sterilized by ultraviolet (UV) Light using Brine Shrimp Lethality Test (BSLT) method. The test groups were divided into K0, K1, K2, and K3 groups. K0 contained pure chitosan as a control group, K1 contained collagen 25%-chitosan 75%, K2 contained collagen 50%-chitosan 50%, K3 contained collagen 75%-Chitosan 25%. Collagen extracted from skin and scalp of snakehead fish (Channa striata) then mixed with chitosan until formed collagen-chitosan wound dressing. This study used Brine Shrimp Level Test (BSLT) method with solution concentration 10, 50, 100, 250, 500, 750 and 1000 ppm. Based on the results showed that K0, K1, K2, and K3 group had LC50 >1000 that proven collagen-chitosan wound dressing was non-toxic materials. As conclusion of the study explained composite wound dressing based on collagen-chitosan in all groups that sterilized under UV-Light along 15 minutes was not toxic based on Brine Shrimp Lethality Test showed LC50 >1000.

Abstract

Crystal violet and Safranin are dyes in Gram staining, which are carcinogenic. Alternative safe materials are needed to minimize the use of carcinogenic properties. Purple sweet potato (Ipomoea batatas L.) peels were the candidate of the alternative dye source because of its high anthocyanin pigment. The purpose of this study was to determine purple sweet potato (Ipomoea batatas L.) peels extract as an alternative to Gentian violet in Gram staining of bacteria. Extracts obtained from purple sweet potato peels studied with varying concentrations of 50%, 60%, and 75% for 1, 3, and 5 min as a substitute for Gentian violet on Bacillus sp. The parameters observed from this study based on visual field clarity, glass slide cleanliness, contrast, bacterial shape, bacterial colour. Each extract concentration compared with a control group using Gentian violet. The results showed that optimum staining in 50% concentration for 5 min, 60% concentration for 5 min, 75% concentration for 3 min, and 5 min respectively. The present study exhibited the potency of Ipomoea batatas L. peels extract as an alternative staining agent.

Abstract

Escherichia coli is the bacteria that can cause diarrhea in humans and often used as a parameter of stool environmental pollution. Culture of E. coli from the sample often requires Mac Conkey as commercial media which is able to distinguish it from other bacteria in the Enterobacteriaceae group. Commercial media such as Mac Conkey certainly has a price that is quite expensive because of its ability as a growth medium for Enterobacteriaceae. Therefore, in the study tested natural ingredients that can be used for growth media, such as coconut water. The purpose of this study was to compare the ability of Mac Conkey media and coconut water to support the growth of E. coli. This research is an experimental study with a completely randomized design. The concentration of coconut water tested was 0%, 20%, 40%, 60%, 80%, and 100%. The results showed that at the concentration of coconut water 20% to 60% the number of E. coli colonies on coconut water media was slightly below the Mac Conkey Agar media, while in coconut water a concentration of 80% showed a greater number of colonies than Mac Conkey. The Mann Whitney test showed a significant difference between the number of colonies on 80% coconut water media and Mac Conkey Agar, which was equal to 0.004 (p < 0.05). Based on these results, coconut water has the potential to be used as a growth medium for E. coli.

Abstract

Snack as food and beverages, present vastly in Surabaya. Start from street vendor to famous stores. The distribution of these snacks trader often found in schools, from primary to high school. Common problems from these food dan baverages sales is their hygine. With a low sales price, other raised problem are the using of raws material for snack production which lead to hazard microbial contamination. This reaserch aim is to monitors the quality of beverages on microbiology that been sold in around primary school of Jemur Wonosari District, Surabaya. Two microbiology parameters that been analize were coliform bacterial and Escherichia coli number. Beverages sample that been used were 9 samples, then analize using most propable number using 15 tube series. The Results then analize descriptively. From 9 samples, all were exceed the standar quality according to Kepmenkes RI No.492/MENKES/PER/2010.
