Abstract

Bacterial resistance to antibiotic is one of the factors triggering infection therapy failure. This study was conducted to determine the prevalence of carbapenem-resistance Klebsiella pneumoniae infection and the phenotype of carbapenem-resistant Metallo-Beta-Lactamase (MBL)-producing Klebsiella pneumoniae isolates at RSUP Dr. Wahidin Sudirohusodo Makassar. This study included Klebsiella pneumoniae identification on each infectious patient’s isolates. The sensitivity test of antibiotics, phenotype confirmatory test, and MBL phenotypic test were conducted using agar diffusion Kirby-Bauer, Vitek-2-Compact, and Double Disc Synergy Test (DDST) method, respectively. As the result, the antibiotic sensitivity test using the Vitek-2-Compact method on 50 clinical samples (pus, sputum, blood. tissue, urine, brain fluid, and feces) found that 10 isolates (20%) were resistant to carbapenem.  The phenotypic test using the Double Disc Synergy Test (DDST) method found that carbapenem-resistant isolates caused by the production of Metallo Beta Lactamase (MBL) enzymes were 2 isolates or 20% of the total carbapenem-resistant isolates.

Abstract

This research was carried out with the aim of determining the inhibitory power of the combination gel of propolis extract and mulberry leaf extract against Propionibacterium acnes bacteria and determining the concentration of propolis extract that provided the greatest inhibitory power against the test bacteria. The research method includes the extraction of mulberry leaves by maceration using 70% ethanol as a solvent and the propolis extract used is a ready-to-use extract. The formulations were designed with various concentrations of propolis (0.5%, 1%, 1.5%) and 0.5% mulberry leaf concentration. Testing the antimicrobial activity of propolis extract in combination with mulberry extract in gel preparations against the growth of Propionibacterium acnes bacteria was carried out using the agar diffusion method. The results showed that the average diameter of the inhibition area at 0.5% concentration was 36.87 mm, at 1% concentration was 44.557 mm and at 1.5% concentration was 45.97 mm, positive control was 49.21 mm. Based on the data obtained, it is concluded that the gel formula with a propolis concentration of 1.5% is the preparation with the largest zone of inhibition.

Abstract

Pharmacy services is a direct service and obligation to the patients regarding the pharmacy substances with the aim to increase the public health services in the field of pharmacy. The purpose of this study is to evaluate the implementation of pharmaceutical standard at the primary health services Puskesmas. This study is conducted in descriptive research, data collection using a cross sectional survey approach. The instruments in this study was a questionnaire sourced from the Regulation of the Minister of Health No. 74 of 2016 concerning pharmaceutical service standards in Puskesmas. Respondents in this study were pharmacists at the Pagesangan Puskesmas. The management activities reach 100% of drug and medical consumables, 93.75% of equipment and supporting facilities, 100% of human resources. These results indicate that the evaluation of the implementation of pharmaceutical service standards at the Puskesmas based on pharmaceutical service indicators has been carried out well at the Pagesangan Puskesmas with an average value of 96.77%. The implementation of pharmaceutical service standards at the Pagesangan Puskesmas has met all the indicators of pharmaceutical service standards at the Puskesmas.

Abstract

Dyspepsia is a collection of several clinical symptoms consisting of pain in the pit of the stomach, flatulence, early satiety, nausea, and vomiting. The incidence of dyspepsia at the Karang Taliwang Public Health Center until 2019 was quite high. Evaluation of drug use at the Puskesmas has never been carried out, so it is necessary to know the pattern of use of dyspepsia drugs at the Karang Taliwang Public Health Center. This review aimed to determine the pattern of dyspepsia drug use in outpatient dyspepsia at Karang Taliwang Public Health Center using prescribing indicators developed by the World Health Organization. This study is a descriptive study of retrospective drug use. Sampling using a consecutive sampling method. Prescription data and medical records were collected through observation sheets. The results of the study on drug use patterns were the average number of drug items per sheet of 2.55 prescription; the percentage of drugs prescribed with generic names was 99,37%; the percentage of prescription drugs with antibiotics was 1.61%; the percentage of prescription drugs with injection preparations was 0%; and the percentage of drugs prescribed according to the Formularium was 96.84%.

Abstract

Songga (Styrchnos lucida R.Br) is a plant that has potential to be developed as an anti-breast cancer against. This study was conducted to predict the secondary metabolite derived from songga, which potential as anti-breast cancer through inhibition of estrogen receptor-α. A molecular docking study was carried out using Autodock Vina assisted by AutodockTool and Biovia Discovery Studio. The study used estrogen receptor-α as a protein and ten compounds from songga as ligands. The 3D structure of estrogen receptor-α and ten ligands were obtained from Protein Data Bank and the PubChem database. The result revealed that strychnine N-oxide is the compound that has the lowest free binding energy and inhibition constant with the value 9.6 kcal/mol and 0.09 µM, respectively. This indicated that strychnine N-oxide has an excellent affinity for estrogen receptor-α. Thus, this compound is predicted to inhibit the estrogen receptor-α and can be developed as anti-breast cancer. However, further investigations such as molecular dynamics simulation, in vitro and in vivo assays should be undertaken.

Abstract

Schizophrenia is a chronic mental disorder characterized by distortions in thinking and behavior. Schizophrenic patients receive antipsychotic therapy (main) and non-antipsychotic (supportive) for a long time. Prescription drugs are given either single or in combination, that can possible for drug interactions. This study objective to determine the potential drug-drug interactions in schizophrenic patients. The design of this study was cross-sectional with purposive sampling technique. The data used is secondary data from the medical records of inpatient schizophrenia patients at the Mutiara Sukma Mental Hospital NTB in 2020 with a retrospective method. The sample data obtained were 109 medical records. The results showed that there were 82,2% of drug interaction cases, namely the combination of risperodone and lorazepam. The number of major interaction cases is 24,2%, moderate is 72,1% and minor is 3,6%. The high incidence of drug interactions in schizophrenic patients requires closely monitored.

Abstract

Self-medication often becomes a choice for various complaints, one of which is menstrual pain. Regarding the description of the use of menstrual pain medication independently, it is still limited. The use of a questionnaire as a research instrument that has been tested for validity and reliability is very necessary. Therefore, this study aims to develop and test the validity and reliability of the description of the independent use of menstrual pain medication in female students. This research is a descriptive survey type with a cross-sectional design. The validity test was carried out using a qualitative and quantitative professional assessment method, namely by using the Content Validity Ratio (CVR). An advance validity test was conducted by 5 experts and 40 respondents. The reliability test was carried out with 40 respondents and used the Chronbach Alpha value. The results of the validity test obtained 28 questions with a CVR value of 1. Facial validity was declared valid after one repair. The reliability of the test obtained Cronbach's Alpha coefficient value of 0.934. Based on the validity and reliability test of the questionnaire, it was declared valid and reliable so that all questions could be used as an instrument to find out the description of the use of menstrual pain medication independently.

Abstract

Candida albicans (C. albicans) is the norm microbiota that lives in humans' oral cavity, digestive tract, and urinary tract. However, C. albicans, under certain circumstances can cause superficial infections of the mucosa such as oropharyngeal candidiasis (OPC) or vulvovaginal candidiasis (VVC) and even systemic infections. Secreted aspartic proteinase (Sap) 1 plays a role in the phenotype change of C. albicans. The study aimed to perform molecular docking using the bioactive compound Ocimum sanctum to inhibit Sap 1 C. albicans. Methods used include preparation of protein structures, ligands preparation, simulation of docking, and analysis and visualization. The results showed Apigenin had the highest binding energy of 7,792 kcal/mol and was followed by methyl eugenol ligand 5,361 kcal/mol, Citral 5,307 kcal/mol, Ursolic acid 4,967 kcal/mol, and Taxol 2,118 kcal/mol. Known interactions from the docking results showed that the four ligands hit the catalytic residue Asp32/Asp218, but only ursolic acid did not hit the catalytic residue. This study concludes that all ligands have binding energies that inhibit Sap 1, resulting in various interactions, amino acid residue contacts, and dissociation constants.

Abstract

Pineapple plants are used ethnomedicinally in various countries, especially in Indonesia. Several studies have shown that pineapple plants have pharmacological effects both in vitro and in vivo on various diseases. Ananas Plant has the potential to be cultivated as medicine products. The toxicity of Ananas comosus (L) Merr important to acknowledge. This literature review aims to provide comprehensive information through a literature review regarding the toxicity of pineapple (Ananas comosus [L.] Merr). The literature review was collected in the period 2010 to 2021 which was obtained from the Google Scholar and Pubmed databases with a total of 9544 and a study was carried out on 10 works of literature. Based on the results of 10 literatures that have met the inclusion criteria, it shows that the in vitro toxicity test on pineapple (Ananas comosus [L.] Merr) has an LC50 of 403.83 to 9000 ppm which belongs to the category of practically non-toxic to moderately toxic. In vivo toxicity test showed that LD50 > 5000 mg/kgBW showed no deaths which were classified as practically non-toxic. From the results of literature research, it can be said that toxic plants (Ananas comosus [L.] Merr) have a toxicity category of practically non-toxic to moderately toxic in vitro studies. The results of the in vivo test showed that there were no deaths in the test animals which were in the category of practically non-toxic to mildly toxic.
