Abstract

By product Muarenesox talabon swim bladders can be used as a raw material for desperately needed in the food, biomedical, pharmaceutical, and cosmeceuticals industries. The aims of the research were to observed the histological and determine chemical characteristics of swim bladder including proximate and amino acids; extraction of acid soluble collagen and determine the characteristics of collagen including proximate, pH, heavy metals, microbial, amino acids, functional groups, molecular weights, and thermal stability. The morphology of cunang swim bladder consists of outer, middle, and inner layers containing collagen fibers; 33.67±0.71%wb and protein whichwere dominated by three amino acids that were glycine, proline, and alanine. Pretreatment by 0.1 M NaOH for 8 hours (K1T4) and acid extraction by 0.25 M acetic acid for 72 hours (M1T3) was the best treatment yielding 14.51± 0.43% of collagen; having 12.12±0.04% wb of moisture; 88.54 ± 0.08% wb of protein; 1.31±0.23% wb of fat; 0.17±0.03% wb of ash. Not detected any heavy metals (Pb, Hg, As, Cd). Acidity pH was 4.31 and negative of E. coli and Salmonella. The main amino acids detected were glycine 241.06 mg/g; proline 88.73mg/g; and alanine 86,98 mg/g; FTIR spectra were revealed the presence of triple helix structures; electrophoresis patterns consisted of 136 kDa of mol weight of α1 and 117 kDa of mol weight of α2 were characterisedto be type I collagen; which had Tmax of 195.59ºC and ΔH 7.8113 J/g. Keywords: acid extraction, swim bladder, collagens, thermal stability

Abstract

The research aimed to discover the runoff suspended solid load of Gunung Kijang estuary to coastal area, East District of Bintan. The research was held in May and September 2015. The total suspended solid (TSS) load in five estuaries was measured on May. On September, measurement was focused in two biggest estuaries, Kawal and Galang Batang. The TSS in coastal water area was measured both on May and September. Regarding to five estuaries measured, the lowest TSS came from Angus River while the highest comes from Kawal River. TSS load in May on both estuary were lower than in September. Suspended solid load at the lowest tide period was higher than at the highest tide at both estuaries. In May TSS load in the Kawal estuary was 528.723 mg/sc and Galang Batang estuary was 172.903 mg/sc. In September TSS load in the Kawal estuary was 720.920 mg/sc while Galang Batang estuary was 446.893 mg/sc. These results indicate that tidal patterns and current were major influence on the TSS load in estuaries. Keywords : Bintan, coastal water, estuary, load, TSS

Abstract

The study aims to explore the antibacterial activity of Stichopus vastus against pathogenic MDR bacteria. Analysis of samples of sea cucumbers included extraction, fractionation, and analysis of bacterial sensitivity test Gas Chromatography-Mass Spectrometry (GC-MS), the extraction process is carried out by solid-liquid extraction method. Fractionation was done with Open-Column Chromatography (OCC). Sensitivity test of bacteria was done using an agar diffusion method according to the Kirby-Bauer (Ref). The study revealed that from 5 species MDR bacteria, which are Coagulant negative stapylococi (CNS), E. coli, Enterobacter 5, Klebsiella sp. dan Pseudomonas sp. There are two MDR bacteria had the most sensitive responses by the extract of sea cucumber, which were Enterobacter-5 and Klebsiella sp. The two bacteria were tested against five bioactive fractions obtained from OCC. Fraction criteria-2 had the highest antibacterial activity against Enterobacter-5 and Klebsiella sp, with serial concentration of 20 µg ∙ disc–1, 40 µg. disc–1 and 80 µg. disc–1.  Largest inhibition zone were obtained from 80 µg. disc–1 againts the two bacteria were (14.73 ± 0.48) mm and (11.22 ± 0.85) mm respectively.  GC-MS Analysis revealed that fraction criteria-2 had (or consisted of) cyclohexhane, ethanol, butanoic and pentanoic acids. Keywords: antibacterial activity, multi drug resistance (MDR), sea cucumber, Stichopus vastus

Abstract

Talise waters, located in North Sulawesi, is one of many coastal areas in Indonesia with sea grass cover extending along the reef flat. This ecological feature is beneficial for living habitat of associated fauna, including brittle stars. Brittle stars are group of Echinodermata reported to be abundantly found in sea grass ecosystem. However, the diversity of this fauna is less reported except as part of echinoderm studies. This research aims to investigate the diversity of brittle stars in Talise waters. Data were collected in July 2009 applying quadrant transect method. In total of 650 individuals belonging to 4 families (Ophiotrichidae, Ophiodermatidae, Ophiocomidae and Ophiuridae) and 10 species were recorded during this study. The diversity (H’=1.58), richness (D=1.69) and Evennes Index (J=0.89) was relatively higher compared to adjacent water in North Sulawesi such as Wori (H =1.64, D =1.38, J=0.85), Kema (H =1.3, D =0.38, J=0.36) but lower than East Likupang waters (H =1.95, D =2.42, J=0.97). Keywords: brittle star, diversity, richness, evennes, Talise waters

Abstract

Fish freshness plays an importantant role in determining quality of fish product. Organoleptic is one of sensorik method for determining fish freshness. The fish deterioration mainly affected by temperature. This study aimed to determine the best method for determining organoleptic quality of skipjact tuna based on chilling methods and storage time. Fish sampling were taken from the Seram Sea, Maluku Province. Chilling methods was done by the different ratio between the ice and fish: first was without using ice, second was ratio 1:1 and the third was ratio 1:2. The time storage were 0, 2, 4, and 6 hours.The results showed that chilling methods ratio between the ice and fish of 1:1 with 6 hours storage time provided the best organoleptic quality with eye 8.87, mucus 8.83, gill 8.67, meat 8.73, odour 8.80, and texture 8.86.  Keywords: chilling methods, organoleptics, skipjact tuna

Abstract

This research was conducted in April, July and October 2013 at 8 (eight) stations: 1. Tj. Intan, 2. Tj. Tempurung, 3. Tj. Bunga Dalam, 4. Tj. Bunga dalam, 5. Tj. Burung, 6. P. Dabung, 7. Tasik Malaya, and 8. Tj. Harapan. This study aims to determine the relationship between several physic-chemical parameters and phytoplankton abundance. The results show the growth of phytoplankton biomass (chlorophyll a) was determined by high concentration of NH4 and NO3, while its growth is limited by turbidity (turbidity). Abstract

The dried sea cucumber Stichopus vastus is one of the commercially species harvested in Indonesian waters. This study aims to highlight the nutritional value and heavy metals content of dried sea cucumber S. vastus. Proximate (moisture, ash, protein, fat and carbohydrate), mineral (sodium, calcium, potassium and iron) and heavy metal (mercury, cadmium, arsenic and lead) were determined by standard method of AOAC, while phosphorous was determined by spectrophotometric method. Chondroitin sulphate was determined by UPLC method, glucosamine sulphate and vitamin (A, B1, B2 and E) by HPLC method. Results show that protein was the major component in proximate analysis of dried sea cucumber S. vastus in the present study. The protein content was 38.70%. Moisture, ash, fat and carbohydrate content were 19.46%, 34.04%, 0.38% and 7.42% respectively. All vitamins and heavy metals examined in this study were not detected. The sodium content was 8054.36 mg/100 g higher than other minerals. Calcium, potassium, phosphorus and iron content were 2449.9 mg/100 g, 159.77 mg/100 g, 5085.2 mg/100 g and 520.8 mg/100 g respectively. Glucosamine sulphate content was found to be 2.429 g/100 g, whereas chondroitin sulphate was found to be 1.115 g/100 g. It can therefore, be concluded that the dried sea cucumber S. vastus from Salemo Island is safe for human consumption and hence can be used as a source of food supplement in the future. Keywords: food supplement, Salemo island, Stichopus vastus

Abstract

Cendrawasih Bay is habitat for olive ridley (Lepidochelys olivacea) which is directly adjacent to the Pacific Ocean. The presence and the diversity of L. olivacea in Cendrawasih Bay has been threatened. Genetics can be the key to conservation because they play an important role in maintaining population and recovering from damage. This study aims to determine the genetic diversity of L. olivacea and its association to the current pattern in the waters of Cendrawasih Bay. Samples of L. olivacea (n=20) were collected in Kwatisore (n=8) and Yapen Island (n=12) in Cendrawasih Bay waters from August 2015 to December 2016. The molecular analysis was based on 791-bp fragment of D-Loop on the non-coding region gen. The current pattern analysis was performed through INDESO data and visualized by using Ferret software. Phylogenetic analysis showed that 2 groups of L. olivacea from a total of 2 haplotypes, whose population from Kwatisore was dispersed in two haplotypes, while the population from Yapen Island was only dispersed in one haplotype. Populations from Kwatisore showed higher variations than populations from Yapen Island. The current pattern analysis suggests that the two study sites, Kwatisore and Yapen islands are different. Both populations are only connected by the Northwest monsoon currents period that indicates a little geneflow between this populations. Abstract

The rapid development of infrastructure that occurred in the area of Tanjung Api-api resulted in changes in the function of mangrove land along the coast. Directly, this also impacts on the sustainability of ecological functions of mangroves. This study aims to estimate changes (prior to infrastructure development and current conditions) of aboveground carbon reserves stored in mangrove forests in Tanjung Api-api. This research was conducted in June-August 2016. Carbon stocks in mangrove along Tanjung Api-api were conducted by surveying the mangrove species and density, and then calculating biomass and carbon stock using allometric equation. While changes in carbon stocks are calculated by converting changes in mangrove area before and after infrastructure development to carbon stock value. The change of mangrove area was done by doing image data processing in 2005 and 2016. The results showed that mangrove Tanjung Api-api has 9 species of mangrove with total mangrove aboveground biomass of Tanjung Api-api is 1.1498 Mg.ha-1 and stored carbon of 0.5235 MgC.ha-1 and able to absorb CO2 of 1.1921 MgC.ha-1. The result of satellite image processing shows that there is a reduction of mangrove land of 1056.6 ha during the last 11 years from 9741.15 ha in 2005 (before development) to 8684.55 ha in 2016. Thus, there is a decrease of mangrove biomass by 1214.894 Mg, carbon stock is reduced by 553.090 MgC and CO2 uptake is reduced by 2029.84 MgC. Keywords : aboveground, CO2, mangrove, Tanjung Api-api

Abstract

Karimata Strait located between Kalimantan and Bangka Islands, while Gaspar Strait located between Bangka and Belitung Islands. Volume transport variations through Karimata and Gaspar Straits during 2010-2014 were studied using three-dimensional numerical model of HAMburg Shelf Ocean Model (HAMSOM). Simulated area is located in 1°0′0′′N - 4°30′0′′N and 104°0′15′′E - 110°30′0′′E with a horizontal resolution of 0.5′ (about 0.94 km). The correlations value of current velocity from model with observation is quite high i.e. 0.85 for zonal component (u) and 0.89 for meridional (v) with 95% of significance level. The results of signal analysis showed that the volume transport is strongly influenced by the seasonal period. In addition to seasonal wind, there was also weak and medium phase of El Niño and La Niña during 2010 - 2014 that affected volume transport. The average volume transport from 2010-2014 through Karimata and Gaspar Straits are -2.75 Sv and -1.98 Sv, respectively (1 Sv = 106 m3/s). The negative value shows the southward volume transport, while positive to north. Total transport to south indicates that there are may be due to parts of Indonesia Through Flow (ITF) flow through South China Sea, Karimata Strait, and Gaspar Strait to the Java Sea.Keywords: volume transport, Karimata Strait, Gaspar Strait
