Abstract

Development is an increase in the ability of the structure, function of the body to be more complex in a regular pattern. Based on initial data obtained at PAUD Muslimat in Petung Village in January 2020 by conducting the KPSP test as many as 7 out of 10 children the results were dubious. The research design is descriptive, the population and samples of all mothers who have children aged 3-4 years are 35 people. The data are presented in the form of frequency distribution tables and cross tabulations and then analyzed descriptively without statistical tests using research data and supporting theories. The results showed that most (60%) mother's knowledge was good, most (57.2%) child development was doubtful, appropriate child development (86.7%) was more in mothers who had good and sufficient knowledge than those with less knowledge ( 60%) questionable child development. The conclusion of appropriate child development is more in mothers who have good and sufficient knowledge than those with less knowledge.

Abstract

Prolonged labor is labor that lasts more than 24 hours in primi and more than 18 hours in multigravida. The incidence of prolonged labor in KB Karunia in 2019-2020 is (79.36%). The purpose of this study was to determine the relationship between anemia and the incidence of prolonged labor in the Karunia family. The population studied were 100 patients and found (79.36%) mothers who had anemia and prolonged labor and (40.54%) mothers who were not anemic experienced normal first stage labor. The research design used in this article is cross-sectional, in which only observations and measurements of variables are carried out at one particular moment. The instruments used are medical records and patient partographs or use secondary data by applying the purposive sampling method. The results showed that the results of the chi square test showed P value of 0.056 > 0.05 which indicated that there was a relationship between the length of the first stage and the incidence of anemia. This shows that anemia in pregnancy has a relationship with the length of the 1st stage. The conclusions mostly show that there is a relationship between anemia and the incidence of the duration of the 1st stage of birth. Family Planning Karunia in 2019-2020.

Abstract

DNA isolation is a basic technique in molecular biology where DNA is separated from various unnecessary components. There are various methods of DNA isolation but the most commonly used methods are column and resin methods. The purpose of this study was to compare the results of PCR using DNA template isolated by column and resin methods. The research method used in this research is descriptive experimental using 8 whole blood samples. Samples were tested quantitatively with UV-VIS spectrophotometer and qualitatively using PCR. This research was conducted from February to April 2021. The results showed that based on quantitative DNA testing, DNA samples isolated using the column method had higher purity than the resin isolation method, which means that the resin method contained a lot of contamination. However, in terms of concentration, the resin method had a higher concentration than the column method and there was no significant difference in the purity and concentration of DNA if the sample was isolated using the column method and the resin method (r=0.169, r=0.252) after the dependent T test was performed. . In the analysis using PCR, samples isolated by the column method showed a fairly clear band compared to the resin method.

Abstract

Diabetes mellitus type 2 (DMT2) is a disorder characterized by hyperglycemia due to impaired insulin secretion and insulin resistance. The purpose of this study was to identify the gene involved in the positive allele of DMT2 marker using A18 primer and is a type of exploratory descriptive study. The samples used were 2 samples of DMT2 patients from the villages of Taman and Balongbend, and 8 samples of DNA collection at the Molecular Biology Laboratory, Muhammadiyah University of Sidoarj. Based on the genetic analysis carried out, the GATB gene (Glutamyl-tRNA Amidotransferase Sub Unit B) was identified in the positive allele with a homology value of "percentage identity" between the sample sequence and the sequence data in the NCBI Genbank of 80 and 88%. This research hypothesizes that the GATB gene has a relationship with the emergence of DMT2 as evidenced by the relationship between the gene and the performance of mitochondria to produce ATP which is needed by pancreatic beta cells to produce insulin.

Abstract

Diabetes Mellitus is a metabolic disorder disease characterized by an increase in blood glucose due to insulin resistance (impaired insulin function) and or a decrease in insulin secretion. Genetically, Type 2 Diabetes Mellitus is influenced by genes that affect glucose metabolism in the body. The purpose of this study was to identify the genes involved in type 2 diabetes mellitus patients who were amplified using D20 primers. The method used in this research is descriptive experimental using 2 whole blood samples and 8 samples collected from DNA isolation in the Molecular Biology Laboratory of Type 2 Diabetes Mellitus patients. The results of the research based on the BLAST program obtained matched gene sequences from the sequenced leucine rich repeats gene. and guanylate kinase domain containing (LRGUK). The LRGUK gene is thought to be associated with Type 2 Diabetes Mellitus because the presence of SNP (Single Nucleotide Polymorphism) in the area around the LRGUK gene (7q32) shows a significant relationship with fasting blood glucose.

Abstract

The centrifugation process is able to separate the blood into several components, including the buffy coat. Buffy coat contains white blood cells that have a cell nucleus which is the source of DNA. This research uses descriptive experimental method. The samples used were 16 venous blood samples consisting of 8 samples without centrifugation (wholeblood) and 8 samples centrifuged (buffycoat). DNA isolation using the resin method. Quantitative analysis was performed using a UV-Vis spectrometer. The results showed that the average concentration and purity of DNA in the centrifuged sample was higher than in the sample without centrifugation. The result of this research is that the centrifuged sample (buffycoat) can be used for the DNA isolation process and has a higher purity and concentration value than the sample without centrifugation (wholeblood).

Abstract

DNA isolation is the first step in molecular biology work. The quality of the DNA template is very important because it can affect the DNA amplification reaction. The application of centrifugation to the sample is expected to produce a Buffy coat layer containing nuclear cells. This study aims to compare the quality of the DNA template isolated from the column with and without centrifugation. This research used descriptive experimental method. The samples used were 16 samples with the division of 8 samples given centrifugation treatment and 8 samples without centrifugation. DNA isolation column method is a DNA isolation method that uses a silica membrane, where the silica membrane works as a filter for DNA that is split from the cell. The DNA isolation method column was carried out according to the standard protocol on the Geneaid DNA mini Kit Blood/Tissue. The concentration and purity of DNA were measured using UV-Vis Spectrophotometry at 260 nm and 280 nm. Visualization of DNA isolation results was carried out using 1% agarose gel. The results of measuring the quality of DNA samples using the column method with centrifugation showed better results with clearer visualization of DNA bands than samples without centrifugation. The results of the T dependent statistical test on both samples of p value < 0.05 showed that there was a significant difference in DNA quality with centrifuged and non-centrifuged treatment.

Abstract

DNA quality test is one of the basic techniques in Molecular Biology which aims to determine the presence or absence of protein and RNA contamination. DNA quantity test can be done using a spectrophotometer. Spectrophotometer is a tool used to measure the concentration of a compound based on its ability to absorb light. The purpose of this study was to determine differences in DNA purity as measured by UV-Vis Spectrophotometer and Nanodrop Spectrophotometer. The method used in this research is descriptive exploratory with purposive sampling technique. The number of samples used is 15 samples. The results of this study are that there are differences in the concentration and purity of DNA from quantitative tests with UV-Vis spectrophotometers and Nanodrops obtained based on statistical tests using the T Dependent Test. The result of this research is that in measuring DNA purity there is a difference in DNA purity because the results of Sig (2-Tailed) use 0.001 or below 0.05 (Ha is accepted). And based on statistical tests on DNA concentration measurements, it is known that there are differences in DNA concentrations.

Abstract

The purpose of this thesis research is to find out and analyze the product, promotion and price affect the school's image, influence the parents' interest in choosing the school and to find out and analyze the school image as a mediator variable capable of influencing the relationship between product, promotion, price and parents' interest in choosing a school. schools at SD Muhammadiyah 5 Porong. This research belongs to the type of quantitative research and uses descriptive statistical analysis. Researching on a particular population or sample. Data collection with a limited questionnaire method. Respondents are the guardians of the students of SD Muhammadiyah 5 Porong as many as 125 people. Validity test analysis. It is necessary to test the validity and reliability of the researcher's information using the W-Start program. .Analysis of hypothesis testing on the relationship between variables should be carried out using path analysis assisted by the SPSS type 21 windows program. The results of the SPSS program analysis show that the influence of the Product, Promotion and Price aspects together in producing School Image is 65%. Meanwhile, the other 35% of m is influenced by other aspects.

Abstract

The musyarakah financing product is a competitive flagship product of Sharia Rural Banks (BPRS) which has not experienced growth like other financing products. The lack of development of musyarakah financing products has caused BPRS to have more products based on fixed income financing which is similar to the conventional pattern that uses a fixed income structure. PT. BPRS Baktimakmur Indah is the largest BPRS with assets of Rp. 163,147,899,000 and the financing provided to the community reaches Rp. 126,807,530 as of December 31, 2019. This research was conducted at BPRS Baktimakmur Indah Branch throughout Sidoarjo which is located at Jln. Raya Bebekan No.21 Throughout the Taman District, Sidoarjo Regency. The purpose of this study was to analyze the implementation of musyarakah financing based on the standard book of musyarakah products published by the financial services authority. By using qualitative research methods through a phenomenological paradigm approach that requires researching a natural setting, with observation data collection techniques, direct interviews and literature from companies to support data needs. After the research was carried out, it was found that the BPRS Baktimakmur Indah branch of the Panjang branch had implemented musyarakah financing in accordance with the standard book of musyarakah products issued by the financial services authority.
