Abstract

Type II diabetes mellitus (T2DM) is a metabolic disorder that has potential causing ulcer complications. Ulcers occur due to peripheral vascular abnormalities and trauma. The occurrence of ulcers allows differences in the hematology characteristics in sufferers. This study aimed to determine the comparison of the blood picture between T2DM with ulcers and T2DM without ulcers. This was analytical descriptive research using 29 samples (10 T2DM samples with ulcers and 19 T2DM samples without ulcers). A complete blood test was performed using Sysmex Kx-21N hematology analyzer. Data were analyzed using Independent T-test and Mann-Whitney U depends on the normality. Normality was done using Shapiro Wilk (Confident Level: 95%). The results of this study indicated that Red blood cell count (RBC) and Hemoglobin Count (HGB) differ significantly between T2DM with ulcers and T2DM without ulcers (p: 0.012 and 0.006). The identification of HGB levels was highly recommended to get proper treatment in T2DM.

Abstract

T2DM can be triggered by two collaborating factors, namely genetics and the environment. This study aimed to identify genetic markers that can be used to detect the possibility of a person having T2D using the random amplified polymorphism DNA (RAPD) method. The study was carried out cross-sectional and involved 60 samples consisting of 30 positive T2D samples and 30 negative samples T2D. The primer used for PCR-RAPD was D20 (5'-ACCCGGTCAC-3’). The PCR-RAPD results were then analyzed using the scoring method and analyzed using the non-parametric Chi-Square test (cl: 95%). Among T2D, 576 bp band were confirmed to be markers in the patients.

Abstract

Typhoid fever is a disease caused by Salmonella typhi infection. This disease is still a global problem. The purpose of this study was to develop a rapid detection method for Salmonella using molecular methods utilizing Inv A and Fim-C Genes. Two methods compared, which are Multiplex and monoplex PCR. The sample is in the form of the patient's blood, which is stored at 3 ml EDTA vacutainer tube consisting of 10 positive samples and five negative samples. The criteria for samples used previously have tested by widal/ immunoserology (titer more than 1/160). The results showed that multiplex PCR for detection of Salmonella using the Inv A gene and fim c gene not recommended because too many bands produced. The application of monoplex in the Inv A gene gives better results than the Fim C gene. In conclusion, the monoplex application on Inv A gene recommended than Fim C gene used to detect S. typhi in human blood samples.

Abstract

Type II Diabetes Mellitus is a disorder caused by genetic and environmental factors. Molecular analysis of T2DM abnormalities has been carried out. But the analysis in sample preparation especially the stages of DNA isolation has not been done much. The aims of this study are to investigate the effect of temperature, storage duration and level of white blood cell (WBC) with Genome Quality in T2DM. The treatment (n:10) which were divided into several tubes and then stored at 4 °Ϲ, 25 °Ϲ and 32 °Ϲ for 21 days. To determining storage duration effect, we use periodically isolation of DNA (3, 15 and 30 days) after sampling. The effect of WBC with DNA quality was carried out using 17 samples. DNA isolation was done by the DNA Isolation Kit manual without modification and then tested qualitatively and quantitatively. Based on this research, it can be concluded there is a correlation between the numbers of WBC with DNA quality. The higher the number of WBC, the higher DNA concentration (r: 0.818. p value: 0.000). The concentration of DNA at a temperature of 4°Ϲ (135.1 ± 165.2 ng / µl) was higher compared with the temperature treatment 25 ° Ϲ (29.7 ± 36.5 ng / µl) and 32 ° Ϲ (22.14 ± 7.13 ng / µl) (p

Abstract

Diabetes mellitus is a metabolic disorder characterized by not produced insulin or disruption of insulin action. Diabetes mellitus was reported correlated with TCF7L2 gene mutation. This study aimed to analyze the TCF7L2 gene in families with type 2 diabetes. 5 samples were used in this study, the methods flow through DNA Isolation, PCR, and sequencing. Samples were amplified by the PCR technique using primers that were designed to detect the TCF7L2 gene, (F: 5'-GGCTTGATTGTTGATTATGGGC3' and R:5 'TCTGGCACTCAGAGAGT 3'). The amplification and sequencing resulted in 368 bp (located at 103631-103972 bp). We found a transition mutation at C103950T. Abstract

Type II diabetes mellitus (T2D) is a genetic disorder that becomes an unsolved problem. This is because T2D can be a disorder caused by lifestyle changes and is inherited from parents. This study aimed to determine the polymorphism that distinguishes between T2D and non T2D patients and the result was predicted to the families that has T2D record. The model in this research was descriptive exploratory and consists of 80 samples (30 samples positive T2D, 30 non DM samples and the others are the member of 5 families). Firstly, the analysis was focused on determining differences in polymorphisms and then the results were applied to the 3 families of T2D using A10 Primer. Difference polymorphism was analyzed using chi-square (cl: 95%). Six bands were obtained from PCR-RAPD which a significantly different band at 997 bp. Analysis of T2D has confirmed that 5 families have a clue band. Based on the analysis this report can be used to screen the possibility of someone to have DM type 2.

Abstract

Diabetes Mellitus type II (DMT2) is a metabolic disorder which its number increases from year to year. The Control of DMT2 sufferer’s number is difficult because DMT2 is caused by genetic and environmental factors. DMT2 can worsen infectious diseases for people with other infectious diseases. This study aimed to determine the polymorphism of sufferers and non-sufferers of T2DM using PCR-RAPD method. The sample of this study consisted of 60 samples which 30 positive samples and 30 negative samples taken from several clinics in Sidoarjo district. The primer used is A 18 (5'- AGGTGACCGT-3 '). Data analysis was performed using the SPSS 16.00 program and analyzed with a Cross-sectional Chi-square with a 95% confidence level. The results of this study produced 17 bands with length of 197 bp, 239 bp, 269 bp, 319 bp, 390 bp, 530 bp, 588 bp, 686 bp, 777 bp, 972 bp, 1175 bp, 1676 bp, 2780 bp, 3843 bp, 6563 bp, 11072 bp & 18434 bp. Four of them were monomorphic and two bands showed significantly different results at 319 bp (p: 0.035) and 18434 bp (p: 0.004).there were significant differences in the resulting band at 319 (p: 0.035) bp and 18434 bp (p: 0.004). Further analysis needs to be done to confirm the genes involved.

Abstract

Pendahuluan: Berdasarkan survey di Pondok pesantren Al-Hamdaniyah kami menemukan santri yang sakit terbaring dikamar hanya beralaskan kain dan selimut. Kondisi ini sangat tidak nyaman karena 1 kamar berisi 10 anak dengan berbagai macam aktivitas yang dapat menimbulkan kegaduhan sehingga dapat mempengaruhi istirahat santri yang sakit. Selain itu dapat dibilang berbahaya karena penyakit kemungkinan dapat menularkan pada santri di sekelilingnya. Di pondok tidak ada UKS dan kader kesehatan sehingga yang sakit kadang tidak terurus. Santri pondok pesantren Al-Hamdaniyah tidak pernah mendapatkan pendidikan tentang pola hidup bersih dan sehat di lingkungan pondok pesantren.Metode: Kegiatan pengabdian masyarakat dilakukan pada tanggal 11 Maret 2020 dilaksanaan workshop dan penyuluhan tentang perilaku hidup bersih dan sehat baik secara individu maupun di lingkungan pondok pesantren, kemudian dilanjutkan dengan bimbingan serta pembentukan kader kesehatan, kemudian dilanjutkan dengan pengadaan UKS (Usaha Kesehatan Sekolah) yang akan dimanfaatkan bagi santri yang sedang sakit untuk istirahat atau sekedar cek kesehatan dan melakukan perawatan sakit ringan. Beberapa santri juga akan kami bentuk untuk menjadi kader kesehatan, agar dapat mengenali tanda gejala awal adanya kesakitan sebelum di periksakan kepada tenaga medis, dan dapat melakukan perawatan mandiri setelah santri berobat hingga santri dapat sembuh dengan baikHasil: kegitan seminar PHBS di lingkungan pondok pesantren berlangsung lancar, dan para santri terlihat antusias mengikuti acara hingga selesai. Pembinaan kader kesehatan telah dilakukan, dan para calon kader kesehatan dapat mengkuti pembinaan dengan baik. Tersedianya UKS baru yang dapat menjadi fasilitas tambahan dan bermanfaat bagi santri.Kesimpulan: Program pengabdian masyarakat ini telah dilaksanakan. Dan menghasilkan sebuah perubahan sesuai dengan target luaran, yakni pengetahuan tentang PHBS individu dan lingkungan pondok pesantren, terbentunya kader kesehatan untuk menjalankan kegiatan UKS, terwujudnya UKS di pondok pesantren Al-Hamdaniyah.
