Abstract

The human population is heterogeneous and consists of populations of immense ethnic diversity. There are considerable allelic differences between human popula-tions as well as individuals within each ethnic group as a result of molecular hetero-geneity of the genome. This, in turn, is responsible for differential allelic expression of genes endowing them with polymorphic characters. The molecular diversity within genes is responsible amongst others, of disease resistance or susceptibility or for that matter drug response. Pharmacogenomics is the key to the understanding of differ-ential drug response in different patients in relation to genetic constitution. The revelation of such information at the molecular level would assist the pharmaceuti-cal industry to address a therapy directed to each individual. The objective of this article is to understand the nuances of the genetic repertoire and correlate it with disease gene identification, genes that have been or can be used as drug targets, identify candidate genes for drug development and recent trends in drug discovery.

Abstract

The pharmaceutical services at a dispensary are prescription dispensing, self-medication and drug information. Patient's satisfaction is an essential factor in improving medicines sale at dispensary. The objective of survey is to know the patient's satisfaction who get the prescription service at Kopkar dispensary. Cross sectional study is conducted by collecting data from 100 adult patients who get the prescription service at Kopkar dispensary on November 2003 by questioner. Respondent are selected by the systematic random sampling. Data were analyzed by comparing between the perception of reality service and the perception of need service using correlation index. The result of this study are: (1) The patients have high satisfaction to the empathy and the cleaning waiting room, (2) The patients have moderate satisfaction to the drug service, the cashier service, the drug information, the drug completed, the drug price, the desk in waiting room, the fan and air conditioning service, and (3) the patients have low satisfaction to the television service.

Abstract

Four flavonoid glycosides have been isolated from the leaves of Crotalaria anagyroides. The glycosides are 1) apigenin 7-O-β-D-apiofuranosyl (1!6)-β-glucopyranosida, 2) apigenin 7-O-β-D-apiofuranosyl (1!2)-β-glucopyranoside, 3) chrysoeriol 7-O-β-D-apiofuranosyl (1!6)-β-D-glucopyranoside, 4) chrysoeriol 7-O-β-D-apiofuranosil (1!2)-β-D-glucopyranoside. Their structures were elucidated by spectroscopic methods of UV, 1H, 13C and 2D-NMR DEPT, COSY and HMBC experiments.

Abstract

The research to accelerate furosemide dissolution rate has been done through physical property modification by solid dispersion forming polyvinylpyrolidone (PVP) carrier with solvent method. Pure furosemide posses property of being practically insoluble in water and has low bioavailability. In current research, six weight ratio of furosemide to PVP being used are 1:1; 1:3; 1:5; 1:9 and 1:15. Physical mixtures are made in equivalent weight ratio. The dissolution rate was examined by paddle method in phosphat buffer pH 5,8. Solid dispersion caracterised with in vitro dissolution study, X-ray diffraction, infra red spectrophotometer and differential scanning calorimetric. The result shows that solid dispersion of furosemide with PVP carrier is higher compare to physical mixture dissolution rate and pure furosemide. The ratio furosemide to PVP who has the highest dissolution rate is 1:15. The analyzing shows the existing of altering crystalline to amorphous state.

Abstract

Almost all district public hospital already has their own formulary. The aims of study are to obtain percentage of noncompliance with the public hospital formulary, to obtain the average additional cost be paid by outpatients as a result of noncompliance with the hospital formulary, and to obtain the average of the outpatient's ability to pay for treatment. A cross sectional study has been carried out to 120 patients in RSU Kabupaten K and 100 patients in RSU Kabupaten B. Subjects of the study were adult outpatients with TB, hypertension and diabetes. Data were collected by well-trained district public hospital staff in interviewing patients. The questioner was first tried out to patients at RSU Kota Jakarta Timur. Data were analyzed by cost analysis. Results of the study are difference in drug item with formulary in RSU Kabupaten K is 66,7% for TB, 96,6% for hypertension; where as in RSU Kabupaten B 44,8% for TB, 82,3% for hypertension and 76,7% for diabetes. Average additional cost that must be paid by outpatients per encounter in RSU Kabupaten K is Rp 10.060 for TB, Rp 26.552 for hypertension; while in RSU Kabupaten B is Rp 5.818 for TB, Rp 8.956 for hypertension and Rp 15.218 for diabetes. The average outpatient's ability to pay for treatment in RSU Kabupaten K is Rp 19.807 and in RSU Kabupaten B is Rp 15.301, which are both less than outpatient treatment cost per encounter.

Abstract

Fruit and leaves of patikala (Etlingera elatior (Jack) R.M.Sm), is a traditional medicinal plants used by people to treat various diseases. The purpose of this study was to determine the total phenolic and flavonoid compounds of E. elatior fruits and leaves extracts. We extracted dried fruits and leaves samples by maceration method using methanol. Qualitative analysis of chemical compounds with certain eluent was done using Thin Layer Chromatography (TLC) to determine the group of active compounds in the extract. The amount of phenolic compounds in the sample was determined by the colorimetric method at a maximum wavelength of 744.8 nm and flavonoid at 431 nm. The results showed that the methanol extract rendemen was 1.93% for fruit and 5.17% for leaf with a total phenolic content of the fruit was 2.29 mgGAE/g b/b extract and leaf was 6.29 mgGAE/g b/b extract. And total flavonoid content of the fruit was 1.7761 % mg Quercetin/g b/b extract and leaf was 5.45 mgQE/g b/b extract.

Abstract

Hylocereus polyrhizus Britton and Rose (Red dragon fruit) has been shown to have antioxidant activity which contains vitamin C, polyphenol, and flavonoid. This research was aimed to investigate antioxidant activity from H. polyrhizus in form of lotion. Lotion were made with 5 concentration from H. polyrhizus methanol extract were 0.04, 0.08, 0.16, 0.32 and 0.64%. Lotion antioxidant activity was measured using DPPH method. The result showed that H. polyrhizus methanol extract had antioxidant activity with inhibition concentration were 19.99±0.33; 25.01±0.08; 39.14±0.04; 66.69±0.12 and 83.37±0.05. The result showed significant differences on antioxidant activity. The physical stability observation of five formula with cycling test method showed unstability because of discolouration (oxidation). As well as the methods of mechanical test showed unstability of lotion because saponification process.

Abstract

Indonesia has many kinds of plants which can be used as natural antioxidant sources. This study was focused on antioxidant activity of Cincau Perdu leaves extract (Premna oblongata Miq.) using 1-Diphenyl-2-picrylhydrazyl (DPPH) methods. P. oblongata leaves was extracted using n-hexane, ethylacetate and methanol. It was found that methanol extract has the highest activity with IC50 value 20.01 μg/mL. The extract with highest antioxidant activity were fractionated with accelerated column chromatography and were earned 6 combination fractions. The antioxidant activity of each combination fractions were tested by DPPH methods. Fraction-5 showed the highest activity as antioxidant with IC50 23.51 μg/mL. The compounds of the active fractions were flavonoid, glikon, saponin and tannin.

Abstract

This study was conducted to observe the profile of DNA Adduct (8-OHdG) formation as DNA damage indicators, by using calf thymus DNA incubated with toxic and carcinogenic compounds. The compounds which could trigger free radicals in this research were PAH(Benzo[a]Pyrene), TiO2, and CuCl2. Calf thymus DNA was incubated with Benzo[a]Pyrene and CuCl2 compounds under pH and temperature variations. The incubation of calf thymus DNA with TiO2-UV radiation (254 nm) was used to induce the formation of reactive oxygen species (ROS) in the process of oxidative DNA damage. From this research, all of compounds have potency to trigger the formation of DNA Adduct (8-OHdG). The ratio of absorbance to assess the purity of DNA at 260 nm and 280 nm (λ260/ λ280 ) was measured at ~1.9. The shifted peaks at λmax were indicating changes on structures of DNA as a result of calf thymus DNA incubation with B[a]P and CuCl2. The highest level of 8-OHdG results in calf thymus DNA incubation with B[a]P and CuCl2 under pH 8.5 and incubation temperature at 60°C, was about 120.856 μg/L. Calf thymus DNA incubation with TiO2-UV radiation (254 nm) under pH 8.5 resulting 8-OHdG level at 57.025 μg/L.

Abstract

Eco-friendly synthesis of imine derivative compound from 2-hydroxy acetophenone and vanillin had been conducted by using a method of stirring (with stirrer) in a water solvent. The whole experiment began with the synthesis of imine compounds, followed by purification and characterization of the synthesis products and finally analysis on their biological and antioxidant activities. The experiments showed that imine compounds could be synthesized by a chemical reaction of 4-amino antipirin aqueous solution with 2-hydroxy acetophenone and vanillin at magnetic stirrer speed of 250 and 450 rpm, respectively, which produced product A, (E)-4-(1-(2-hydroxyphenyl) ethylideneamino)-1,5-dimethyl-2-phenyl-1H-pyrazol-3(2H) -one and product B, (E)-4-(4-hydroxy-3-(vinyloxy) benzylideneamino)-1,5-dimethyl-2-phenyl-1H-pyrazol-3(2H)-one. Yield percentage (%) obtained from the synthesis of product A and B were 40.68% and 19.76% respectively. Toxicity and antioxidant activity tests showed that the product B were toxic and more active as an antioxidant than product A. While the antibacterial activity test showed that both products had a response to Staphyllococcus aureus bacteria and Escherechia coli bacteria.
