Abstract

Purpose: The purpose of this study is to predict the in silico molecular interactions of bitter melon quercetin with GLP-1 receptors using the comparable compounds of myricetin and allosteric modulators. Methods: The in silico test was used to predict the quercetin molecular interactions by docking using the Molegro Virtual Docker computer program. The receptor used was human GLP-1, code PDB: 5VEX with allosteric modulators ligand (97V_1201 [A]). As part from predicting molecular interactions, this study also carried out the prediction of its pharmacokinetic properties (ADME) and the toxicity of quercetin and myricetin using the pkCSM online tool program. The data analysis was performed by comparing the docking bond energies between quercetin, the allosteric modulator ligands and the myricetin comparators at the target receptor. The lower the bond energy of the ligands to the target receptor, the more stable the bonds are. This can be used to predict the biological activity of the compound. Results: The in silico test results showed that the bond energy of quercetin = -70.2678 kcal/mol, myricetin = -105,298 kcal/mol and allosteric modulators = -126,992 kcal/mol. Conclusions: The above test results indicate that quercetin has antidiabetic potential by activating the GLP-1 receptor although it is lower than the myricetin and allosteric modulators. The results of the in silico test using the pkCSM online tool program showed that the quercetin compound had good pharmacokinetic properties and a low toxicity level.

Abstract

The effect of human immunodeficiency virus (HIV) infection on male fertility as assessed by sperm quality is still in controversy in various studies. On the other hand, HIV seropositive men who are stable under the influence of antiretroviral therapy (ARV) experienced an increase in quality and life expectancy that allow them to have offspring. This study aims to investigate the sperm quality of HIV seropositive men in Indonesia. In addition, this study also compared sperm quality in HIV seropositive men who were stable under the influence of ARV (case group) with HIV seronegative men (control group). Semen samples from HIV seropositive male patients at the HIV Integrated Clinic, Ciptomangunkusumo Hospital, Jakarta, Indonesia were collected and analyzed for sperm in the laboratory, including: volume, pH, concentration, motility, morphology and sperm vitality. Abstract

Toxoplasma gondii (T.gondii) is one of the most important protozoa causing abortion in pregnant women and fatal manifestations in immunocompromized individuals. The present study was conducted to assess the effect of Myristica fragrans (M.fragrans) alone and loaded on Chitosan nano particles (CS NPs) on chronic toxoplasmosis as a more effective safe candidate in comparison with spiramycin. One hundred and ten laboratory bred Swiss albino mice were infected by oral inoculation of T.gondii tissue cysts, treated for 14 days starting from the 4th week post infection (p.i) and sacrified at the 10th week p.i. The effect of drugs was assessed by counting the number of tissue cysts in homogenized brain tissues, histopathological examination and SEM of brain tissue. There was a significant decrease in the number of tissue cysts detected in the brain between the control group and all treated groups (P<0.001). Histopathological examination revealed marked improvement of inflammation in all treated groups and the SEM of brain tissues revealed remarkable changes in the shape and structure of the T.gondii tissue cysts.

Abstract

Electrospinning is most widely used technique for fabrication of nanofibres. It is cost effective, simple and versatile process. The resulting nanofibres possess unique properties such as a high surface-area-to-volume and aspect ratio, low density, and high pore volume. These properties make the nanomaterials more advantageous than conventional materials in energy harvesting, conversion, and storage devices. There are different aspects of electrospinning, but the current discussion is based on the use of electrospun fibrous matrices for drug delivery. The article explains about all the available approaches to incorporate drugs onto or within electrospun fibrous matrices, along with their release mechanisms and about the applications of such drug carrying fibrous matrices for regeneration of various tissues, such as neural, vascular, cardiac, skin, and bone.

Abstract

Golden age is a very important period to pay attention to the development of children, especially if there is a suspicion that the growth and development is different from children of his age. Objective: To develop early detection instruments for growth and development in children with special needs. Method: The research design was Research and Development carried out in 2 stages. The first stage was identifying the DDTK instrument from the SDIDTK and Denver II manuals and then conducting FGDs with 70 participants. The second stage is test results development of 30 parents and their children with special needs to get recommendations on the results of instrument development. Methods to identify children with special needs by means of observation, interviews, documentation, orders and a combination of 3 methods. Data analysis using descriptive analysis, validity test and using product moment and reliability using Alfa Chronicle. There are eight questionnaires to detect blind, deaf, mentally retarded, disabled, disabled, autistic, ADHD and special intelligent. The development of this instrument has the addition of aspects of behavior and physical appearance that exist in children. The development of instruments for early detection of development of children with special needs consists of 8 questionnaires. The results of the instrument trial showed good results and the opinion of special school teachers and ABK therapists was very positive with the development of this instrument so it is recommended to be disseminated to posyandu cadres, the community and health workers.
