Biocaster : Jurnal Kajian Biologi (B : JKB), focus to bridge the gap between research and practice, providing information, ideas and opinion, in addition to critical examinations of advances in biology research, teaching, and learning. Through the coverage of policy and curriculum developments, the latest results of research into the teaching, learning and assessment of biology are brought to the fore. Biocaster : Jurnal Kajian Biologi (B : JKB), receiving and publishing article in the form of research and review (scientific article) in the field of biology education and utilization of biological research in teaching and learning. Each article must be linked to the theme "21st century skills in biology education" and / or "Efforts to support the achievements of the goals set forth in the Sustainable Development Goals (SDGs) through biology education". All articles are published in English and under go a peer-review process. Abstract

The aim of this study is to synthesise the findings of various studies on the effectiveness of aquatic plants in reducing ammonia concentrations in aquaculture systems, to identify the mechanisms of phytoremediation involved, and to compare the effectiveness of different plant species based on previous studies. The method used was a systematic literature review with a descriptive-qualitative approach. This study did not conduct experiments or collect primary data, but rather used secondary data from relevant scientific articles. Literature was obtained from Google Scholar, ScienceDirect, and other indexed journals using keywords related to ammonia, aquaculture, phytoremediation, and water plants. The inclusion criteria included original research articles from 2015–2026 that were available in full text and presented quantitative data on ammonia reduction. Fifteen articles that met the criteria were analysed using thematic synthesis through grouping, comparison, and interpretation of data. The results of the synthesis showed that floating water plants such as Eichhornia crassipes, Pistia stratiotes and Lemna minor were the most effective, with ammonia reduction of over 90% in several studies. The main mechanisms include nitrogen absorption by the plants, rhizofiltration, and microbial activity through nitrification in the root zone. The effectiveness is influenced by biomass, root systems, and environmental conditions such as pH, temperature, and nutrient concentration. In conclusion, the synthesis of various studies shows that phytoremediation based on floating aquatic plants is effective and has the potential to be a sustainable and environmentally friendly technology for the treatment of aquaculture waste.

Abstract

This study analyzes the habits of mind of biology education students based on 16 indicators from Costa and Kallick. The study used a descriptive quantitative approach with 22 purposively selected biology education students as subjects. Data were collected using a validated 4-point Likert scale questionnaire with high reliability (Cronbach's Alpha = 0.88). Data were then analyzed using descriptive statistics, the Friedman test, and Kendall's W test. The results showed that students' habits of mind were in the moderate category and uneven across all indicators. Persistence, empathy, thoroughness, and curiosity were in the high category, while flexible thinking was in the low category. The Friedman test showed significant differences between indicators with a moderate effect size (Kendall's W = 0.32). These findings demonstrate the need for biology learning designs that emphasize the development of flexible thinking through open inquiry, problem-based learning, and metacognitive reflection.

Abstract

Mangrove ecosystems in the Air Telang Protected Forest are experiencing ecological pressure due to the dominance of the liana Derris trifoliata, which has the potential to alter the structure of vegetation communities. This study aimed to analyze the patterns of floral associations in mangrove areas affected by the dominance of Derris trifoliata. The study employed a descriptive survey method with a purposive sampling approach across four observation stations. Species association analysis was conducted using the Jaccard Index based on species presence data. The results identified 34 plant species associated with Derris trifoliata, consisting of ferns, monocots, and dicots, indicating a heterogeneous vegetation community. Similarity values among stations ranged from 0.154 to 0.594, with the highest similarity observed between Stations 2 and 3, and the lowest between Stations 2 and 4. The association values between Derris trifoliata and other species ranged from 0.25 to 1.00, with a very high association found in Nypa fruticans and a high association in Acrostichum aureum. The dominance of neutral associations indicates ecological pressure that tends to simplify the structure of mangrove vegetation communities. Ecologically, the presence of Derris trifoliata plays a role in shaping species distribution patterns through interactions influenced by environmental heterogeneity and mangrove habitat conditions.

Abstract

Traditional knowledge regarding the utilization of medicinal plants as ingredients for traditional herbal medicine (jamu) in Karangrejo Village, Kampung Jamu Kromengan, represents a form of local wisdom that is still maintained by the community. This study aimed to identify the diversity of medicinal plants and analyze their utilization levels based on quantitative ethnobotanical indices. The research employed a mixed-method approach with an exploratory sequential design through interviews, observations, documentation, and questionnaire surveys. The results revealed 12 species of medicinal plants belonging to 6 families, with the Zingiberaceae family dominating (7 species). Rhizomes were the most frequently utilized plant part, with a PPUV value of 48.8%, indicating the community’s high reliance on rhizomatous plants in jamu preparation. The highest ICF value (0.97) in the categories of relieving muscle aches and increasing appetite indicated a strong level of community agreement regarding the use of certain plants for these health conditions. The highest RFC values were found in kencur and rice, the highest SUV in turmeric, the highest FUV in the Zingiberaceae family, and the highest FL in temulawak for increasing appetite, indicating that plants from the Zingiberaceae family have high utilization, trust, and ethnobotanical importance in traditional jamu practices. In conclusion, jamu practices in Karangrejo Village are dominated by the use of rhizomatous plants, which play an important role in traditional medicine and reflect the strong local knowledge of the community regarding the use of medicinal plants.

Abstract

This study aims to identify, analyze, and document the use of plants in the Wiwit Kopi ritual in Segunung Traditional Village, Wonosalam District, including plant species, habitus, organs used, methods and stages of use, philosophical meaning, and conservation efforts. This study uses a qualitative descriptive approach with data collection techniques in the form of non-participatory observation, documentation, and semi-structured interviews. The determination of informants was carried out using a purposive sampling technique for one key informant and a snowball sampling technique for three additional informants. Data analysis consists of data condensation, data presentation, and drawing conclusions using triangulation techniques to ensure data validity. The results showed that there were 21 plant species from 14 families used in the Wiwit Kopi ritual with a dominant herbaceous habitus of 57.1%. The most commonly used plant organ was the leaf at 19.0%, while the conservation status category was dominated by Not Evaluated (NE) at 66.7%. The most prominent findings indicate that the use of plants in the Wiwit Kopi ritual not only serves as a complement to the ritual but also represents philosophical values ​​and supports culturally based conservation through the use of local plants passed down through generations.

Abstract

Prolonged use of disposable masks can increase bacterial accumulation due to exposure to droplets, aerosols, and respiratory humidity. This study aims to identify the types of bacteria and determine the number of bacterial colonies on disposable masks based on the duration of use by educational laboratory staff at Bali International University. This study used an analytical observational design with a cross-sectional approach. Samples consisted of 15 disposable masks grouped by duration of use: 3 hours, 6 hours, and 9 hours, with 5 samples each. Examination was carried out through bacterial culture, microbiological identification, and total plate count calculation. The results showed that the number of bacterial colonies increased with increasing duration of mask use. The average total plate count at 3 hours of use was 31.00 ± 9.62 CFU/g, increased to 138.00 ± 33.47 CFU/g at 6 hours, and reached 646.00 ± 122.39 CFU/g at 9 hours. The dominant bacteria found were Bacillus sp. and Staphylococcus sp., with Bacillus sp. dominating across all sample groups. This study concluded that the longer disposable masks were used, the higher the number of bacterial colonies accumulated. Regular mask replacement every 4–6 hours is recommended to maintain hygiene and safety in the laboratory.

Abstract

The lack of knowledge and information among the public about the characteristics and behavior of hammerhead worms has raised concerns about the dangers to human health when coming into contact with these worms. The purpose of this study is to analyze the formation of negative public perceptions regarding the hammerhead worm Bipalium kewense based on information circulating in online media, as well as to examine its consistency with scientific facts available in the literature. This study was conducted using a narrative literature review approach focused on analysis. The results indicate that the characteristics of the Bipalium kewense worm, such as its morphology, reproductive biology, predation strategies, locomotive organs, toxin content, behavior, and conservation status, indirectly contribute to the formation of negative public perceptions, particularly in residential areas, regarding the presence of this worm. In fact, all these traits, which reflect the worm’s life mechanisms, are forms of biological adaptation commonly found in various groups of invertebrates to ensure their survival. The public’s lack of understanding regarding the life processes of the hammerhead worm leads to these scientific facts being more readily perceived as something terrifying rather than as a natural biological phenomenon. This situation is further fueled by the dissemination of digital information that tends to emphasize the threat aspect rather than providing a balanced scientific explanation. It is concluded that the low level of biological and ecological literacy among residents in residential areas plays a significant role in the formation of misconceptions regarding the presence of B. kewense, as to date, no scientific evidence has been found to indicate that this worm causes.

Abstract

The low amount of IAA production by plants causes plants to often obtain IAA from bacteria that live around the roots or rhizosphere. Exploration of IAA bacteria from Buttu Puang, a protected forest area in West Sulawesi, has never been done. One of the typical plants that inhabit Buttu Puang is the dengen plant (Dillenia serrata). The purpose of this study was to screen and identify the ability of rhizosphere fungi from dengen plants in the Buttu Puang protected forest area to produce IAA. The implication of the study is that isolates with the highest activity can be developed as PGRs producing the phytohormone IAA in the future. The research method began by isolating fungi from the rhizosphere soil. Next, the ability to produce IAA was tested by adding the IAA precursor (tryptophan) to the growth medium. After that, the supernatant was then added with Salkowski. A positive test was indicated by the formation of a pink color at the end of incubation. The results of the study showed that 13 isolates of rhizosphere fungi were found from dengen plants in Buttu Puang Forest, namely isolates DG 1.1, DG 1.2, DG 1.3, DG 1.4, DG 2.1, DG 2.2, DG 2.3, DG 2.4, DG 2.5, DG 3.1, DG 3.2, DG 3.3 and DG 3.4. Qualitative and quantitative tests using Salkowski reagent and spectrophotometer measurements at a wavelength of 530 nm showed that only two isolates were able to produce IAA, namely isolate DG 2.1 (104.09 ppm), and isolate DG 2.5 (39.26 ppm). These results indicate significant differences in physiological potential among isolates, which are thought to be related to the physiological characteristics and IAA biosynthesis pathways of each genus. Macroscopic and microscopic identification showed that isolate DG 2.1 was suspected to be a member of the genus Penicillum, while isolate DG 2.5 was suspected to be an Aspergillus. These findings confirm that the phytohormone-producing rhizosphere microorganisms from dengen plants in Buttu Puang Forest have the potential to be developed as plant growth regulators (PGRs) in the future.

Abstract

The purpose of this study was to identify and document the diversity of medicinal plants used by the people of Srimulyo Village, Dampit District, Malang Regency, which is still on the slopes of Mount Semeru. This study used a mixed method with a sequential exploratory design. Qualitative data were obtained through interviews with key informants and additional informants, while quantitative data were obtained through questionnaires to 98 respondents. Informants were selected using purposive sampling and snowball sampling which were analyzed qualitatively and quantitatively using the Informant Consensus Factor (ICF) index, Relative Frequency of Citation (RFC), Use Value (UV) analyzed from Species Use Value (SUV), Plant Part Use Value (PPUV) and Family Use Value (FUV), as well as Fidelity Level (FL). The results of the study identified 15 plant species from 10 families used to treat digestive disorders, with the dominant family Zingiberaceae (FUV=2.27) and the highest use value in Zingiber officinale (SUV=1.23). Leaves are the most widely used part of the plant (35.14%), generally through boiling. The ethnobotanical index shows a high consensus of use in typhoid fever (ICF=1.00) and constipation (ICF=0.98), with Psidium guajava as the most frequently mentioned species (RFC=0.78). Several species such as Musa sp., Cymbopogon citratus, Colocasia esculenta, Carica papaya, Piper retrofractum, and Strobilanthes crispus show absolute specificity of use (FL=100%). This ethnobotanical data directly contributes to local knowledge conservation efforts while strengthening the biocultural resilience of communities in adapting to the environment on the slopes of Mount Semeru from generation to generation.

Abstract

Gram-negative bacteria in layer chickens have the potential to be a source of infection and a reservoir of pathogens that impact animal and human health, as well as the safety of poultry-derived food. The presence of these bacteria in the cloaca reflects the level of microbial contamination in the farm environment and can be an indicator of low barn sanitation and biosecurity. This study aimed to isolate and identify gram-negative bacteria in the cloaca of layer chickens in Mamben Daya Village, Wanasaba District, East Lombok Regency. The study used a descriptive laboratory design with 35 cloacal swab samples taken randomly from smallholder farms. Bacterial isolation was carried out through a pre-enrichment stage using Buffered Peptone Water (BPW), followed by enrichment with Selenite F Broth, and cultivation on selective Xylose Lysine Deoxycholate (XLD) media. Identification was carried out through gram staining and biochemical tests including sugar tests, Sulfide Indole Motility (SIM), Triple Sugar Iron Agar (TSIA), and Simmons Citrate Agar (SCA). The results showed that 34 of 35 samples (97.14%) were positive for gram-negative bacilli. Biochemical tests identified bacteria from the Enterobacteriaceae family, namely Escherichia coli, Escherichia spp., Citrobacter spp. (Citrobacter freundii and Citrobacter diversus), and Klebsiella oxytoca, with Escherichia coli as the dominant isolate. The dominance of Escherichia coli indicates a high risk of contamination of the farm environment and poultry-derived food products, which could impact public health. This finding underscores the importance of implementing biosecurity, barn sanitation, and regular microbiological monitoring to support livestock health and food safety.
