Indonesian Journal of Foof Technology is the official publication of the Departement of Food Technology Jenderal Soedirman University. The Journal is envisaged as the major platform for communicating advances/developments in the post-harvest science, engineering, and technology aspects of food/food products to researchers, academicians, the industry as well as students of food science and technology. This peer-reviewed journal publishes novel and high-quality reviews, original research, and short communications covering the wide range of post-harvest related science, engineering, and technological aspects of foods, food products, and related byproducts with special emphasis on the basic and applied research findings that will impact the quality, safety and/or shelf life of fresh or processed food products including findings that will help improvise the efficiency of a process/technology, effective utilization of by-products or wastes resulting from food processing operations. Manuscripts dealing with pre-harvest aspects will only be considered if the work clearly indicates the relevance to post-harvest practices/processing that will directly impact the safety, quality, or shelf life of the food or food products. Critical reviews on new perspectives in food handling and processing, innovative and emerging technologies and trends, and future research in food products and food industry byproducts are also welcome. Abstract

Background: Contamination of food by pathogenic bacteria such as Escherichia coli and Pseudomonas aeruginosa causes significant food poisoning globally. The use of natural preservatives serves as an alternative to synthetic preservatives. Phytochemical compounds with antibacterial activity hold great potential for the development of natural food preservatives. Purpose: This study aimed to identify the phytochemical constituents and evaluate the antibacterial potential of fresh Katuk (Sauropus androgynus (L.) Merr) leaf methanolic extract against both bacteria. Method: The extract was obtained via maceration, followed by phytochemical screening and antibacterial activity assays using the agar well diffusion method at extract concentrations of 20%, 40%, 60%, and 80% (w/w), with chloramphenicol 0.1% as the positive control and distilled water as the negative control. Data were statistically analyzed using One-Way ANOVA followed by Tukey's Post-hoc HSD test (p = 0.05). Result: The phytochemical screening results showed that the extract contained all tested compound groups. The antibacterial test against E. coli demonstrated significant activity starting from 60% (inhibition zone diameter of 10,70 ± 0,31 mm) to 80% (10,93 ± 0,35 mm) with no statistically significant difference (p > 0.05). Conversely, inhibition against P. aeruginosa was only observed at the highest concentration of 80% with an average diameter of 10,28 ± 0,23 mm. The extract concentration had a significant effect on the inhibition zone diameter (p < 0.0001). It can be concluded that the presence of flavonoids, tannins, and phenolics contributed to the antibacterial activity of the fresh Katuk leaf extract, highlighting its potential to be developed as a natural antibiotic and food preservative.

Abstract

Background: Functional beverages has a widely potential benefit for health due to bioactive compounds, including phenolic, serving as a natural antioxidants. Soursop leaves (Annona muricata L.) and lemongrass (Cymbopon citratus) combined with powdered honey may contain a phenolic compound potential for herbal beverages, especially the herbal brewed tea, but yet hasn’t been widely developed. Purpose: This research focused on identifying the effect of different formulations of soursop leaves and lemongrass on the total phenolic content of a herbal brewed tea with powdered honey as a natural sweetener. Method: A completely randomized design with a single factor was applied for the five different formulations, namely F1 (90:0:10), F2 (0:90:10), F3 (45:45:10), F4 (25:65:10), and F5 (65:25:10), replicated four times each. The observed variable was the total phenolic content of each herbal brewed tea formulation produced. Result: The ANOVA results showed that different formulations significantly affected the total phenolic content of herbal brewed tea (p<0.05), ranged from 2.82 to 8.06 mg GAE/g. The highest phenolic content was obtained in F5 (8.06 ± 0.34 mg GAE/g), followed by F1 (7.71 ± 0.11 mg GAE/g), F3 (5.44 ± 0.28 mg GAE/g), F4 (4.15 ± 0.36 mg GAE/g), and F2 (2.82 ± 0.19 mg GAE/g). Thus, F5 (65:25:10) was considered the most optimal formulation and had the potential to produce a suitable herbal brewed tea made from soursop leaves, lemongrass, and powdered honey.
